Candida albicans is an opportunistic fungal pathogen in humans. It typically lives as a harmless commensal as part of the human gut and skin flora, but under some conditions it causes mucosal infections. In immunocompromised patients it can also cause lifethreatening systemic infections. C. albicans is a multimorphic organism, and the ability to undergo morphological transition from budding to hyphal growth is associated with its virulence. This morphological switch was reported to be regulated by different signaling pathways. One key pathway is cAMP-PKA signaling pathway. The single adenylyl cyclase in C. albicans is CaCdc35, which was reported to interact with …

The devastating effects of Alzheimer’s and related amyloidogenic diseases have inspired the synthesis and evaluation of numerous ligands to understand the molecular mechanism of the aggregation of the beta-amyloid peptide. Our review focuses on the current knowledge in this field with respect to molecules that have been demonstrated to interact with either oligomeric or fibrillar forms of the beta-amyloid peptide. We describe natural proteins, peptides, peptidomimetics, and small molecules that have been found to interfere with beta-amyloid aggregation. We also detail recent efforts in selecting molecules that target beta-amyloid isolated from antibody, protein, and peptide libraries. These new molecules will …

On April 23, 2007, The William Monroe Trotter Institute held a Roundtable convening more than fifty individuals at UMass Boston for a two-hour conversation on the education of black children in Boston. Roundtable attendees represented a balanced mix of professional and lay persons, including educators, advocates, academics, elected officials and policymakers, and donors. Attendees included the Suffolk County Sheriff, the Acting Superintendent of Boston Public Schools (BPS), and the president of the Boston Teacher’s Union.

To limit the damage to nearby healthy tissue, accurate modeling is required of the mechanical behavior of a target soft tissue subject to surgical manipulations. Heart failure is recognized as a major cause of death among diabetics. In type I and II diabetes, glucose uptake, glycolysis and pyruvate oxidation are impaired, and fatty acid utilization increases. These alterations in metabolism contribute toward cardiac contractile function. There is a gap in our understanding on how alterations in glycolytic metabolite fructose-2,6-bisphosphate (F-2,6-P 2 ) affects cardiac dysfunction. Two cardiac-specific overexpression of PFK-2 (phosphofructose kinase-2) transgenic models were used to investigate the role of altered metabolism by F-2,6-P 2 in provoking cardiomyopathy. Previous studies on the mechanistic induction of anti-tumor responses by IL-12 cytokine therapy have focused on the adaptive immune response, specifically the activation NK cells and T cells as the primary targets of IL-12 treatment. In contrast, little attention has been given to the potential role of macrophages in the initiation of anti-tumor responses by IL-12 therapy despite reports that macrophages play a major role in promoting tumor growth and metastasis and in suppressing anti-tumor immune responses. Embryonic development requires the orchestration of temporally precise genetic events that culminate in the formation of a complete organism. The molecular mechanisms responsible for ontogenesis are regulated by environmental and somatic factors in utero that activate or repress the expression of numerous genetic elements resulting in fetal programming of adult diseases. The aryl hydrocarbon receptor (AHR) is an important nuclear transcription factor both during embryogenesis and throughout maturity in multiple organisms. The combination of viral mediated gene delivery, tissue engineering, and Schwann cell (SC) transplantation offer a promising strategy to enhance axonal regeneration and functional recovery following spinal cord injury (SCI). The rationale and feasibility of using such combination has been extensively reviewed in Chapter 1. The study presented in Chapter 2 showed that lentiviral vectors are most suitable in situations where stable long term transgene expression is needed, while adenovirus and retrovirus are more suited for transient gene delivery. The purpose of this dissertation was to demonstrate that a new method of direct intracellular energy delivery was effective in maintaining viability of in vitro Human Umbilical Vein Endothelial Cells (HUVECs) when exposed to chemical ischemia for prolonged periods of time and in preventing in vivo ischemia reperfusion injury in a composite tissue transplantation model. Ischemia and reperfusion injury is a well recognized phenomenon that occurs in situations like organ transplantation, shock, cardiac surgery, etc, where tissues are temporarily deprived of nutrients and oxygen for energy production. In spring 2006, the Frazier International History Museum, in conjunction with the Ekstrom Library Photographic Archives at the University of Louisville, mounted an exhibit of 47 photographs taken by a young French foot soldier during World War I. The exhibit, titled "André Jeunet: Images of WWI by a Pacifist Soldier," ran from May 19 through September 4, 2006. Archaeologists use soil analysis to detect chemicals, like phosphate, to indicate areas of anthropogenic activity. Phosphate detection is a multi-step process, which makes standard techniques time consuming. Kinetic studies decreased the analysis time for the malachite green (MG) method of phosphate detection. The 3-minute method allows extraction and analysis to be complete in 15 minutes. Continued studies resulted in two-color spectral monitoring, which provided values instantaneously. Arsenate (As(V)) interfere with the MG method and results in overestimation of phosphate. As(V) must be reduced to non-interfering arsenite. Two As(V) reducing agents--L-Cysteine and thiosulfate--were investigated. The thiosulfate method was suitable for field …

