Use Of Box-Pcr Subtyping Of Escherichia Coli And Enterococcus Spp. To Determine The Source Of Microbial Contamination At A Florida Beach, Miriam J. Brownell

USF Tampa Graduate Theses and Dissertations

Siesta Key Beach, located on the Gulf Coast of Florida, is frequently mentioned among the top ten beaches in the US. In summer 2004, high levels of indicator bacteria caused health warnings to be posted, and a storm drainage system was implicated as a possible source of microbial contamination. A study was initiated to determine whether indicator bacteria that persisted in the stormwater system could contribute to high microbial loads in receiving waters. Two sampling events, one within 48 hours of a rain event and the other during dry conditions, were conducted. Water and sediment samples were taken at various …

Ligand-Induced Conformational Changes Of Extracellular Loop Two In The Dopamine Transporter, Jon D. Gaffaney Jun 2006

Ligand-Induced Conformational Changes Of Extracellular Loop Two In The Dopamine Transporter, Jon D. Gaffaney

Theses and Dissertations

The dopamine transporter (DAT) is a transmembrane spanning protein responsible for the clearance of dopamine from the dopaminergic synapse. Uptake blockers such as cocaine inhibit transport resulting in elevated levels of dopamine in the synapse. DAT is predicted to contain twelve transmembrane spanning domains (TMs) connected by alternating intra- and extracellular loops, however, the spatial arrangement of these regions is unknown. Extracellular loop 2 (EL2) is composed of approximately 80 amino acids and connects TM3 and TM4, two regions associated with uptake blocker binding. Using limited proteolysis we analyzed the aqueous accessibility of EL2 in the presence or absence of …

The Characterization And Distribution Of Renshaw Cells In The Rat Spinal Cord, Richard A. Zaruba Jun 2006

The Characterization And Distribution Of Renshaw Cells In The Rat Spinal Cord, Richard A. Zaruba

Theses and Dissertations

There is an abundance of electrophysiological and pharmacological data describing Renshaw cells (RC), however comparatively little is known about their distribution, metabolism, ionic regulation, modulation or exact role in motor behaviors. In this dissertation enzyme histochemistry and immunohistochemistry in conjunction with light, fluorescent and confocal microscopic methodologies were used to address the hypothesis that "the distribution of RC is contingent on their functional role in specific α motoneuronal pools and their unique electrophysiological and functional characteristics are reflected by their neurochemistry and synaptic inputs." The highest concentration of RC (number of RC per mm2) was found in the thoracic region …

A Device To Continuously Monitor Dissolved Oxygen And Temperature At User-Selected Depths And Locations In Culture Ponds, Bartholomew W. Green, Philip R. Pearson

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Continuous measurement of dissolved oxygen concentration and temperature at two depths in the water column were needed for a 2004 field study conducted during actual harvests of channel catfish Ictalurus punctatus and for a small-scale research study conducted in 2005. In the on-farm field study, we collected data in grading nets loaded with high densities of channel catfish. The small-scale study was conducted in 0.1-acre channel catfish research ponds. Commercially available dissolved oxygen and temperature monitoring units did not meet study requirements, so we developed a free-floating data collection unit composed of state-of-the-art sensors, a data logger, and readily available …

Prevalence Of Shiga Toxin–Producing Escherichia Coli And Salmonella Enterica In Rock Pigeons Captured In Fort Collins, Colorado, Kerri Pedersen, Larry Clark, William F. Andelt, M.D. Salman

United States Department of Agriculture Wildlife Services: Staff Publications

The potential role of rock pigeons (Columba livia) in the epidemiology of shiga toxin– producing Escherichia coli (STEC) and Salmonella enterica is unclear. Our objective was to determine the prevalence of STEC and S. enterica in pigeons at urban and dairy settings as a function of season. Prevalence of STEC and S. enterica was estimated by bacteriologic culture of cloacal swabs collected from pigeons trapped at urban and dairy locations in and around Fort Collins, Colorado from January to November 2003. Presumptive E. coli isolates were tested for the presence of virulence genes SLT-1, SLT-2, eae, hlyA …

Mixed Messages: The Supreme Court’S Conflicting Decisions On Juries In Death Penalty Cases, Ken Miller, David Niven Jun 2006

Mixed Messages: The Supreme Court’S Conflicting Decisions On Juries In Death Penalty Cases, Ken Miller, David Niven

