Regulatory Intricacies That Confer Temporal Control Of Conjugation And Antibiotic Production Functions In Streptomyces: New Variations On Old Themes, Kevin Lee Schully Jan 2005

Regulatory Intricacies That Confer Temporal Control Of Conjugation And Antibiotic Production Functions In Streptomyces: New Variations On Old Themes, Kevin Lee Schully

LSU Doctoral Dissertations

The complex life style of streptomycetes requires that the appropriate program of genes be expressed at the appropriate times. For example, conjugation occurs solely within the substrate mycelium stage of development while antibiotic production occurs in the aerial mycelium. Thus, complex regulatory mechanisms involving numerous modes of control have evolved that confer temporal regulation of functions governing these expression programs. Conjugation of the Streptomyces lividans plasmid pIJ101 utilizes only seven plasmid functions in its transmission. Two of these, tra and clt, are essential while three additional functions, spdA, spdB and kilB augment the process, with KorA and KorB regulating the …

Alu Retrotransposition-Mediated Genomic Variation Within The Primate Order, Pauline Ann Callinan Jan 2005

Alu Retrotransposition-Mediated Genomic Variation Within The Primate Order, Pauline Ann Callinan

LSU Doctoral Dissertations

Retrotransposons are an active family of mobile elements within primate genomes and the Short INterspersed Element (SINE) Alu is the most abundant member. These non-autonomous elements are responsible for introducing genomic diversity on an intra- and inter- species level that is useful in studies of forensic identity, population genetics, and evolutionary biology. In a computational survey of the human sex chromosomes, 344 recently integrated Alu elements were detected and subjected to empirical testing by polymerase chain reaction to determine presence/absence polymorphism. Sixteen elements were found to be polymorphic on the X chromosome, and only one polymorphic element on the Y …

Ability Of College Students To Simulate Adhd On Objective Measure Of Attention, Randee Lee Booksh Jan 2005

Ability Of College Students To Simulate Adhd On Objective Measure Of Attention, Randee Lee Booksh

LSU Doctoral Dissertations

Complaints of ADHD symptoms in college students are not uncommon and college students frequently self-refer for assessment of ADHD. Some may seek out a diagnosis to obtain academic accommodations and/or stimulant medication. Diagnosing ADHD in college students is largely reliant on self-report measures, and to a lesser extent, objective measures of attention. The typical college student has easy access to information about ADHD, potentially facilitating efforts to simulate self-reported symptoms. The present study examined the ability of college students to effectively simulate ADHD on objective and self-report measures of attention, and examined the relationship between knowledge of ADHD and ability …

Parallel Molecular Dynamics Simulations Of Pressure-Induced Structural Transformations In Cadmium Selenide Nanocrystals, Nicholas Jabari Ouma Lee Jan 2005

Parallel Molecular Dynamics Simulations Of Pressure-Induced Structural Transformations In Cadmium Selenide Nanocrystals, Nicholas Jabari Ouma Lee

LSU Doctoral Dissertations

Parallel molecular dynamics (MD) simulations are performed to investigate pressure-induced solid-to-solid structural phase transformations in cadmium selenide (CdSe) nanorods. The effects of the size and shape of nanorods on different aspects of structural phase transformations are studied. Simulations are based on interatomic potentials validated extensively by experiments. Simulations range from 105 to 106 atoms. These simulations are enabled by highly scalable algorithms executed on massively parallel Beowulf computing architectures. Pressure-induced structural transformations are studied using a hydrostatic pressure medium simulated by atoms interacting via Lennard-Jones potential. Four single-crystal CdSe nanorods, each 44Å in diameter but varying in length, in the …

Osteoporosis And Osteopenia Management In Women: Survey, Case-Referent Study, And Interventional Exercise Trial, Rania A. Mekary Jan 2005

Osteoporosis And Osteopenia Management In Women: Survey, Case-Referent Study, And Interventional Exercise Trial, Rania A. Mekary

