A Survey Of Nuclear Ribosomal Internal Transcribed Spacer Substitution Rates Across Angiosperms: An Approximate Molecular Clock With Life History Effects, Kathleen M. Kay, Justen B. Whittall, Scott A. Hodges

Biology

Background: A full understanding of the patterns and processes of biological diversification requires the dating of evolutionary events, yet the fossil record is inadequate for most lineages under study. Alternatively, a molecular clock approach, in which DNA or amino acid substitution rates are calibrated with fossils or geological/climatic events, can provide indirect estimates of clade ages and diversification rates. The utility of this approach depends on the rate constancy of molecular evolution at a genetic locus across time and across lineages. Although the nuclear ribosomal internal transcribed spacer region ( nrITS) is increasingly being used to infer clade ages in …

Lack Of Il-15 Results In The Suboptimal Priming Of Cd4+ T Cell Response Against An Intracellular Parasite, Crescent L. Combe, Magali M. Moretto, Joseph D. Schwartzman, Jason P. Gigley, David J. Bzik, Imtiaz A. Khan Apr 2006

Lack Of Il-15 Results In The Suboptimal Priming Of Cd4+ T Cell Response Against An Intracellular Parasite, Crescent L. Combe, Magali M. Moretto, Joseph D. Schwartzman, Jason P. Gigley, David J. Bzik, Imtiaz A. Khan

Dartmouth Scholarship

IFN-gamma-producing CD4+ T cells, although important for protection against acute Toxoplasma gondii infection, can cause gut pathology, which may prove to be detrimental for host survival. Here we show that mice lacking IL-15 gene develop a down-regulated IFN-gamma-producing CD4+ T cell response against the parasite, which leads to a reduction in gut necrosis and increased level of survival against infection. Moreover, transfer of immune CD4+ T cells from WT to IL-15-/- mice reversed inhibition of gut pathology and caused mortality equivalent to levels of parental WT mice. Down-regulated CD4+ T cell response in the absence of IL-15, manifested as reduced …

Defocused Orientation And Position Imaging (Dopi) Of Myosin V, Rolfe G. Petschek Apr 2006

Defocused Orientation And Position Imaging (Dopi) Of Myosin V, Rolfe G. Petschek

Faculty Scholarship

The centroid of a fluorophore can be determined within 1.5-nm accuracy from its focused image through fluorescence imaging with one-nanometer accuracy (FIONA). If, instead, the sample is moved away from the focus, the point-spread-function depends on both the position and 3D orientation of the fluorophore, which can be calculated by defocused orientation and position imaging (DOPI). DOPI does not always yield position accurately, but it is possible to switch back and forth between focused and defocused imaging, thereby getting the centroid and the orientation with precision. We have measured the 3D orientation and stepping behavior of single bifunctional rhodamine probes …

Lysine Residues In N-Terminal And C-Terminal Regions Of Human Histone H2a Are Targets For Biotinylation By Biotinidase, Yap Ching Chew, Gabriela Camporeale, Nagarama Kothapalli, Gautam Sarath, Janos Zempleni Apr 2006

Lysine Residues In N-Terminal And C-Terminal Regions Of Human Histone H2a Are Targets For Biotinylation By Biotinidase, Yap Ching Chew, Gabriela Camporeale, Nagarama Kothapalli, Gautam Sarath, Janos Zempleni

Department of Nutrition and Health Sciences: Faculty Publications

In eukaryotic cell nuclei, DNA associates with the core histones H2A, H2B, H3 and H4 to form nucleosomal core particles. DNA binding to histones is regulated by posttranslational modifications of N-terminal tails (e.g., acetylation and methylation of histones). These modifications play important roles in the epigenetic control of chromatin structure. Recently, evidence that biotinidase and holocarboxylase synthetase (HCS) catalyze the covalent binding of biotin to histones has been provided. The primary aim of this study was to identify biotinylation sites in histone H2A and its variant H2AX. Secondary aims were to determine whether acetylation and methylation of histone H2A affect …

Cathexellanea, Pir Rothenberg Apr 2006

Cathexellanea, Pir Rothenberg

Theses and Dissertations

Cathexellanea is a portmanteau word blending "cathexis," a psychoanalytical term meaning an investment of mental or emotional energy on a person, object or idea; and "miscellanea," a collection of objects or writings. One theme running through these stories concerns characters attempt to define themselves through their desires. "Bottomless" explores the idea of emotional and physical fulfillment. "Croquembouche" revolves around self-loathing and corruptibility. In "Destripado," the protagonist discovers that one way to dispel fear is to become the thing that causes it. "Ghosts" asks whether it is a past lover the protagonist longs for, or her phantoms. "Versions" deals with the …

Perception And Implementation Of Sustainable/Green Design In India, Shalini Nagaraj Magadi Apr 2006

Perception And Implementation Of Sustainable/Green Design In India, Shalini Nagaraj Magadi

Master's Theses and Doctoral Dissertations

Profound effects of global warming, species extinction, pollution of air and water, resource depletion, and population growth demand that India, the second most populated country in the world, address the need for ecological responsibility. This phenomenological study explored awareness and perceptions of green/sustainable (G/S) design in India’s design community. Architects in the five geographic regions were interviewed. All were familiar with S/G design and said knowledge was “very important.” However, none felt “well-versed.” They believed lack of client awareness (49%) and materials (28%) were the major problems encountered. Although the vast majority was willing to promote S/G design, none had …
