The Drosophila Boundary Element-Associated Factors Beaf-32a And Beaf-32b Affect Chromatin Structure, Matthew K. Gilbert, Yian Yee Tan, Craig M. Hart Aug 2006

The Drosophila Boundary Element-Associated Factors Beaf-32a And Beaf-32b Affect Chromatin Structure, Matthew K. Gilbert, Yian Yee Tan, Craig M. Hart

Faculty Publications

Binding sites for the Drosophila boundary element-associated factors BEAF-32A and -32B are required for the insulator activity of the scs' insulator. BEAF binds to hundreds of sites on polytene chromosomes, indicating that BEAF-utilizing insulators are an important class in Drosophila. To gain insight into the role of BEAF in flies, we designed a transgene encoding a dominant-negative form of BEAF under GAL4 UAS control. This BID protein encompasses the BEAF self-interaction domain. Evidence is provided that BID interacts with BEAF and interferes with scs' insulator activity and that BEAF is the major target of BID in vivo. BID expression during …

Health In Cuba, Richard S. Cooper, Joan F. Kennelly, Pedro Orduñez-Garcia

Faculty Publications

The poorer countries of the world continue to struggle with an enormous health burden from diseases that we have long had the capacity to eliminate. Similarly, the health systems of some countries, rich and poor alike, are fragmented and inefficient, leaving many population groups underserved and often without health care access entirely. Cuba represents an important alternative example where modest infrastructure investments combined with a well-developed public health strategy have generated health status measures comparable with those of industrialized countries. Areas of success include control of infectious diseases, reduction in infant mortality, establishment of a research and biotechnology industry, and …

Sustainable Development Or Just Plain Development? An Analysis Of Sustainability Indicators For Chattanooga, Tennessee, Cheryl Horn Aug 2006

Sustainable Development Or Just Plain Development? An Analysis Of Sustainability Indicators For Chattanooga, Tennessee, Cheryl Horn

Masters Theses and Doctoral Dissertations

The city of Chattanooga, Tennessee, and the surrounding area has undergone a notable transformation in recent decades. Many of these changes have been used to promote the city as an environmentally friendly place to live, and the phrase sustainable development is frequently used to describe local growth and planning. However, no comprehensive analysis has been undertaken to determine whether Chattanooga is truly headed toward becoming a sustainable community in terms of environmental, social, and economic considerations. Based on the concept of sustainable development, as defined by the United Nations and others, and on sustainability studies conducted in four other cities, …

Identification And Characterization Of Cellular Targets Of The Bacterial Cytotoxin Yopt In Saccharomyces Cerevisiae, Paul David Caccamo Aug 2006

Identification And Characterization Of Cellular Targets Of The Bacterial Cytotoxin Yopt In Saccharomyces Cerevisiae, Paul David Caccamo

Masters Theses

Human pathogenic Yersinia spp. utilize Type III Secretion to deliver six effector proteins into host cells. The injected proteins resemble eukaryotic signal transduction molecules and are capable of disrupting signaling pathways in order to subvert the functions of the targeted cell or disrupt communication with surrounding cells. As a result, bacterial invaders are not only able to survive the threat of a host immune response, but can thrive despite it. YopT is a cysteine protease that cleaves RhoGTPases, releasing them from the membrane and thus from their role as initiators of signaling pathways, including the nucleation of actin and …

Stable Hydrogen Isotopic Composition Of Fishes Reflects That Of Their Environment, Gregory Whitledge, Brett Johnson, Patrick Martinez Aug 2006

Stable Hydrogen Isotopic Composition Of Fishes Reflects That Of Their Environment, Gregory Whitledge, Brett Johnson, Patrick Martinez

Publications

Otolith microchemistry and isotopic analyses have emerged as effective techniques for providing insights into fish environmental history that are difficult to obtain by other means. Stable hydrogen isotope ratio (2H/1H or D/H, expressed as δD) is a possible environmental marker that has not been employed in fish provenance research, although it has been applied as a natural tracer of terrestrial organism migrations. We illustrate the potential of δD to serve as a new natural marker of fish environmental history by demonstrating that significant linear relationships (r2 >= 0.97) exist between fish otolith and muscle δD and δD of waters that …

Learned Fear And Reaction To Novel Stimuli: Behavioral And Hormonal Stress Responses In The Maternal Rat, Brandi Nicole Rima Aug 2006

Learned Fear And Reaction To Novel Stimuli: Behavioral And Hormonal Stress Responses In The Maternal Rat, Brandi Nicole Rima

