The importance of polarized growth for fungi has elicited significant effort directed at better understanding underlying mechanisms of polarization, with a focus on yeast systems. At sites of tip growth, multiple protein complexes assemble and coordinate to ensure that incoming building material reaches the appropriate destination sites, and polarized growth is maintained. One of these complexes is the polarisome that consists of Spa2, Bud6, Pea2, and Bni1 in Saccharomyces cerevisiae. Filamentous hyphae differ in their development and life style from yeasts and likely regulate polarized growth in a different way. This is expected to reflect on the composition and …

High-temperature, controlled incineration and thermal treatment of contaminated soils, sediments, and wastes at Superfund sites are often preferred methods of remediation of contaminated sites under the Comprehensive Environmental Response, Compensation, and Liability Act of 1980 and related legislation. Although these methods may be executed safely, formation of toxic combustion or reaction by-products is still a cause of concern. Emissions of polycyclic aromatic hydrocarbons (PAHs); chlorinated hydrocarbons (CHCs), including polychlorinated dibenzo-p-dioxins and dibenzofurans; and toxic metals (e.g., chromium VI) have historically been the focus of combustion and health effects research. Tumor-associated eosinophilia has been observed in numerous human cancers and several tumor models in animals; however, the details surrounding this eosinophilia remain largely undefined and anecdotal. We used a B16-F10 melanoma cell injection model to demonstrate that eosinophil infiltration of tumors occurred from the earliest palpable stages with significant accumulations only in the necrotic and capsule regions. Furthermore, the presence of diffuse extracellular matrix staining for eosinophil major basic protein was restricted to the necrotic areas of tumors, indicating that eosinophil degranulation was limited to this region. Antibody-mediated depletion of CD4+ T cells and adoptive transfer of eosinophils suggested, respectively, …

Genes encoding reproductive proteins often diverge rapidly due to positive selection on nucleotide substitutions. While this general pattern is well established, the extent to which specific reproductive genes experience similar selection in different clades has been little explored, nor have possible targets of positive selection other than nucleotide substitutions, such as indels, received much attention. Here, we inspect for the signature of positive selection in the genes encoding five accessory gland proteins (Acps) (Acp26Aa, Acp32CD, Acp53Ea, Acp62F, and Acp70A) originally described from Drosophila melanogaster but with recognizable orthologues in the D. pseudoobscura subgroup. Most MALDI instrumentation uses UV lasers. We have designed a MALDI-IM-oTOF-MS which employs both a Nd:YAG laser pumped optical parametric oscillator OPOTEK, λ = 2.8-3.2 μm at 20 Hz) to perform IR-LDI or IR-MALDI and a Nd:YLF laser (Crystalaser, λ = 249 nm at 200 Hz) for the UV. Ion mobility (IM) gives a fast separation and analysis of biomolecules from complex mixtures in which ions of similar chemical type fall along well-defined "trend lines". Our data shows that ion mobility allows multiply charged monomers and multimers to be resolved; thus, yielding pure spectra of the singly charged protein ion …

Using an ecoregional approach, multimetric indices were developed to describe stream condition in each geographically distinct region of Georgia. Collected from predetermined reference and impaired condition streams, macroinvertebrate data were used to develop multimetric indices. Multimetric indices identified a range of stream conditions, as well as, the overall health of aquatic ecosystems. Based on multimetric indices, the Multimetric Rating System grouped streams according to overall stream condition. The Multimetric Rating System used multiple indices to describe various facets of stream condition. Quantifying periphyton (attached algal) contributions to autotrophic production in lakes is confounded by properties of substratum that affect community biomass (as chlorophyll content) and productivity. We compared chlorophyll content and productivity of natural algal communities (phytoplankton, epipelon, epilithon, epixylon, and epiphyton) experiencing high (>10%) incident radiation in lakes in the US, Greenland, and Quebec, Canada. Chlorophyll content and productivity differed significantly among regions, but they also differed consistently among communities independent of region. Chlorophyll content of periphyton on hard substrata (rocks and wood) was positively related to water-column total P (TP), whereas chlorophyll content of algae on sediment (epipelon) …

