Seasonal trends and short-term fluctuations in abundance of Plodia interpunctella (Hubner) and Cadra cautella (Walker) infesting maize stored on two South Carolina farms were studied during three storage seasons (September 1990–June 1993). Coils of corrugated paper placed on the grain surface were used to trap mature larvae seeking pupation sites. Temperatures in the grain (18-cm-deep) and in the bin headspace were recorded hourly, and grain moisture content was measured weekly. Weekly mean numbers of moth larvae, and adults of two natural enemies, trapped in the coils were used for tracking changes in their abundance over time. The most significant findings …

A cryptic plasmid of the wall-less plant pathogenic mollicute, Spiroplasma kunkelii CR2-3X, was cloned and its sequence analyzed. The 14,615 bp plasmid, designated pSKU146, has a nucleotide content of 28 mol% G + C, and contains 18 potential protein-coding regions (open reading frames, ORFs), of which six encode proteins that exhibit similarity to virulence-associated proteins involved in cell-to-cell adhesion or conjugal DNA transfer. One ORF encodes a 96 kDa protein, SkARP1, that is highly similar to SARP1 adhesin involved in attachment of Spiroplasma citri to insect vector gut membrane. Susceptibility to carbaryl in six Mexican strains of the southern cattle tick, Boophilus microplus (Canestrini), was evaluated with the Food and Agricultural Organization larval packet test. Tick strains from the cattle fever tick quarantine zone in Texas were more susceptible to carbaryl than to coumaphos or diazinon. Compared with the susceptible reference (Gonzalez) strain, Mexican tick strains demonstrated 10.9Ð59.5-fold resistance to carbaryl. SigniÞcant cross-resistance was found between carbaryl and the organophosphate acaricides coumaphos and diazinon. Bioassay results with synergists suggested that metabolic detoxification mechanisms did not play a major role in carbaryl resistance. Resistance to carbaryl was likely conferred by …

Much of biological research conducted today is multidisciplinary, partly because of the availability of ever simpler to use molecular tools and the realization that ecological systems are complex and multitrophic. Although there is a general trend toward academic specialization and cooperation, some fields still require that scientists be aware of the “bigger picture.” We consider that the study of insect-transmitted plant pathogens is one of those fields, where insect, pathogen, and plant are inseparably linked. In weed biological control, the host range of a prospective agent is one of the most important considerations in its implementation. Aim Extrapolation of tower CO₂ fluxes will be greatly facilitated if robust relationships between flux components and remotely sensed factors are established. Long-term measurements at five Northern Great Plains locations were used to obtain relationships between CO₂ fluxes and photosynthetically active radiation (Q), other on-site factors, and Normalized Difference Vegetation Index (NDVI) from the SPOT VEGETATION data set.

Background: The Mycobacterium tuberculosis 19-kDa lipoprotein has been reported to stimulate both T and B cell responses as well as induce a number of Th1 cytokines. In order to evaluate the Mycobacterium avium subsp. paratuberculosis (M. avium subsp. paratuberculosis) 19-kDa lipoprotein as an immunomodulator in cattle with Johne's disease, the gene encoding the 19-kDa protein (MAP0261c) was analyzed.

Results: MAP0261c is conserved in mycobacteria, showing a 95% amino acid identity in M. avium subspecies avium, 84% in M. intracellulare and 76% in M. bovis and M. tuberculosis. MAP0261c was cloned, expressed, and purified as a fusion protein with the maltose-binding …

We describe here the complete genome sequence of a common clone of Mycobacterium avium subspecies paratuberculosis (Map) strain K-10, the causative agent of Johne’s disease in cattle and other ruminants. The K-10 genome is a single circular chromosome of 4,829,781 base pairs and encodes 4,350 predicted ORFs, 45 tRNAs, and one rRNA operon. In silico analysis identified >3,000 genes with homologs to the human pathogen, M. tuberculosis (Mtb), and 161 unique genomic regions that encode 39 previously unknown Map genes. Analysis of nucleotide substitution rates with Mtb homologs suggest overall strong selection for a vast majority of these shared mycobacterial …

Indeed, approximately 50% of the Earth landmass is frozen at some time during the annual cycle, with 20% of the land underlain by permafrost (Sharratt et al., 1997). Infestations of corn rootworms (Coleoptera: Chrysomelidae) create economic and environmental concerns in the Corn Belt region of the United States. To supplement the population control tactics of areawide pest management programs, we believe that a better understanding of the spatial relationships between biotic and abiotic or physical factors at the landscape scale is needed. Our research used several geographical information systems (GIS) and spatial analytical techniques to examine relationships between corn rootworm metapopulation dynamics, soil texture, and elevation. Fusarium verticillioides (teleomorph Gibberella moniliformis) is a pathogen of maize worldwide and produces fumonisins, a family of mycotoxins that have been associated with several animal diseases as well as cancer in humans. In this study, we sought to identify fungal genes that affect fumonisin production and/or the plant–fungal interaction. We generated over 87,000 expressed sequence tags from nine different cDNA libraries that correspond to 11,119 unique sequences and are estimated to represent 80% of the genomic complement of genes. A comparative analysis of the libraries showed that all 15 genes in the fumonisin gene cluster were differentially expressed. …

Taxonomy: There are three members of the genus Polerovirus (family Luteoviridae ) that induce yellowing of sugar beet: Beet mild yellowing virus (BMYV), Beet chlorosis virus (BChV) and Beet western yellows virus -USA (BWYV-USA, Fig. 1). Non-beetinfecting isolates of BWYV found particularly within Europe have now been re-named Turnip yellows virus (TuYV). Species-specific antibodies are unavailable, but the viruses can be distinguished by RT-PCR using primers specifically designed to the 5 ′ end of their respective genomes. We have developed a novel, non-intrusive fluid velocity measurement method based on photobleaching of a fluorescent dye for microfluidic devices. The residence time of thefluorescent dye in a laser beam depends on the flow velocity and approximately corresponds to the decaying time of the photobleaching of the dye in the laser beam. The residence time is inversely proportional to the flow velocity. The fluorescence intensity increases with the flow velocity due to the decrease of the residence time. A calibration curve between fluorescence intensity and known flow velocity should be obtained first. The calibration relationship is then used to calculate …

Navigating Dangerous Constitutional Straits: A Prolegomenon on the Federal Marriage Amendment and the Disenfranchisement of Sexual Minorities considers the nature of the constitutional amendment process and the Supreme Court's historical approach to incorporating new amendments into the existing constitutional text. Professors Krotoszynski and Spitko argue that, given the Supreme Court's consistent narrow reading of new amendments to preserve pre-existing constitutional rights, the Federal Marriage Amendment must be broadly written to override expressly due process and equal protection guarantees against not only sexual orientation discrimination, but also gender discrimination. In our view, however, if a proposed Federal Marriage Amendment were sufficiently …

Most discussions of white privilege emphasize the individual benefit to the holder of privilege. Yet dynamics beyond the individual combine to reinforce and reinvent white privilege. Socio-cultural factors operate in conjunction with material forces, enabling whites to self-perpetuate as a dominant racialized identity. Material forces such as the distribution of societal goods and resources, the division of labor, and immigration policies, create a world that privileges whiteness. Socio-cultural factors, including discursive practices, patterns of behavior, and the thinking patterns that language creates, further strengthen white privilege, contributing to its endurance.