Master's Theses

The present thesis examines the relationship between reproductive experience and the behavioral, neural, and hormonal processes of learned fear in the female rat. Multiple research models indicate that reproductive experience functions to decrease the female's stress response in potentially harmful environments, thus providing her with numerous survival benefits, including decreased fearfulness, increased aggression, and refined hunting skills. Based on existing understandings of maternal experience and unconditioned fear, this study was designed to determine how nulliparous (no reproductive experience, NP), primiparous (one reproductive experience, PP) and multiparous (more than one reproductive experience, MP) rats comparatively respond to a Pavlovian paradigm of …

Salicylic Acid Activates Sigma Factor B By Rsbu-Dependent And -Independent Mechanisms, Marco Palma, Arnold Bayer, Leon I. Kupferwasser, Tammy Joska, Michael R. Yeaman, Ambrose Cheung

Dartmouth Scholarship

Salicylic acid (SAL) may impact Staphylococcus aureus virulence by activating the sigB operon (rsbU-V-W-sigB), thus leading to reductions in alpha-toxin production and decreased fibronectin binding (L. I. Kupferwasser et al., J. Clin. Investig. 112:222-233, 2003). As these prior studies were performed in strain RN6390 (an rsbU mutant) and its rsbU-repaired variant, SH1000, the current investigation was designed to determine if the SAL effect occurs via rsbU- and/or rsbV-dependent pathways in an rsbU-intact S. aureus strain (FDA486). We thus quantified the transcription from two sigB-dependent promoters (asp23 and sarA P3) in FDA486 in response to SAL exposure in vitro, using isogenic …

La RepresentacióN Del Indio En La NovelíStica De José MaríA Arguedas, Teresa Gizzi Aug 2006

La RepresentacióN Del Indio En La NovelíStica De José MaríA Arguedas, Teresa Gizzi

Theses, Dissertations and Culminating Projects

Mary Shelley And The Early Goddess, Anne Schreuder Aug 2006

Mary Shelley And The Early Goddess, Anne Schreuder

Theses, Dissertations and Culminating Projects

For my thesis, I intend to focus on elements of Greek idealism in Mary Shelley’s Valperga, Frankenstein, and The Last Man. I will define “classicism” and seek to understand the ideals of the ancients that are concerned with the assertion of a masculine identity and fear the interference of the female voice in the quest for that assertion. I will show how Mary Shelley’s revisionist history undermines the classical ideals of masculinity in the search for the lost feminine. I will use Shelley’s two plays, Proserpine and Midas to support my claim that Shelley is interested in revisionist history of …

Rain Town, Melanie Crow Aug 2006

Rain Town, Melanie Crow

Dissertations

My project, a collection of poetry, examines both loss and transformation. Personal loss informs the poems, but the work also addresses how the speaker is accorded insight into the world because of loss and is transformed. The work takes into account mythology, science, and the act of writing in order to understand what it means to live in mutability. Many of the poems demonstrate an acute awareness of the body. Robert Lowell and Jorie Graham are both writers who have informed my poems in this way; both explore a dual consciousness of loss and renewal and both represent moments of …

Vertebral Age Estimation: An Examination Of The Seventh Cervical, Seventh Thoracic, And Fourth Lumbar Vertebrae, Holly Hernandez

Masters Theses

This project examines the utility of vertebral osteophyte development to estimate age at death. Skeletal joint degeneration is a common method for estimating individuals' age at death, however studies regarding vertebral degeneration and age estimation are lacking. My study follows the work of Stewart (1958) in comparing stages of osteophytic lipping of vertebral bodies to age at death.

I examined the seventh cervical (C7), seventh thoracic (T7), and fourth lumbar (L4) vertebrae in 100 individuals of known ancestry, sex, and age at death from the Hamann-Todd skeletal collection. Degenerative stages and composite scores were analyzed through descriptive statistics and chi …

Prolonged, extensive exposure to asphalt fume has been associated with several adverse health effects. Inhaled polycyclic aromatic hydrocarbons (PAHs) from asphalt fume exposure are of concern. The objective of this study was to characterize both qualitative and quantitative differences between fumes generated at 150°C and 180°C using a well controlled laboratory road paving fume generation system. Fumes were characterized by (1) total vapor and particulate concentration, (2) simulated boiling point profile and (3) specific PAH content. The mean concentrations of the vapor fractions generated at 180°C and 150°C were 23.3 mg/m3 and 11.2 mg/m3, respectively, demonstrating a statistically significant increase …