LSU Doctoral Dissertations

Three studies are presented; the first epidemiological study was a survey of Baton Rouge/New Orleans physicians who manage osteoporosis. When the respondents’ answers were compared to selected guidelines, they displayed poor knowledge regarding osteoporosis prevention, diagnosis, and treatment (X2=39.88; P<0.0001). Rheumatologists and endocrinologists scored globally better on osteoporosis management when compared to ObGyn (OR= 6.98) (CI=2.4; 22.8). Physicians with more years of experience were more knowledgeable of osteoporosis care (OR=1.04) (CI=1.014; 1.071). The second epidemiological study was a case referent study looking at osteoporosis treatment and patient adherence of selected women in Baton Rouge. Results showed that osteoporosis management depended partly on patient adherence. Barriers to patient’s adherence varied from physiological to psychological reasons. Physicians need to provide more explanation and motivation to the patients. Spine was the area most affected and showed more improvement with treatment than the femur (P<0.01). Bisphosphonates were the most effective treatment for the spine (P<0.05). In patients with osteoporosis, being on just calcium and exercise is not enough to counteract bone resorption. They also need to be on an anti-resorptive therapy. Low body mass index, genetics, and history of fractures were negatively correlated to bone mineral density (BMD) increase (P<0.05). The third interventional pilot study was a non-randomized controlled study done on osteopenic postmenopausal women to examine the effect of calcium supplements and core/lower back strengthening exercises on lumbar density and muscle strength. Repeated measures analysis showed that both groups increased in isometric lumbar strength with time (P=0.02). When fosamax and exercise were taken as treatments, with baseline BMD as a covariable, the one-tailed P value of the two-way ANOVA showed a numerically positive but not significant increase in the exercise group (yearly change=2.373 ± 2.625; T value=0.9; P=0.21). Also, the exercise group showed increased feelings of well-being as opposed to the control group who showed no change or worsening. One woman in the control group fractured a bone. Ultimately, osteoporosis management relies on physicians’ knowledge, involvement, and patient adherence. Calcium, exercise and anti-resorptive treatments are needed in case of osteoporosis. Calcium supplements and site-specific strengthening exercises may be enough in case of osteopenia.

Pah And Pcb Distribution In Sediment Fractions And Sorptive Phases, Trudy J. Estes Jan 2005

Pah And Pcb Distribution In Sediment Fractions And Sorptive Phases, Trudy J. Estes

LSU Doctoral Dissertations

Contaminant distribution is integral to environmental evaluations of contaminated sediment on every level. The technical and economic feasibility of treatment or beneficial use is a function of the accessibility or separability of the contaminants within the sediment. The magnitude of release to the environment during dredging and disposal is dependent upon the partitioning behavior of the contaminants. Yet, little attention has been paid to contaminant distribution in the fine sediment fractions. Further, conventional distribution coefficients provide little information regarding the relative contaminant loading of each of the sorptive phases. Bioavailability of contaminants in oil and grease, soot and organic carbon, …

Dna Supercoiling With A Twist, Edwin Kamau Jan 2005

Dna Supercoiling With A Twist, Edwin Kamau

LSU Doctoral Dissertations

The level of torsion in double-stranded DNA regulates base-pair stability and DNA conformation. It is important in initiation and regulation of specific DNA metabolic processes as well as chromatin assembly. High mobility group proteins (HMGB) are architectural proteins whose HMG DNA binding domains confer significant preference for distorted DNA, such as supercoiled DNA and 4-way junctions. HMGB proteins play a role in transiently regulating or conserving DNA torsion. Topoisomerases regulate DNA supercoiling, which has been argued to provide a coherent explanation for the main modes of transcriptional control - stringent control, growth-rate control and growth-phase control during the normal cell …

