Background: 2-Chloroethyl ethyl sulphide (CEES) is a sulphur vesicating agent and an analogue of the chemical warfare agent 2,2′-dichlorodiethyl sulphide, or sulphur mustard gas (HD). Both CEES and HD are alkylating agents that influence cellular thiols and are highly toxic. In a previous publication, we reported that lipopolysaccharide (LPS) enhances the cytotoxicity of CEES in murine RAW264.7 macrophages. In the present investigation, we studied the influence of CEES on nitric oxide (NO) production in LPS stimulated RAW264.7 cells since NO signalling affects inflammation, cell death, and wound healing. The Influence Of Membrane Lipid Order On Cell Shape And Microvesiculation In Human Erythrocytes, Laurie Jackson Gonzalez
Theses and Dissertations
Exposure of human erythrocytes to elevated intracellular calcium causes alterations in cell shape and stimulates shedding of the cell membrane in the form of microvesicles. We hypothesized that both the shape transition and microvesiculation are influenced by microscopic membrane physical properties such as lipid order. To test this hypothesis, membrane properties were manipulated by varying the experimental temperature, membrane cholesterol content, and the internal ionic environment. Changes in membrane order were assessed using steady-state fluorescence spectroscopy with an environment-sensitive probe, laurdan. Our observations led us to the following conclusions: 1) the modest temperature dependence of membrane structure observed with laurdan …

This background paper traces the development within American health care of two interrelated trends and activities: an evidence-based approach to medical practice and the critical evaluation of new technologies with respect to their costs and effectiveness. Over the past 35 years each of these developments has increasingly shaped the coverage decisions of public and private health insurers, and their importance for coverage policy is certain to grow. The paper also contrasts the different approaches to such “evidence-” or “value-based” coverage policy in the mixed public and private U.S. health care enterprise with the approach taken in Great Britain’s single-payer National …

Management of water resources requires an understanding of the full range of natural variability and its consequences. The weather record, which spans the last 100 years or so, provides a snapshot of the contemporary history of drought and its impacts, but this record is relatively short and is inadequate for understanding long-term trends or for evaluating the magnitude of human impacts. A variety of so-called paleoclimatic records - such as tree rings, lake sediments, and sand dunes - record the history of the environment and can be used to extend the record of climatic variation to older intervals of time. …

SUMMARY. We consider a semiparametric regression model that relates a normal outcome to covariates and a genetic pathway, where the covariate effects are modeled parametrically and the pathway effect of multiple gene expressions is modeled parametrically or nonparametrically using least squares kernel machines (LSKMs). This unified framework allows a flexible function for the joint effect of multiple genes within a pathway by specifying a kernel function and allows for the possibility that each gene expression effect might be nonlinear and the genes within the same pathway are likely to interact with each other in a complicated way. Water Optimizer is a tool for analyzing alternative water management strategies when the available water supply is limited. It can be used by producers to determine the profit maximizing crops to produce and the optimum amount of water to apply to each crop, given a particular water supply. It can also be used to evaluate the economic consequences of public policies which expand or reduce the amount of irrigation.
The Water Optimizer model can compute the profit maximizing strategy for producers located in any county in Nebraska where there is significant irrigation. This can be done for three different soil …

An Intellectual Property Rights (IPRs) system is effective when infringers can be identified, successfully sued for damages and deterred from further infringement. The effectiveness of IPRs in plant varieties is limited due to high detection costs of unauthorized use of seed that embodies intellectual property (e.g., genetically modified (GM) seed) and high enforcement costs. Seed companies have traditionally performed limited research and development (R&D) in self-pollinating plants mainly because seed saving limits their ability to recoup their investment.1

Factors Influencing Off-Flavor In Beef, Jennie Marie James Hodgen
Department of Animal Science: Dissertations, Theses, and Student Research
Projects were conducted to increase knowledge of liver-like off-flavor origins in muscles from the beef chuck and round. Effects of cooking rate and holding time on off-flavor of various steaks from ten carcasses were determined. Off-flavor from these muscles was lowest when the steaks were cooked slowly (on a 149°C grill versus a 249°C grill) and when held for one hour prior to evaluation. The M. infraspinatus had the least off-flavor, and the M. vastus intermedius had the most intense off-flavor. These data suggest a carcass with one off-flavored muscle is likely to have other off-flavored muscles in the chuck …

Suppression Of Spontaneous Genome Rearrangements In Yeast Dna Helicase Mutants, Kristina H. Schmidt, Richard D. Kolodner

Saccharomyces cerevisiae mutants lacking two of the three DNA helicases Sgs1, Srs2, and Rrm3 exhibit slow growth that is suppressed by disrupting homologous recombination. Cells lacking Sgs1 and Rrm3 accumulate gross-chromosomal rearrangements (GCRs) that are suppressed by the DNA damage checkpoint and by homologous recombination-defective mutations. In contrast, rrm3, srs2, and srs2 rrm3 mutants have wild-type GCR rates. GCR types in helicase double mutants include telomere additions, translocations, and broken DNAs healed by a complex process of hairpin-mediated inversion. Spontaneous activation of the RadS3 checkpoint kinase in the rrm3 mutant depends on the Mec3/Rad24 DNA damage sensors and results from …

Treatment of pulping wastewater by using a novel electrochemical reactor,i.e.the three phase fluidized bed electrode,reactor were invesigated.The effects of air flow rate,applied voltage,pH value and FeSO4 concentration on electrolysis efficiency were investigated.It was clearly shown that the 20 V of voltage,0.3 m3·h-1 of air flow rate,1000 mg L-1 of FeSO4 concentration and pH≈2.5 were its best conditions.COD removal could be 70.1% under these conditions.

A new technique was developed for the deposition of colloidal metal nanoparticles on carbon nanotubes. It involves fast evaporation of a suspension containing sonochemically functionalized carbon nanotubes and colloidal nanoparticles. It was demonstrated that metallic nanoparticles with different sizes and concentrations can be deposited on the carbon nanotubes with only a few agglomerates. the technique does not seem to be limited by what the nanoparticles are, and therefore would be applicable to the deposition of other nanoparticles on carbon nanotubes. PtPd and CoPt3 alloy nanoparticles were used to demonstrate the deposition process. It was found that the surfactants used to …

The Rio Grande has been identified as one of the most endangered rivers in the United States by American Rivers. Biologically, the consequences of the massive anthropogenic alterations of this river have been substantial. The overall aim of this study was to characterize the ecological implications of these alterations in the aquatic food web of the middle Rio Grande (MRG) of New Mexico. Our approach was to analyze the stable isotope composition of current food web components and of historical museum preserved fish specimens.