Measuring Chemotaxis In Borrelia Burgdorferi The Lyme Disease Spirochete, Richard Gerrit Bakker Aug 2006

Measuring Chemotaxis In Borrelia Burgdorferi The Lyme Disease Spirochete, Richard Gerrit Bakker

Graduate Theses, Dissertations, and Problem Reports (ETD)

Borrelia burgdorferi is the spirochete causative agent of Lyme disease. The chemotaxis and motility systems of these bacteria are far less well described than that of Escherichia coli or Salmonella enterica. This dissertation explores the role of the CheA proteins in the chemotactic response and describes the first defined attractants for B. burgdorferi.;In order to test hypotheses, we developed or optimized three protocols. To characterize the motion of cells, two motion tracking systems were optimized. The Hobson BacTracker allowed for tracking cell motions in real time. This hardware/software chimera, while powerful for the specific application, utilizes a cumbersome interface. Therefore, …

The Effect Of A Diet Supplemented With Flaxseed Oil On The Lipid Content And Fatty Acid Profile Of Rainbow Trout (Oncorhynchus Mykiss) Muscle Tissue, Jason P. Nguyen Aug 2006

The Effect Of A Diet Supplemented With Flaxseed Oil On The Lipid Content And Fatty Acid Profile Of Rainbow Trout (Oncorhynchus Mykiss) Muscle Tissue, Jason P. Nguyen

Graduate Theses, Dissertations, and Problem Reports (ETD)

This experiment examined the effect of nine diets supplemented with varied combinations of flaxseed oil and vitamin E on the total lipid content and fatty acid profile of rainbow trout over four months. Mean values were tested for significant differences between groups using one-way analyses of variance.;The most flaxseed oil supplementation, 23.5%, resulted in the highest concentration of total omega-3 fat in fillets (P<0.05). This was due to the increased (P<0.05) concentration of alpha-linolenic acid (ALA). The 23.5% flaxseed oil trout had the lowest (P<0.05) proportion of saturated fatty acids and the highest amount of total unsaturated fatty acid. Unexpectedly, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) contents significantly decreased as the experiment progressed. No significant differences were seen among total fat contents. The results indicate that feeding a diet supplemented with flaxseed oil and vitamin E produces omega-3 enhanced trout with higher ALA but lower EPA and DHA.

A Component-Based Approach To Verification And Validation Of Formal Software Models, Dejan Desovski Aug 2006

A Component-Based Approach To Verification And Validation Of Formal Software Models, Dejan Desovski

Graduate Theses, Dissertations, and Problem Reports (ETD)

Formal methods for verification of software systems often face the problem of state explosion and complexity. We present a divide and conquer methodology that leads to component based verification and analysis of formal requirements specifications expressed using Software Cost Reduction (SCR) models. The proposed methodology has the following steps: model partitioning, partition verification (either by model checking or testing) and composition of verification results.;We define a novel decomposition methodology for SCR specifications based on min-cut graph algorithms. The methodology identifies components in given SCR specifications and automates related abstraction methods. It also provides guidance on how to perform verification and …

The Study Of Selected Twentieth-Century American Piano Compositions Appropriate For Use With Freshman And Sophomore College Piano Majors: Performance And Pedagogical Suggestions, Li-Jung Lee

Graduate Theses, Dissertations, and Problem Reports (ETD)

Studying and performing twentieth-century compositions are important for piano students to become well-rounded musicians. This study presents a performance analysis of repertoire from three highly influential American composers: Four Piano Blues (1926-1948) by Aaron Copland, Excursions Op. 20 (1944) by Samuel Barber and Suite de Danzas Criollas Op. 15 (1946) by Alberto Ginastera. Although these works are not the most technically demanding works by these composers, the nationalistic character of the Argentine and American folk melodies and dances; the popular styles of ragtime, blues and jazz; as well as their incorporation into twentieth-century compositional technique will challenge students in their …

Journal of Extension

As the concern over global warming grows, interest in sequestering carbon in terrestrial ecosystems is expected to intensify. Nonindustrial private forest (NIPF) landowners in the southeastern United States can play a major role in sequestering atmospheric carbon. Sequestering carbon through reforestation/afforestation incentive programs requires participation by university Extension personnel to effectively communicate knowledge to landowners. This article discusses above and belowground carbon sequestration, carbon sequestration programs available to nonindustrial private forest landowners, and activities university Extension personnel may engage in to facilitate the implementation of such programs.