Fabrication Of Core-Shell Nanoparticles, Zhanhu Guo Jan 2005

Fabrication Of Core-Shell Nanoparticles, Zhanhu Guo

LSU Doctoral Dissertations

Metallic Cu and Au shells were fabricated around cobalt nanoparticles. A new technique to coat nanoparticles with carbon coatings and poly(methyl methacrylate) (PMMA) was developed. The copper shell formation is a self-limiting process. A thin copper shell (0.82 nm) around the cobalt nanoparticle (1.56 nm) enhanced the magnetic property by increasing the blocking temperature from 124 K to 235 K for nanoparticles with a copper shell. The formed gold shell (0.67 nm) enhanced the cobalt nanoparticle magnetic property by increasing the blocking temperature above room temperature. The magnetic moment in the Co-Cu and Co-Au core-shell nanoparticle is much higher than …

An Analysis Of Human Alu Element Lineages, Anthony Bronson Carter Jan 2005

An Analysis Of Human Alu Element Lineages, Anthony Bronson Carter

LSU Doctoral Dissertations

The Alu Ya and Yb-lineages are distinct groups of Short INterspersed Elements (SINEs) found exclusively in primates. These lineages are comprised of several distinct subfamilies, which contain elements that are evolutionarily-related because they share common diagnostic mutations. Some of these Alu subfamilies are still actively mobilizing in the human genome. We have recovered 4333 Ya and Yb-lineage elements that reside in the human genome draft sequence and focused our analyses on the 4051 autosomal Alu elements. A total of 2672 autosomal loci were subjected to polymerase chain reaction (PCR)-based assays that allow analysis of individual elements for insertion presence and …

Red Rice Competition And Control In Cultivated Rice, Christopher Todd Leon Jan 2005

Red Rice Competition And Control In Cultivated Rice, Christopher Todd Leon

LSU Doctoral Dissertations

Studies were conducted to evaluate rice competitiveness with red rice and how to utilize glufosinate- and imazethapyr-resistant rice in water-seeded rice to control red rice. In the interference study, CL 121, Cocodrie, Drew, and Jasmine were seeded to obtain 95, 190, and 290 plants/sq m. Red rice density was 0 or 20 red rice plants/sq m. Jasmine, a tall, vigorous tillering, mid-season cultivar was more competitive with red rice. With the exception of Cocodrie grown red rice free, no benefit existed from increasing the seeding rate above 190 plants/sq m. Another study examined the effect of permanent flood establishment in …

Synthesis Of Symmetric And Asymmetric Water-Soluble Porphyrin Derivatives, Caroline Owendi Ongayi Jan 2005

Synthesis Of Symmetric And Asymmetric Water-Soluble Porphyrin Derivatives, Caroline Owendi Ongayi

LSU Doctoral Dissertations

Novel symmetric and asymmetric water-soluble tetrabenzoporphyrin are synthesized in relatively good yields. These compounds are synthesized by the condensation of butanopyrrole with several aldehydes that contain masked water solubilizing groups followed by metal insertion, oxidation and demetallation and deprotection of the masked solubilizing groups. The biological evaluation (in vitro and in vivo) studies are described in addition to the photophysical evaluations of the corresponding compounds. A novel versatile route to the syntheses of novel free-bases and metallo-benzoylbiliverdin is discussed. The treatment of metal free-dodecasubstituted porphyrin with sodium nitrite and trifluoroacetic acid at room temperature in the presence of oxygen affords …

Effects Of Body Size On Goose Behavior: Lesser Snow Goose And Ross's Goose, Jon Einar Jonsson Jan 2005

Effects Of Body Size On Goose Behavior: Lesser Snow Goose And Ross's Goose, Jon Einar Jonsson

LSU Doctoral Dissertations

Body size is highly variable among geese, both at intra- and interspecific levels. Interspecific variation in several behaviors has been attributed to differences in body size in geese: incubation constancy, tendency to maintain family units, and time spent foraging. Body size has important physiological implications for birds, mostly because mass-specific metabolic rate is greater for birds of smaller mass. The Body-size Hypothesis predicts that smaller species deplete their energy reserves at relatively faster rates than do larger species. Hypotheses and conclusions concerning effects of body size on waterfowl behavior often are based on comparisons of species that confront different climates, …