Recent attention has focused on the possibility that even low-level Pb exposure can produce subtle neurological impairments in the absence of overt signs of toxicity. Previous behavioral assessments of learning and memory using DMTS procedures have documented learning impairments in monkeys exposed to Pb during early postnatal development, and although a variety of operant behavioral assays have documented detrimental effects of dietary Pb treatment in rats, the DMTS procedure has not been examined in rats following short-term low levels of Pb through dietary exposure. Traditional coral reef monitoring efforts lack assessment of coral health at tissue and cellular levels. This thesis investigated tissue, cellular, and gross morphological responses of the Caribbean staghorn coral, Acropora cervicornis, to elevated sedimentation and phosphate using state-of-the-art histological techniques. Branch fragments of A. cervicornis were collected offshore Broward County, FL, acclimated for 12 weeks, subjected to a 4-week experimental period, and given a 1-week recovery period in laboratory aquaria. In an extension and partial reformulation of Conceptual Metaphor Theory (Lakoff & Johnson, 1980), Ritchie (2003, 2004, 2006) proposed that the linguistic expressions cited as evidence of complex conceptual metaphors can be parsimoniously interpreted in terms of perceptual simulators (Barsalou, 1999), often within extended fields of meaning, which may be but are not necessarily anchored in underlying conceptual metaphors. Cameron (2003, 2007) added substance and precision to the focal concept of communicative context, and showed how metaphors can be analyzed both as part of an overall pattern of figurative language in a communicative event. Interpreting The Fourteenth Amendment: Two Don'ts And Three Dos, Garrett Epps

A sophisticated reading of the legislative record of the framing of the Fourteenth Amendment can provide courts and scholars with some general interpretive principles to guide their application of the Amendment to current legal problems. The author argues that two common legal conceptions about the Amendment are, in fact, misconceptions. The first is that the Amendment was chiefly concerned with the immediate situation of freed slaves in the former slave states. Instead, he argues, the legislative record suggests that the framers were broadly concerned with the rights not only of freed slaves but also of foreign-born immigrants in the North …

On Thursday, 01 February 2007, Scott Raasch of rural Madison County called to say he had what he believed to be a Eurasian Tree Sparrow (Passer montanus) coming to a feeder at his house just east of Enola. The bird had been present for about a week, although he had just identified the species upon purchasing a new field guide. He reported that the bird was very wary and only stayed for a short time at the feeder. The bird was most often observed in the early morning or just before dusk.

This was essentially a normal fall season in Nebraska, with no major weather events or significant shifts in breeding or migration ranges, although a possible exception is a first record of Lesser Goldfinch breeding in the state. One observer made the point, though, that the hard freeze in the spring affected seed and fruit crops for fall, most conspicuously resulting in fewer large American Robin flocks. Excellent water conditions in the Rainwater Basin were welcome, but few herons and egrets were present, and no unusual breeding events were reported there.

The 2007 NOU Fall Field Days were held at the 4-H Camp in the Nebraska National Forest near Halsey on September 28-30, 2007. Among the approximately 45 in attendance were two first time attendees: Juanita Rice of Fairmont and Tim Hajda of Broken Bow.

Wayne Mollhoff spoke on Friday night about the progress on the new breeding bird atlas: "A Funny Thing Happened on the Way to the Atlas," and on Saturday night, Bob Rooney did a multimedia presentation entitled "A Sandhills Odyssey."

Detection Of Polychlorinated Biphenyls In The Rio Grande Basin Above Cochiti Dam: Sources & Significance, Anthony D. Edwards

Polychlorinated biphenyls (PCBs) have been detected with increasing frequency in water and fish tissue samples in the Rio Grande Basin above Cochiti Dam (RGBACD), which correlates with the use of improved analytical methods for quantifying PCBs. Levels of PCBs identified in some fish have exceeded levels considered safe for human consumption, resulting in a Fish Consumption Advisory' for Abiquiu and Cochiti Reservoirs and the Rio Grande from Pojoaque Creek to Frijoles Canyon (Bandelier National Monument). In June of 2007, the New Mexico Environment Department (NMED) listed seven water bodies in the RGBACD as impaired due to elevated concentrations of PCBs …
