Transplantation of male germ cells into sterilized recipients has been widely used in mammals for conventional breeding and transgenesis purposes. This study presents a workable approach for germ cell transplantation between male chickens. Testicular cells from adult and prepubertal donors were dispersed and transplanted by injection directly into the testes of recipient males sterilized by repeated gamma irradiation. We describe the repopulation of the recipient seminiferous epithelium up to the production of heterologous sperm in about 50% of transplanted males. In comparison to males transplanted with testicular cell preparations from adult donors, in which the first ejaculates with sperm were …

Fumonisin Production In The Maize Pathogen Fusarium Verticillioides: Genetic Basis Of Naturally Occurring Chemical Variation, Robert H. Proctor, Ronald D. Plattner, Anne E. Desjardins, Mark Busman, Robert A. E. Butchko

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Fumonisins are polyketide-derived mycotoxins produced by the maize pathogen Fusarium verticillioides. Previous analyses identified naturally occurring variants of the fungus that are deficient in fumonisin C-10 hydroxylation or that do not produce any fumonisins. In the current study, gene deletion and genetic complementation analyses localized the C-10 hydroxylation deficiency to a cytochrome P450 monooxygenase gene in the fumonisin biosynthetic gene (FUM) cluster. Sequence analysis indicated that the hydroxylation deficiency resulted from a single nucleotide insertion that caused a frame shift in the coding region of the gene. Genetic complementation localized the fumonisinnonproduction phenotype to the polyketide synthase …

Human Emanations And Related Natural Compounds That Inhibit Mosquito Host-Finding Abilities, Ulrich R. Bernier, Daniel Kline, Kenneth H. Posey

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

One mechanism by which the action of semiochemicals can be classified is based on the behavioral impact within or external to the species of interest. As such, one can classify a chemical as one of the following2,3:

1. Pheromone, if it results in response between insects of the same species
2. Kairomone, if it results in response in another species that benefits the species receiving the chemical cue
3. Allomone, if it results in response in another species that benefits the species releasing the chemical cue

However, the distinctions can be more specific by classification of chemical cues …

Laboratory Evaluation Of Avian Odors For Mosquito (Diptera: Culicidae) Attraction, S. A. Allan, Ulrich R. Bernier, D. L. Kline Jan 2006

Laboratory Evaluation Of Avian Odors For Mosquito (Diptera: Culicidae) Attraction, S. A. Allan, Ulrich R. Bernier, D. L. Kline

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Attraction of Culex quinquefasciatus Say, Culex tarsalis Coquillett, Culex nigripalpus Theobald, and Aedes aegypti (L.) to avian and other host odors was investigated in a dual-port olfactometer. Although attraction to a human arm was high for Ae. aegypti (>80%) and low for all Culex spp. (<25%), all species responded similarly to a chicken (55.3-73.6%). Responses of Ae. aegypti, Cx. quinquefasciatus, and Cx. nigripalpus to feathers were low (<20%) but greater than to controls. There was no difference in attraction of Cx. tarsalis to feathers or controls. Responses to CO2 (5 ml/min) were low for all species (<15%) except Cx. tarsalis, which were moderate (24.5%). When feathers were combined with CO2, the resulting attraction was additive or lower than responses …

Markets For Markets: Origins And Subjects Of Information Markets, Miriam A. Cherry, Robert L. Rogers Jan 2006

Markets For Markets: Origins And Subjects Of Information Markets, Miriam A. Cherry, Robert L. Rogers

All Faculty Scholarship

This Article focuses on why information markets have covered certain subject areas, sometimes of minor importance, while neglecting other subject areas of greater significance. To put it another way, why do information markets exist to predict the outcome of the papal conclave and the Michael Jackson trial, but no information markets exist to predict government policy conclusions, Supreme Court decisions, or the rulings in Delaware corporate law cases? Arguably, from either a dollar value or a social utility perspective, these areas of law and business would be more important than the outcome of, say, the Jackson trial. Why, then, do …

Interhemispheric Modulation Of Corticomotor Excitability Following I-Wave Periodicity Transcranial Magnetic Brain Stimulation (Itms), Lucy Catherine Millar Jan 2006

Interhemispheric Modulation Of Corticomotor Excitability Following I-Wave Periodicity Transcranial Magnetic Brain Stimulation (Itms), Lucy Catherine Millar

Theses : Honours

AIMS: Transcranial Magnetic Stimulation (TMS) has been recently demonstrated to have potential therapeutic benefits by promoting cortical plasticity through modulation of corticospinal excitability. We have previously shown in healthy adult subjects that paired-pulse TMS (1.5ms ISI) applied over Ml at 0.2Hz for 15min (known as iTMS), can raise corticospinal excitability for a period (~10min) that outlasts the intervention. Since interhemispheric changes in corticomotor excitability are considered to have fundamental importance in the control of voluntary movement, and recovery of motor function following unilateral damage, importance is placed on understanding the mechanisms involved. The aims of the current study were therefore …

