The phospholipase A(2) (PLA(2)) enzymes are activated by binding to phospholipid membranes. Although the N-terminal alpha-helix of group I/II PLA(2)s plays an important role in the productive mode membrane binding of the enzymes, its role in the structural aspects of membrane-induced activation of PLA(2)s is not well understood. In order to elucidate membrane-induced conformational changes in the N-terminal helix and in the rest of the PLA(2), we have created semisynthetic human group IB PLA(2) in which the N-terminal decapeptide is joined with the C-13-labeled fragment, as well as a chimeric protein containing the N-terminal decapeptide from human group IIA PLA(2) …

Optical imaging technique has strong potential for sensitive cancer diagnosis, particularly at the early stage of cancer development. This is a sensitive, non-invasive, non-ionizing (clinically safe) and relatively inexpensive technique. Cancer imaging with optical technique however greatly relies upon the use of sensitive and stable optical probes. Unlike the traditional organic fluorescent probes, fluorescent nanoparticle probes such as dye-doped nanoparticles and quantum dots (Qdots) are bright and photostable. Fluorescent nanoparticle probes are shown to be very effective for sensitive cancer imaging with greater success in the cellular level. Two-photon fluorescence microscopy is a powerful tool for the study of dynamic cellular processes and live-cell imaging. Many commercially available fluorescent probes have been used in multiphoton-based imaging studies despite exhibiting relatively low two-photon absorption cross-section values in the tunability range of ultrafast Ti:sapphire lasers commonly used in multiphoton microscopy imaging. Furthermore, available fluorophores may be plagued with low fluorescence quantum yield and/or photoinstability (i.e., photobleaching) on exposure to the high peak power and photon density provided by the ultrafast laser source. Discrete models are used in describing various microscopic phenomena in many branches of science, ranging from biology through chemistry to physics. Arrays of evanescently coupled, equally spaced, identical waveguides are prime examples of optical structures in which discrete dynamics can be easily observed and investigated. As a result of discretization, these structures exhibit unique diffraction properties with no analogy in continuous systems. Recently nonlinear discrete optics has attracted a growing interest, triggered by the observation of discrete solitons in AlGaAs waveguide arrays reported by Eisenberg et al. in 1998. Recent findings suggest that leptin may be regulated in response to abrupt changes in energy homeostasis. Therefore, it is conceivable that transient changes in energy balance induced by exercise may also regulate leptin synthesis and secretion. As such, we hypothesized that acute increases energy expenditure (i.e. exercise), may regulate leptin concentrations in obese individuals. Fifteen healthy obese males underwent either a single exercise session of moderate intensity (58.4 % ± 4.0 of VO2max) for 60 min (n=8), or served as controls (n=7). The exercise session elicited an energy expenditure of 567±80 Kcal. No significant changes in plasma leptin (pre 23.5± …

Enhanced Biological Phosphorous Removal (EBPR) performance was found to be adequate with reduced Return Activated Sludge (RAS) flows (50% of available RAS) to the anaerobic tank and smaller than typical anaerobic zone volume (1.08 hours hydraulic retention time or HRT). Three identical parallel biological nutrient removal (BNR) pilot plants were fed with strong, highly fermented (160 mg/L VFAs), domestic/industrial wastewater from a full scale wastewater treatment facility (WWTF). The pilot plants were operated at 100%, 50%, 40% and 25% RAS (percent of available RAS) flows to the anaerobic tank with the remaining RAS to the anoxic tank. In addition, varying …

Traditional couple counseling research focused on why people end relationships, with research only recently addressing what factors contribute to relationship satisfaction and stability. Yet, throughout this research, minimal attention has been paid to the role of play in couple counseling. The research available on play in couple counseling had varied definitions of couple play and was not based on current couple counseling theory. Since their inception, the actual use of nuclear weapons in conflict is extremely limited. There have been only two documented occurrences which were committed exclusively by the United States. By contrast, however, state posturing with nuclear weapons occurs with regularity transcending historical situations, national wealth, military power, or even the actual possession of nuclear weapons. Rationalist arguments that depict nuclear posturing as a means of deterrence appear insufficient given its tendency to unbalance perceptions of equilibrium, and the public nature in which it occurs. The present investigation modeled the expectancy memory organization and likely memory activation patterns of marijuana expectancies of children across age and marijuana use. The first phase of the study surveyed 142 children to obtain their first associate to marijuana use. From their responses, the Marijuana Expectancy Inventory for Children and Adolescents (MEICA) was developed. The second phase of the study administered the MEICA to a second sample of 392 children to model marijuana expectancy organization and probable memory activation paths of marijuana users versus never-users. Conventional methods of body fluid identification use labor-intensive, technologically diverse techniques that are performed in a series, not parallel, manner and are costly in terms of time and sample. Furthermore, for some frequently encountered body fluids, such as saliva or vaginal secretions, no confirmatory technique exists. William James is well-known for arguing that different words which describe the same practical state of affairs are, in fact, equivalent in meaning. This theory of meaning is one important strain of American pragmatism, the movement which remains America’s primary philosophical contribution to intellectual history, and it invites us to consider a unique perspective on the contemporary dialogue between science and religion. Specifically, it raises the question of the necessity of philosophical foundations when there is practical agreement.

This paper argues that when practical agreement can be reached there are certain purposes for which philosophical foundations can be strategically ignored. …

Febrile seizures occur between the age of 3 months and 5 years with a temperature of 38 degrees C or higher, and are either simple or complex. Eight gene loci have been identified to be associated with certain cases of autosomal dominant familial febrile seizures, and 12 genes have been associated with some of the familial epilepsy syndromes that can start with febrile seizures. The mutations and the protein products are known for only some of these 20 genes. The risk of recurrence of convulsions in a further febrile illness is on average 30%, and of developing epilepsy is on …

We propose a novel method to overcome significant problems of baseline drift and sensitivity degradation in amperometric biosensors based on oxidase enzyme reactions. A novel glucose microsensor with a built-in electrochemical oxygen manipulation microsystem is introduced to demonstrate three novel functionalities; one-point in situ self-calibration (zero-point), broadening of dynamic range and increase in sensitivity. The influence of electrochemically generated oxygen microenvironment on the sensor output within a fluidic structure is investigated.

Backpropagation algorithm is the most commonly used algorithm for training artificial neural networks. While being a straightforward procedure, it suffers from extensive computations, relatively slow convergence speed and possible divergence for certain conditions. The efficiency of this method as the training algorithm of a radial basis function neural network (RBFN) is compared with that of particle swarm optimization, for neural network based identification of a small power system with a static compensator. The comparison of the two methods is based on the convergence speed and robustness of each method.

Rich Mesic Forest, a Northeastern variant of the species-rich Mixed Mesophytic Forest association of eastern North America, is an Acer saccharum-dominated forest type typically associated with calcareous bedrock and nutrient-rich, mull soils. Rich Mesic Forest (RMF) is a priority for conservation in the Northeast due to its limited areal extent, high plant species richness, and numerous rare taxa, yet the community characteristics and environmental correlates of this forest type are incompletely understood. There is an urgent need for tools to unravel the complex interactions and functionalities of genes. As such, there has been much interest in reverse-engineering genetic regulatory networks from time series gene expression data. We use an artificial neural network to model the dynamics of complicated gene networks and to learn their parameters. The positive and negative regulations of genes are defined by a weight matrix, and different genes are allowed to have different decaying time constants. We demonstrate the effectiveness of the method by recreating the SOS DNA Repair network of Escherichia coli bacterium, previously discovered through experimental data.