Elasmosaur cervical vertebrae are common fossils, but their taxonomic utility is limited due to a lack of understanding concerning their shape within and among taxa. In this paper, we analyze data from complete elasmosaur necks in an attempt to quantify and understand the variation in centrum dimensions. In accord with previous studies, variation in cervical centrum shape is found to stem from at least three sources: ontogeny, intracolumn variation, and intercolumn or taxonomic variation. Ontogenetic variability seems reminiscent of that seen in Cryptoclidus, with an overall positive allometry in the length of all centra that is accentuated in the mid-cervical …

Background and Objectives: This study was designed to examine modes of cell death after photodynamic therapy (PDT). Study Design: Murine leukemia L1210 cells and human prostate Bax-deficient DU145 cells were examined after PDT-induced photodamage to the endoplasmic reticulum (ER). Phase contrast, fluorescence and electron microscopy were used to identify changes in cellular morphology, chromatin condensation, loss of mitochondrial membrane potential, and formation of phagolysosomes. Western blots were used to assess the processing of LC3-I to LC3-II, a marker for autophagy. Inhibitors of apoptosis and/or autophagy were used to delineate the contributions of the two pathways to the effects of PDT. …

Most birds develop brood patches before incubation; epidermis and dermis in the brood patch region thicken, and the dermal connective tissue becomes increasingly vascularized and infiltrated by leukocytes. However, current dogma states that waterfowl incubate without modifications of skin within the brood patch region. The incubation periods of lesser snow geese (Chen caerulescens caerulescens; hereafter called snow geese) and Ross's geese (C. rossii) are 2-6 days shorter than those of other goose species; only females incubate. Thus, we hypothesized that such short incubation periods would require fully developed brood patches for sufficient heat transfer from incubating parents to eggs. We …

A molecular phylogenetic analysis recovers a pattern consistent with a drift vicariance scenario for the origin of Greater Antillean cichlids. This phylogeny, based on mitochondrial and nuclear genes, reveals that clades on different geographic regions diverged concurrently with the geological separation of these areas. Middle America was initially colonized by South American cichlids in the Cretaceous, most probably through the Cretaceous Island Arc. The separation of Greater Antillean cichlids and their mainland Middle American relatives was caused by a drift vicariance event that took place when the islands became separated from Yucatan in the Eocene. River corridor wetland restoration and freshwater diversion from the lower Mississippi River are being considered as two major options to reduce nitrogen input to the Gulf of Mexico. However, it is largely uncertain how much nitrogen can actually be retained from the overflowing waters by these wetland systems. This study quantified the nitrogen inflow and outflow for the largest distributary basin of the Mississippi River, the Atchafalaya River Swamp basin. Researchers have become increasingly interested in the social context of chronic pain conditions. The purpose of this article is to provide an integrated review of the evidence linking marital functioning with chronic pain outcomes including pain severity, physical disability, pain behaviors, and psychological distress. We first present an overview of existing models that identify an association between marital functioning and pain variables. We then review the empirical evidence for a relationship between pain variables and several marital functioning variables including marital satisfaction, spousal support, spouse responses to pain, and marital interaction. [Excerpt] "When Charles Darwin published On the Origin of Species By Means of Natural Selection in 1859, it sparked some of the most contentious debates in American intellectual history, debates that continue to rage today. Although these debates have numerous political ramifications, the question posed in this paper is narrow: Does the Establishment Clause permit a particular assessment of current evolutionary theory – intelligent design (“ID”) – to be taught as science in American elementary and secondary public schools? Coastal watersheds and nearshore marine areas are the most valuable and dynamic places on Earth. Human population growth is great in these regions, which are home to some of the most sensitive habitats in the world. Coastal areas provide more than half of the overall service value derived from the global environment (Costanza et al., 1997).