The Effects Of Light Reduction Treatments On Mobile Epifaunain An Amphibolis Griffithii (Black) Den Hartog Seagrass Ecosystem, Helen Barwick Jan 2006

The Effects Of Light Reduction Treatments On Mobile Epifaunain An Amphibolis Griffithii (Black) Den Hartog Seagrass Ecosystem, Helen Barwick

Theses : Honours

One of the main anthropogenic disturbances to seagrass meadows in Australia is reduction in light availability, through nutrient enrichment or suspended sediments. Dredging can create suspended sediment plumes from the expulsion of particulates into the water column and in tum reduces light penetration to seagrass ecosystems. Preliminary investigations have demonstrated that light reduction for different intensities and durations results in reduced seagrass and epiphytic algae biomass. The main aim of this study was to determine the effects of different intensities and durations of light reduction on epifaunal assemblages in Amphibolis griffithii seagrass meadows in Jurien Bay, Western Australia. This was …

Changes In Demersal Wild Fish Aggregations Beneath A Sea-Cage Fish Farm After The Cessation Of Farming, Fernando Tuya, Pablo Sanchez-Jerez, Tim Dempster, Arturo Boyra, Ricardo Haroun Jan 2006

Changes In Demersal Wild Fish Aggregations Beneath A Sea-Cage Fish Farm After The Cessation Of Farming, Fernando Tuya, Pablo Sanchez-Jerez, Tim Dempster, Arturo Boyra, Ricardo Haroun

School of Natural Sciences Publications

Demersal, non-cryptic, wild fish were counted in replicate 100 m2 transects beneath a floating sea-cage fish farm and two nearby sandy locations at Gran Canaria (Canary Islands, eastern Atlantic) four times before and after the cessation of farming. Cessation of farming involved the removal of farmed fish and ceasing of the daily feeding, although farm structures (cages and moorings) remained. A “beyond-BACI” sampling design provided the framework to detect the effect of the cessation of farming, which produced qualitative and quantitative changes in the composition and structure of the fish assemblages beneath the sea-cage fish farm compared with two nearby …

The White Death: Representations Of Salt Affected Landscapes In The Wheatbelt, Grant Currall Jan 2006

The White Death: Representations Of Salt Affected Landscapes In The Wheatbelt, Grant Currall

Theses : Honours

This exegesis examines the significance of salt-affected landscapes in Western Australia's Wheatbelt, particularly sites affected due to pastoral settlement. It explores ways in which landscape imagery has been constructed and controlled by cultures to represent a particular relationship between settlement and the land. A focus on both American and Australian photographic practitioners and their understanding of landscape aesthetics will be incorporated to reveal how unconventional subject matter like dryland salinity can be re-presented to provide an alternative perspective of cultures relationship with nature in the Wheatbelt. Finally, an enquiry into the impact of national identity on landscape will also explore …

Interactive E-Assessments, Geoffrey Crisp

EDU-COM International Conference

It is useful for teachers to reflect on the future formats that will be necessary for assessment tasks. There is currently much discussion concerning the need for immersive and authentic learning environments. Teachers will need to investigate assessments that mix real and virtual environments, testing declarative knowledge and also conditional and procedural capabilities. Teachers will need to contemplate using assessments that measure approaches to problem solving and student responses in terms of efficiency, ethical considerations and the involvement of others. This image of assessment is still sometime away, but will be within reach in the short term. Are academic staff …

Improving Community Water Conservation Behaviour In Chachoengsao, Thailand, Kuakul Sathapornvajana, Lynne Cohen, Neil Drew, Julie Ann Pooley

EDU-COM International Conference

The issue of water supply throughout the world is of concern for many reasons. A major factor is water pollution by industry, agriculture and residential sectors. Less than 3% of water is fresh and potable, while the remainder is saline. It is projected that by the year 2025, two-thirds of the world‘s population will encounter moderate to severe water shortages. As a result of unsustainable development over the past decade, Thailand has faced pollution problems as well as the depletion of many natural resources. These problems have impacted the country‘s main rivers (Chaopraya River, Thachine River, and the Bangpakong River), …

The Islamic Azad University And Iran’S Social Development, Ratchaporn Simbar Jan 2006

The Islamic Azad University And Iran’S Social Development, Ratchaporn Simbar

EDU-COM International Conference

Higher education in an era of emerging organizational model, being recognized as one of the principal social affairs and a delicate endeavour, can not and should not be left with the government alone. In an attempt to lighten the government‘s responsibility to provide the nation with higher education, Islamic Azad University was established in Iran. With the growing tendencies of Iranian families in making their children enter university along with the soaring demand for higher education, the government itself could not have been able to satisfy the needs. Meanwhile, training and education can be seen as the main factors in …