ExpressO

The right to a jury determination of a capital defendant's fate has expanded recently. The era of judges making factual determinations then determining whether to apply a death sentence or judges having the power to overrule a jury's life sentence to impose death is over. The expanded right to access a jury and have it hold determinative power over a defendant's life has not, however, been accompanied by commensurate attention to the instructions that guide those jurors through the applicable law toward their verdict. Nor have adequate procedures been designed to produce a truly representative jury panel. In brief, the …

Vertical Migration Of The Toxic Dinoflagellate Karenia Brevis And The Impact On Ocean Optical Properties, Oscar Schofield, John Kerfoot, Kevin Mahoney, Mark A. Moline, Matthew Oliver, Steven Lohrenz, Gary Kirkpatrick Jun 2006

Vertical Migration Of The Toxic Dinoflagellate Karenia Brevis And The Impact On Ocean Optical Properties, Oscar Schofield, John Kerfoot, Kevin Mahoney, Mark A. Moline, Matthew Oliver, Steven Lohrenz, Gary Kirkpatrick

Biological Sciences

Vertical migration behavior is found in many harmful algal blooms; however, the corresponding impact on ocean optical properties has not been quantified. A near-monospecific population of the dinoflagellate Karenia brevis was encountered off the west coast of Florida. The community was tracked for 24 hours by following a Lagrangian drifter deployed at the beginning of the experiment. A suite of inherent optical and cellular measurements was made. Over the 24 hour period, the K. brevis population increased during the day with concentrations peaking in the late afternoon (1600 local daylight time) in the upper 2 m of the water column. …

"Quartet:" Dissecting A Story, Elizabeth H. Williams '06 Jun 2006

"Quartet:" Dissecting A Story, Elizabeth H. Williams '06

Honors Projects

From the beginning of my investigation of Quartet, the text has intimidated me; after all, the script is infused with a multitude of literary, historical, and philosophical references and quotes all of which Heiner Mueller, himself, prefers to dissociate himself from. "The theatre has nothing to do with ideas," Mueller once stated. Paradoxically his theatre is one that is full of ideas and whether Mueller chose, in his own lifetime, to recognize the ideas of his theatre or not, it is precisely his provocative ideas and images that has been the basis for this paper. In his dramatic text, Quartet, …

Investigating The Role Of Small, Acid-Soluble Spore Proteins (Sasps) In The Resistance Of Clostridium Perfringens Spores To Heat., Deepa Raju, Michael Waters, Peter Setlow, Mahfuzur R Sarker

SoM Articles

BACKGROUND: Clostridium perfringens type A food poisoning is caused by enterotoxigenic C. perfringens type A isolates that typically possess high spore heat-resistance. The molecular basis for C. perfringens spore heat-resistance remains unknown. In the current study, we investigated the role of small, acid-soluble spore proteins (SASPs) in heat-resistance of spores produced by C. perfringens food poisoning isolates.

RESULTS: Our current study demonstrated the presence of all three SASP-encoding genes (ssp1, 2 and 3) in five surveyed C. perfringens clinical food poisoning isolates. beta-Glucuronidase assay showed that these ssp genes are expressed specifically during sporulation. Consistent with these expression results, our …

Determination Of Particle (Pm10 And Pm2.5) And Gas-Phase Ammonia (Nh3) Emissions From A Deep-Pit Swine Operation Using Arrayed Field Measurements And Inverse Gaussian Plume Modeling, Randal S. Martin, Vishal Doshi, Kori Moore Jun 2006

Determination Of Particle (Pm10 And Pm2.5) And Gas-Phase Ammonia (Nh3) Emissions From A Deep-Pit Swine Operation Using Arrayed Field Measurements And Inverse Gaussian Plume Modeling, Randal S. Martin, Vishal Doshi, Kori Moore

Space Dynamics Laboratory Publications

The contribution of agricultural emissions of primary (direct) and secondary (precursor) pollutants to air quality is rapidly being recognized as an important fraction of local and regional air pollution budgets. However, a significant uncertainty still exists in the magnitude and rate of these types of emissions, especially under “in field” conditions common within the central and western United States. Described herein are the results of a study conducted at a deep-pit swine production facility in central Iowa. The facility consisted of three separate, parallel barns, each housing around 1,250 pigs with an average weight of approximately 90 pounds per animal. …

Oedipus Rex: Metaphysics And The Fundamental Human Struggle, Joseph Mack Lorenzo Jun 2006

