Goldenseal (Hydrastis canadensis L.) is an important medicinal plant, native to Canada and Eastern United States, used as an antibiotic, anticonvulsant and in treatments for inflammations and dyspepsia. Over collection of wild population and native habitat destruction has lead the species to be listed as an endangered species in the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES), Appendix II. This brought a revived interest in conservation of germplasm.;Calcium alginate solid and hollow bead encapsulation protocols were evaluated. The hypothesis of this project is that a functionally relevant association exists between CFTR and Hsp90. In testing this hypothesis, (a) the association of CFTR and Hsp90 is confirmed via crosslinking and immunoprecipitation of CFTR from stably transfected HEK-293 cells, electrophoretic separation of cross-linked proteins, and protein identification via SDS-PAGE, western blotting, and mass spectrometry. Also, this hypothesis is tested by (b) functionally assessing the effect of Hsp90 on CFTR function via Ussing chamber analysis, two-electrode voltage clamp analysis, preliminary in vivoanimal studies, and protein phosphorylation. A variable resolution x-ray (VRX) computed tomography (CT) scanner can image objects of various sizes with greatly improved spatial resolution. The scanner employs an angulated discrete detector and achieves the resolution boost by matching the detector angulation to the scanner field of view (FOV) determined by the size of an object being imaged. A comprehensive evaluation of spatial resolution in an experimental version of the VRX CT scanner is presented in this dissertation. Calpain is a class of intracellular cytoplasmic cysteine proteases.1 The enzyme participates in different intracellular signaling pathways that are mediated by Ca2+.2 The two major isoforms of calpain universally distributed in most mammalian tissues are calpain 1 (µ-calpain) and calpain 2 (m-calpain). The exact in vivo function of the enzyme is not clear, but calpain has been implicated in a variety of physiological and pathological conditions,3 such as cancer, stroke, cardiac ischaemia, muscular dystrophy, cataract and Alzheimer’s disease. Calpain inhibitors are therefore of interest as therapeutic agents and as biomedical tools.

Although the majority of coronary artery bypass graft (CABG) surgery patients are extubated within 6 to 8 hours following surgery, 20% to 40% of patients remain intubated 12 hours after surgery due to dysfunctional ventilator weaning response (DVWR). DVWR associated with increased morbidity and mortality (30% to 43%) following CABG surgery. Finding significant antecedence to predict DVWR could help to identify and prevent the complications from DVWR after CABG surgery. Literature review revealed that there is an association between cardiopulmonary indicators (CPI) and DVWR after CABG surgery. Cardiopulmonary indicators are the selected hemodynamic parameters that have an association with DVWR. …

Since the mid- to late 1990s, the scientific and medical research community has sought to increase its access to healthy children for research protocols that involve harm or a risk of harm. This move reverses longstanding policy within that community generally to exclude healthy children from such protocols on the grounds that the research as to them is non-therapeutic, that they are particularly vulnerable to research-related abuses, and that they are unable themselves to give informed consent to their participation. The primary goal of this dissertation was to examine the long-term effect of prenatal alcohol exposure (PAE) on the primary somatosensory (SI) and primary motor cortex (MI). For over 30 years, PAE has been shown to produce a triad of symptoms (growth deficits, craniofacial dysmorphologies and central nervous system disruptions) diagnosed as fetal alcohol syndrome (FAS) in children and adults. Since low to moderate levels of prenatal alcohol exposure are less likely to produce FAS symptoms in children, the term “PAE” in this dissertation typically refers to heavy/abusive/chronic levels of prenatal alcohol exposure (in humans this equates to 5+ drinks/per …

Delta-Like 4 Notch Ligand Regulates Tumor Angiogenesis, Improves Tumor Vascular Function, And Promotes Tumor Growth In Vivo, J-L Li, Rca Sainson, W Shi, R Leek, Et Al

The vascular endothelial growth factor (VEGF) plays a key role in tumor angiogenesis. However, clinical trials targeting the VEGF pathway are often ineffective, suggesting that other factors/pathways are also important in tumor angiogenesis. We have previously shown that the Notch ligand Delta-like 4 (DLL4) is up-regulated in tumor vasculature. Here, we show that DLL4, when expressed in tumor cells, functions as a negative regulator of tumor angiogenesis by reducing the number of blood vessels in all five types of xenografts, but acts as a positive driver for tumor growth in two of them (human glioblastoma and prostate cancer). Global Analysis Of An Expectations Augmented Evolutionary Dynamics, Angelo Antoci, Antonio Gay, Massimiliano Landi, Pier Luigi Sacco

We consider a deterministic evolutionary model where players form expectations about future play. Players are not fully rational and have expectations that change over time in response to current payoffs and feedback from the past. We provide a complete characterization of the qualitative dynamics so induced for a two strategies population game, and relate our findings to standard evolutionary dynamics and equilibrium selection when agents have rational forward looking expectations.

Two species with different ecologies and natural histories use different morphological characteristics and thermoregulatory strategies to survive seasonal environmental changes. The purpose of this project was to investigate two such species, the eastern red bat (Lasiurus borealis) and big brown bat (Eptesicus fuscus). Red bats roost in the foliage of trees and occasionally leaf litter and have dense fur, as well as a furred interfemoral membrane. These two species differ in their morphology and roosting ecology in that big brown bats are generally larger with less dense fur, no fur on the interfemoral membrane, and are less exposed to environmental …

I studied the effect of environmental conditions on the foraging behavior of Carolina chickadees (Poecile carolinensis) in Kiewig woods, property of Indiana State University, in West Terre Haute in west-central Indiana. My study method uses Optimal Foraging Theory (Brown 1988) to measure the giving up density (GUD) of Chickadees under different environmental conditions ( different resource abundances, different habitat types, different resource types and different seasons). For my experiment, I set up 24 feeding stations in three kinds of habitats ( open area, secondary growth woods and mature woods) within 4 sites and paired trays in each habitat X site. …

MicroRNAs (miRNAs), a large class of short noncoding RNAs found in many plants and animals, often act to post-transcriptionally inhibit gene expression. We report the generation of deletion mutations in 87 miRNA genes in Caenorhabditis elegans, expanding the number of mutated miRNA genes to 95, or 83% of known C. elegans miRNAs. We find that the majority of miRNAs are not essential for the viability or development of C. elegans, and mutations in most miRNA genes do not result in grossly abnormal phenotypes. These observations are consistent with the hypothesis that there is significant functional redundancy among miRNAs …