Symposium Introduction: The Myth Of Moral Justice: Why Our Legal System Fails To Do What's Right, Thane Rosenbaum

Faculty Scholarship

I wrote The Myth ofMoral justice,' primarily, as a moral critique of the legal system. In examining the rituals and practices of the law under moral criteria-its obsessive focus on zero-sum contests, its dedication to cold rules and procedural technicalities over human emotion, its failure to acknowledge the spiritual pain of those who come before it, its inability to create an atmosphere where apologies, reconciliation, and the restoring of moral balance to relationships is possible, its preference for judicial economy over truth, its privileging of secrets and indifference to lies, and its failure to promote an atmosphere of mutual caring …

Modification Of Human Immunodeficiency Virus Type 1 Reverse Transcriptase To Target Cells With Elevated Cellular Dntp Concentrations, Varuni K. Jamburuthugoda, Pauline Chugh, Baek Kim Jan 2006

Modification Of Human Immunodeficiency Virus Type 1 Reverse Transcriptase To Target Cells With Elevated Cellular Dntp Concentrations, Varuni K. Jamburuthugoda, Pauline Chugh, Baek Kim

Biology

Retroviruses and DNA viruses utilize cellular dNTPs as substrates for their DNA polymerases during viral replication in infected cells. However, because of S phase-dependent dNTP biosynthesis, the availability of cellular dNTPs significantly varies among cell types (e.g. dividing versus nondividing cells and normal versus tumor cells). Here we tested whether alterations in the dNTP utilization efficiency and dNTP binding affinity of viral DNA polymerases can switch viral infection specificity to cell types with different dNTP concentrations. We employed an HIV-1 reverse transcriptase (RT) mutant (Q151N), which is catalytically active only at high dNTP concentrations because of its reduced dNTP binding …

Identification Of The Heparin-Binding Determinants Within Fibronectin Repeat Iii1: Role In Cell Spreading And Growth, Liqiong Gui, Katherine Wojciechowski, Candace D. Gildner, Hristina Nedelkovska, Denise C. Hocking Jan 2006

Identification Of The Heparin-Binding Determinants Within Fibronectin Repeat Iii1: Role In Cell Spreading And Growth, Liqiong Gui, Katherine Wojciechowski, Candace D. Gildner, Hristina Nedelkovska, Denise C. Hocking

Biology

Fibronectins are high molecular mass glycoproteins that circulate as soluble molecules in the blood, and are also found in an insoluble, multimeric form in extracellular matrices throughout the body. Soluble fibronectins are polymerized into insoluble extracellular matrix (ECM) fibrils via a cell-dependent process. Recent studies indicate that the interaction of cells with the ECM form of fibronectin promotes actin organization and cell contractility, increases cell growth and migration, and enhances the tensile strength of artificial tissue constructs; ligation of integrins alone is insufficient to trigger these responses. Evidence suggests that the effect of ECM fibronectin on cell function is mediated …

Hair has been used extensively for detection of drugs. Hair analysis has also been shown to be quicker and less invasive than other methods used for analysis of environmental pollutants in biological systems. This study examined the ability of hair to pick up explosives [2, 4, 6-trinitro toluene (TNT), hexahydro-1, 3, 3-trinitro-s-triazine (RDX), pentaerythritol tetranitrate (PETN), ethylene glycol dinitrate (EGDN), 2, 4-dinitrotoluene (2, 4-DNT), nitroglycerine (NG), triacetone triperoxide (TATP) and diacetone diperoxide (DADP)] via vapor diffusion. The results showed that the sorption of explosive to black hair is significantly higher than brown, blond and bleached hair. The hair did not …

Effects Of Endocrine-Disrupting Chemicals On Fish Reproduction And Reproductive Indicators, Lesley Judith Mills Jan 2006

Effects Of Endocrine-Disrupting Chemicals On Fish Reproduction And Reproductive Indicators, Lesley Judith Mills

Open Access Dissertations

The central hypothesis tested in this dissertation is that endocrine-disrupting chemicals (EDCs) found in the aquatic environment disrupt reproductive processes in fish. To gain a broad perspective of existing evidence, a review of the current literature was conducted. From this literature review, the lack of a dependable method or indicator to assess reproductive fitness of fish in situ was identified as an outstanding need. In laboratory experiments, reproductive endpoints (egg production, fertility and viablity) were measured in the marine fish Tautogolabrus adspersus , or cunner, implanted with estrogens. Estrone and ethynylestradiol significantly reduced egg production in treated cunner at the …