Medical Image Enhancement, Alina Monica Trifas Jan 2005

Medical Image Enhancement, Alina Monica Trifas

LSU Doctoral Dissertations

Each image acquired from a medical imaging system is often part of a two-dimensional (2-D) image set whose total presents a three-dimensional (3-D) object for diagnosis. Unfortunately, sometimes these images are of poor quality. These distortions cause an inadequate object-of-interest presentation, which can result in inaccurate image analysis. Blurring is considered a serious problem. Therefore, “deblurring” an image to obtain better quality is an important issue in medical image processing. In our research, the image is initially decomposed. Contrast improvement is achieved by modifying the coefficients obtained from the decomposed image. Small coefficient values represent subtle details and are amplified …

Stm And Arpes Studies Of Epitaxial Multilayer Ag On Cu(110) And Ni(110), Weichang Zhao Jan 2005

Stm And Arpes Studies Of Epitaxial Multilayer Ag On Cu(110) And Ni(110), Weichang Zhao

LSU Doctoral Dissertations

Ag nanostructures of multilayer coverages (< 30 monolayers) epitaxially self-assembled on Cu(110) and Ni(110) have been explored by scanning tunneling microscopy (STM), angle-resolved photoelectron emission spectroscopy (ARPES), and low energy electron diffraction (LEED). We have studied varied nanostructure morphologies self-assembled depending on different deposition/annealing processes and coverages, their atomic structures, growth behaviors and mechanisms, and the electronic structures of nanowires. At nominal coverages of 1.2 ML < ¦È < ~10 ML, there are two epitaxial structures on Cu(110) and Ni(110). One is a Ag(110) multilayer film, which has a superstructure with lateral periodic units of eight and three/four substrate lattice constants along [¯110] and along [001] respectively. Another is that of Ag(110) nanowires surrounded by pseudohexagonal Ag(111) monolayer. The Ag(110) nanowires are triangular in cross section. The two side surfaces are faceted and the long axis is atomically straight along [¯110]. Typical lengths are within the range of 100 ~ 5000 Å, widths 70 ~ 300 Å, side slopes 10 ~ 30º, and heights 5 ~ 60 Å. The Ag nanowires present extraordinary anisotropy with observed aspect ratios (length:width) of up to 20:1. The Ag(110) nanowires are in-registry with the substrate along [001], but not along [¯110]. At coverages of ~ 10 ML < ¦È < ~25 ML, there also exist two different nanostructures, the nanowires and a Ag(110) atomically-flat film with some pits as deep as down to the substrate and a one-dimensional quasiperiodic superstructure along [001]. There are two basic separations of the superstructural stripes: one is three lattices wide (~11 Å) and the other is two lattices wide (~7 Å). Both of nanostructures are stable at temperature up to at least 200 ¡ãC and not inter-transformable. The growth of the nanowires is driven by the elastic strain mechanism, but the growth of the atomically-flat film is driven by electronic growth mechanism originated from the electron quantum confinement in the vertical direction of the film. The ARPES of the nanowires shows dispersion in the vertical and the [¯110] directions, but no dispersion in the [001] direction because of the limited width (~ 200 Å).

Pursuing Enlightenment In Vienna, 1781-1790, Heather Morrison Jan 2005

Pursuing Enlightenment In Vienna, 1781-1790, Heather Morrison

LSU Doctoral Dissertations

Radical transformations came about in Vienna during the 1780s, as intellectuals in the city embraced the Enlightenment and explored ways in which the movement could be spread. In 1781, Joseph II and his state reformed censorship. In an instant, the Viennese had access to the great scholarly works of seventeenth- and eighteenth-century Europe. In an instant, Vienna spawned a multitude of writers, publishing houses, reading rooms and all the accoutrements of a culture of print. The newly generated intellectual culture produced an amazing amount of pamphlets, an era termed the Broschürenflut in Austrian history. Public debate on the state, religion, …