Oedipus Rex: Metaphysics And The Fundamental Human Struggle, Joseph Mack Lorenzo

Theses and Dissertations

The fundamental human struggle is a spiritual struggle demonstrated by a metaphysical analysis of the drama Oedipus Rex. Drama presents a mimesis of human action and dramatic plots are comprised of these physical actions. This fundamental dramatic action consists of the embodied action of the human being (i.e., substance), comprised of body and soul (i.e., matter and form). Through these embodied actions Aristotelian metaphysical first principles become tangible for the audience. This metaphysical reality is both fundamental to the human being and universal to the human experience. Therefore the fundamental human struggle is a spiritual struggle, viz., the exercise …

Intermediary Metabolism And Life-History Trade-Offs: Differential Metabolism Of Amino Acids Underlies The Dispersal-Reproduction Trade-Off In A Wing-Polymorphic Cricket, Anthony J. Zera, Zhangwu Zhao Jun 2006

Intermediary Metabolism And Life-History Trade-Offs: Differential Metabolism Of Amino Acids Underlies The Dispersal-Reproduction Trade-Off In A Wing-Polymorphic Cricket, Anthony J. Zera, Zhangwu Zhao

Anthony Zera Publications

Although the differential flow of metabolites through alternate pathways of intermediary metabolism is thought to be an important functional cause of life-history trade-offs, this phenomenon remains understudied. Using a radiolabeled amino acid, we quantified genetic differences in in vivo amino acid metabolism between morphs of the wing-polymorphic cricket Gryllus firmus that trade off early-age reproduction and dispersal capability. Lines selected for the flight-capable morph, which delays reproduction, oxidized a greater proportion of radiolabeled glycine and converted a greater amount into somatic lipid, mainly triglyceride (flight fuel). By contrast, lines selected for the flightless, reproductive morph converted a substantially greater proportion …

Effectiveness Of A Computer–Facilitated, Interactive Social Skills Training Program For Boys With Attention Deficit Hyperactivity Disorder, Kevin Fenstermacher, Daniel Olympia, Susan M. Sheridan Jun 2006

Effectiveness Of A Computer–Facilitated, Interactive Social Skills Training Program For Boys With Attention Deficit Hyperactivity Disorder, Kevin Fenstermacher, Daniel Olympia, Susan M. Sheridan

Department of Educational Psychology: Faculty Publications

Children with attention deficit hyperactivity disorder (ADHD) are at significant risk for a variety of comorbid conditions, including social skills deficits. Although interventions addressing various aspects of social difficulties with these children have been developed, few researchers have integrated new technology with existing social skills intervention literature and investigated such approaches empirically. The purpose of this study was to investigate the effectiveness of a computer-mediated social skills training program for children diagnosed with ADHD. The program presented specific social skill sequences to four children with ADHD in a variety of computer–facilitated formats with video peer modeling, social problem–solving, and reinforcement …

Modelling Wildlife Rabies: Transmission, Economics, And Conservation, Ray T. Sterner, Graham C. Smith Jun 2006

Modelling Wildlife Rabies: Transmission, Economics, And Conservation, Ray T. Sterner, Graham C. Smith

United States Department of Agriculture Wildlife Services: Staff Publications

Rabies is a fatal zoonotic disease of mammals; it exacerbates the uncertainty of conserving populations of some threatened mammals (TM). Modeling affords an inexpensive, a priori way of studying key parameters of wildlife rabies transmission, rabies management economics, and TM conservation. Numerous models of rabies transmission have been published. Linear density dependent models predicted that a threshold density (KT 6 1.0), possibly attained by culling or contraception, would eliminate an epizootic through reduced contacts among host animals. Density independent models predicted less advantage of culling and contraception in rabies control due to limited contacts among territorial host animals. Recent stochastic, …

An Integrated Approach To Measuring Emissions From Confined Animal Feeding Operations At The Whole Facility Scale, G. E. Bingham, J. Hatfiels, J. H. Prueger, T. D. Wilkerson, V. V. Zavyalov, R. L. Pfeiffer, L. Hipps, R. Martin, P. Silva, W. Eichinger

Space Dynamics Laboratory Publications

Agricultural operations produce a variety of particulates and gases that influence air quality. Agriculture, through wind erosion, tillage and harvest operations, burning, diesel-powered machinery and animal production operations, is a source of particulate matter that can enter human lungs and cause pulmonary problems. Animal production operations can be a source of gaseous emissions such as ammonia, odor-causing volatile organic compounds, hydrogen sulfide, greenhouse gases (methane, nitrous oxides) and airborne pathogens. These emissions can negatively impact human health, property values, and the environment. The presence of buildings and other structures often make whole facility measurement capability a requirement for understanding the …
