Alternative First Exon Splicing Regulates Subcellular Distribution Of Methionine Sulfoxide Reductases, Hwa-Young Kim, Vadim Gladyshev

Background: Methionine sulfoxide reduction is an important protein repair pathway that protects against oxidative stress, controls protein function and has a role in regulation of aging. There are two enzymes that reduce stereospecifically oxidized methionine residues: MsrA (methionine-Ssulfoxide reductase) and MsrB (methionine-R-sulfoxide reductase). In many organisms, these enzymes are targeted to various cellular compartments. In mammals, a single MsrA gene is known, however, its product is present in cytosol, nucleus, and mitochondria. In contrast, three mammalian MsrB genes have been identified whose products are located in different cellular compartments. The purpose of this project was to continue to provide educational efforts on wetlands functions and values, prime wetlands designation, aerial and on-the-ground inventory findings, wildlife habitat, and New Hampshire’s Prime Wetlands laws and rules for the communities of Hampton and Hampton Falls, which was the project focus area. The project incorporated the first phase (phase I) of this wetland inventory report, which focused on the Taylor River Watershed. That initial assessment included the Taylor River as it flows through both Hampton and Hampton Falls, a portion of Ash Brook and Old River as it flows through Hampton, and Grapevine …

This dissertation presents the theory and practice of designing, synthesizing and using peptidomimetics to disrupt protein-protein interactions. Our general strategy is to design and synthesize peptidomimetics that will mimic peptide secondary structures (alpha-helices and beta-sheets). Chapter One is a theoretical examination of the feasibility of using beta-sheet mimics called molecular clamps to inhibit substrate-receptor interactions by blocking the substrate rather than the receptor or enzyme. Several natural and synthetic examples of this approach are given in support of this concept. Background- The effects of pulmonary arterial hypertension on cardiovascular and physical function are well documented. Limited information exists regarding the effects of pulmonary arterial hypertension on cognitive function despite patient reports of problems with memory and attention. Our primary purpose was to determine if a prospectively identified cohort of pulmonary arterial hypertension patients had cognitive sequelae. Our secondary purpose was to determine the relationships between cognitive sequelae and neuropsychological test scores with depression, anxiety, and quality of life. Methods- Forty-six adults with pulmonary arterial hypertension underwent assessment of cognitive function, depression, anxiety, and quality of life using standardized neuropsychological tests …

With start-up funding provided by the Regenstrief Foundation, Purdue University has created the Regenstrief Center for Healthcare Engineering (RCHE) to design, implement, and sustain interdisciplinary solutions to improve the safety, quality, efficiency and accessibility of healthcare delivery systems. RCHE’s primary goal is to bring a systems-analysis approach to improving the processes of healthcare delivery, not to provide tools or techniques for medical research, diagnosis, or treatment. This article will describe RCHE’s business model; that is, how RCHE “engineers” interdisciplinary solutions, using the multiple perspectives of healthcare and multiple project time scales to organize and leverage healthcare delivery transformation. Microcin C is a ribosome-synthesized heptapeptide that contains a modified adenosine monophosphate covalently attached to the C-terminal aspartate. Microcin C is a potent inhibitor of bacterial cell growth. Based on the in vivo kinetics of inhibition of macromolecular synthesis, Microcin C targets translation, through a mechanism that remained undefined. Here, we show that Microcin C is a subject of specific degradation inside the sensitive cell. The product of degradation, a modified aspartyl-adenylate containing an N-acylphosphoramidate linkage, strongly inhibits translation by blocking the function of aspartyl-tRNA synthetase.

With the growing racial and ethnic diversity of the U.S. population and workforce, scholars have begun to address the ways in which coalition building across groups will continue to be necessary but will become even more complex. The growing scholarship has focused on analyzing how best to promote effective coalition building. Thus far, scholars have not examined what that growing racial and ethnic diversity will mean in the individuated context of racial and ethnic discrimination claims. In other words, what will anti-discrimination litigation look like when all the parties involved are non-White but a racial hierarchy is alleged to exist …

Stress-activated protein kinase/c-Jun kinase (SAPK/JNK) is thought to be necessary for preimplantation embryonic development (Maekawa et al., 2005). However, media increases SAPK/JNK phosphorylation and these levels negatively correlate with embryonic development (Wang et al., 2005). Culture-induced stress could confuse analysis of the role of SAPK in development. In this study, we tested how SAPK/JNK inhibitors influence embryonic development in optimal and non-optimal media and define the contribution of cell survival and proliferation to the embryonic response to these media. SAPK/JNK inhibitors retard embryonic development in suboptimal Ham’s F10, but improve development in optimal potassium (K+) simplex optimized media (KSOM) +AA. …

A Comparative Genomic Analysis Of Chlorella Nc64a Virus Ny-2a And Chlorella Pbi Virus Mt325 From The Family Phycodnaviridae, Lisa A. Fitzgerald

The family Phycodnaviridae consists of a morphologically similar but genetically diverse group of large dsDNA viruses which infect both fresh and marine water eukaryotic algae. Two viruses, the 370 kb Chlorella NC64A virus NY-2A and the 313 kb Chlorella Pbi virus MT325, from the family Phycodnaviridae, genus Chlorovirus, were sequenced, analyzed, and compared to the prototype chlorella virus PBCV-1. The NY-2A genome, the largest chlorella viral genome sequenced to date, contains 886 open reading frames (ORFs) of 65 codons or larger and encodes 404 putative proteins and 7 tRNAs. We present a conceptual approach for evaluating the biological and hydrological controls of nutrient removal in different sized rivers within an entire river network. We emphasize a per unit area biological parameter, the nutrient uptake velocity (νf), which is mathematically independent of river size in benthic dominated systems. Standardization of biological parameters from previous river network models to νf reveals the nature of river size dependant biological activity in these models. We explore how geomorphic, hydraulic, and biological factors control the distribution of nutrient removal in an idealized river network, finding that larger rivers within a basin potentially exert considerable …

A Feminist Perspective On The Precautionary Principle And The Problem Of Endocrine Disruptors Under Neoliberal Globalization Policies, Erica Hesch Anstey

Industrialization and "development" during the last 200 years have led to an increase of pesticides, an intensified use of synthetic chemicals, higher levels of environmental pollution, and more exposure to hazardous working conditions. Environmental toxins, many of which are endocrine disruptors, are stored in fat tissue, increasing reproductive health risks for both women and men. Women’s bodies are particularly vulnerable as sites for creating, growing, feeding, and nurturing the next generation. And yet, women’s lives are consistently devalued, especially in a capitalist economy, so that a woman’s rights to her own reproductive health are no longer guaranteed.