Plasmid-Mediated High-Level Gentamicin Resistance In Bacteria For The Turtle Farm Environment In Louisiana, Maria Alejandra Diaz Jan 2005

Plasmid-Mediated High-Level Gentamicin Resistance In Bacteria For The Turtle Farm Environment In Louisiana, Maria Alejandra Diaz

LSU Doctoral Dissertations

The sale of small turtles is banned from the US market due to concerns about their excretion of Salmonella spp. To produce a safe pet for the export market, the Louisiana pet turtle industry uses 1000 μg/ml gentamicin sulfate baths to eradicate Salmonella spp. from turtle eggs. In 1999, we analyzed bacterial samples recovered from turtle farms and found that strains of Salmonella enterica subsp. arizonae and other bacteria such as Salmonella enterica subsp. enterica, Enterobacter cloacae, Citrobacter freundii, Pseudomonas aeruginosa and Stenotrophomonas maltophilia, were resistant to high concentrations of gentamicin (>2000 μg/ml) and to other aminoglycosides. The goal …

Nutrient Interactions, Plant Productivity, Soil Accretion, And Policy Implications Of Wetland Enhancements In Coastal Louisiana, Christopher Brantley Jan 2005

Nutrient Interactions, Plant Productivity, Soil Accretion, And Policy Implications Of Wetland Enhancements In Coastal Louisiana, Christopher Brantley

LSU Doctoral Dissertations

Ecosystem response, stakeholder interactions, and the policy implications to a wetland assimilation project are reported here for the City of Mandeville, St. Tammany Parish, Louisiana. Between September 1998 and October 2004, input of secondarily treated wastewater effluent was found to have a net positive effect on the downstream wetland receiving basin. The major hydrologic inputs to the system are the effluent, precipitation, and back water flooding from Lake Pontchartrain. Nutrient levels were generally low except in the immediate vicinity of the outfall and removal efficiencies of N and P ranged from 44% to 87% and 25% to 93%, respectively. On …

Civic Orientation Predictors Of Black Students: An Exploratory Study, Joe Lee Lott Jr Jan 2005

Civic Orientation Predictors Of Black Students: An Exploratory Study, Joe Lee Lott Jr

LSU Doctoral Dissertations

Institutions are becoming more cognizant of preparing students for global leadership through measures such as service-learning and community service. Colby and her colleagues (2003) suggest that institutions should guide students to believe that they are members of a shared social structure. Black students are a group whose civic participation has sharply declined between the 1970s and 1990s (Putnam, 2000) when 50 years ago they were one of the most active civic groups (Davis, 1993). This exploratory study investigated the relationships between Black students’ citizenship perspectives and independent variables such as institution type, racial identity, gender, classification, major, age, and socioeconomic …

Anisotropic Diffusion Of N-Butane And N-Decane On A Stepped Surface, Shrijay Subramaniyan, Jee-Ching Wang Jan 2005

Anisotropic Diffusion Of N-Butane And N-Decane On A Stepped Surface, Shrijay Subramaniyan, Jee-Ching Wang

Chemical and Biochemical Engineering Faculty Research & Creative Works

The diffusion of single n-butane and n-decane molecules on a model stepped surface, Pt(655), and on a corresponding flat surface, Pt(111), is investigated using molecular-dynamics simulations and anisotropic united atom model. The surface step on Pt(655) causes the alkane molecules to adsorb on the lower terrace in all-trans conformations with their long molecular axes adjacent and parallel to the step edge, and to diffuse anisotropically along the surface step via a constant wiggly motion without rotation or marked deviation from the parallel adsorption configuration. At relatively high temperatures, the alkane molecules can temporarily break away from the step edge but …

Reevaluation Of Species Concepts Of Uppermost Cretaceous Unionidae (Mollusca: Bivalvia) : Hell Creek Formation, Montana, U.S.A., Marron J. Bingle

Theses and Dissertations

Species are the basic units of life relative to taxonomy, and a practical definition of species is imperative to paleontology in order to promote reproducible, unambiguous criteria for accurately interpreting past species relationships. The need for a well-defined rubric for identifying species is evident in the freshwater mussel assemblage (Family Unionidae) in the Hell Creek Formation. The Hell Creek Formation unionid species were first described by Whitfield in 1903 and 1907. His descriptions were often based on differences of a single trait. In 1976, Russell reevaluated the original species descriptions, I but still based the classification of species on only …

(First paragraph) Growing up as a child in southern England, my early memories of snow include trudging home from school with my father, gazing at the seemingly enormous snowdrifts that smoothed the hedgerows, fields and bushes, while listening to the soft “scrunch” of the snow under my Wellington boots. In the country, snow stretching as far as I could see was not a particularly uncommon sight. The quietness of the land under a foot of snow seemed eerie. I cannot remember the first time I looked at snowflakes per se; my interests as a small child were primarily in their …

Biological Sciences Faculty Publications

Based on a continuous 20-year data base ofmonthly sampling in Chesapeake Bay and tidal regions of its major tributaries, 1454 phytoplankton taxa have been identified in these waters. They represent a diverse assemblage of species with a dominant diatom flora throughout the year, in addition to large seasonal representation by chlorophytes, cyanobacteria, cryptophytes and dinoflagellates. Included among this flora were 34 potential harmful or toxin producing species. The phytoplankton compositions associated with the seasonal successional patterns are discussed, in addition to characterizing the dominant floral relationships, with comparison to early composition records within the Bay. Several of the present day …

A Defensin-Like Gene Expressed In The Black-Legged Tick, Ixodes Scapularis (Acari: Ixodidae), Wayne L. Hynes, S. M. Ceraul, S. M. Todd, K. C. Seguin, Daniel E. Sonenshine

Biological Sciences Faculty Publications

The black-legged tick Ixodes scapularis linnaeus (Acari: Ixodidae) is an important vector of microbial pathogens. Knowledge of the tick's innate immune response, particularly defensin and other antimicrobial peptides, is important for understanding how microbes survive in this tick. A defensin gene (slnA) from I. scapularis was obtained by reverse transcription-polymerase chain reaction (RT-PCR) using mRNA extracted from tissues of female ticks. RT-PCR indicated the gene was expressed in the midgut, haemocytes, and fat-body, although no evidence of a peptide was found. Sequencing a cloned cDNA fragment revealed a 225 bp open reading frame encoding a 74 amino acid pre-prodefensin, including …

Benthic Fisheries Ecology In A Changing Environment: Unraveling Process To Achieve Prediction, Mark J. Butler Iv

Biological Sciences Faculty Publications

Marine fisheries and the ecosystems that sustain them are increasingly beset by environmental deterioration, and the problem is particularly acute in coastal zones where human Populations are increasing. In the best of circumstances, fishery managers are faced with the multiple, often conflicting, demands of resource users, politicians, and scientists when considering strategies for resource management. A further challenge is that management decisions must be made against a backdrop of a deteriorating environment and the shifting status of coastal ecosystem integrity. Traditional tools for single-species management may be inadequate in these settings. Furthermore. the necessary empirical data to appropriately parameterize models …

Injection of the hormone 20-hydroxyecdysone (20-E) into partially fed (virgin) female adults of the American dog tick, Dermacentor variabilis, while they are attached and feeding on the rabbit host, initiated the expression of the vitellogenin (Vg) gene, and Vg protein secretion and uptake by the ovary. The induction of egg production by 20-E in this bioassay was dose dependent in the range of 1-50 times the concentration normally found in a replete, vitellogenic female. Ticks examined 4d after the 50x treatment were still attached to the host, had numerous enlarged vitellin-filled (brown) oocytes in their ovaries, but had not …
