Trehalose Loading Through The Mitochondrial Permeability Transition Pore Enhances Desiccation Tolerance In Rat Liver Mitochondria, Xiang Hong Liu, Alptekin Aksan, Michael A. Menze, Steven C. Hand, Mehmet Toner

Trehalose has extensively been used to improve the desiccation tolerance of mammalian cells. To test whether trehalose improves desiccation tolerance of mammalian mitochondria, we introduced trehalose into the matrix of isolated rat liver mitochondria by reversibly permeabilizing the inner membrane using the mitochondrial permeability transition pore (MPTP). Measurement of the trehalose concentration inside mitochondria using high performance liquid chromatography showed that the sugar permeated rapidly into the matrix upon opening the MPTP. The concentration of intra-matrix trehalose reached 0.29 mmol/mg protein (∼190 mM) in 5 min. Mitochondria, with and without trehalose loaded into the matrix, were desiccated in a buffer …

Reconstruction Of Late Holocene Precipitation For Central Florida As Derived From Isotopes In Speleothems, Limaris R. Soto

Little is known about the paleo-precipitation of the Florida Peninsula. In order to better understand Floridas late Holocene climate variability (last 4,200 years), the isotopic composition was analyzed of four speleothems from two caves, in West-Central Florida. Two speleothems were collected from BRC Cave in Hernando County, and two others from Briar Cave in Marion County. This study represents the first speleothem-based paleoclimate records for Florida.

Uranium-series disequilibrium analyses were determined by using thermal ionization mass spectrometry (TIMS) to provide accurate determination of chronology of the deposition of the speleothems. Stable isotopic analyses of oxygen and carbon were performed using …

The Anchor began in 1887 and was first issued weekly in 1914. Covering national and campus news alike, Hope College’s student-run newspaper has grown over the years to encompass over two-dozen editors, reporters, and staff. For much of The Anchor's history, the latest issue was distributed across campus each Wednesday throughout the academic school year (with few exceptions). In recent years The Anchor moved to monthly print issues and a more frequently updated website. The Anchor is now published in print twice per academic semester. Occasionally, the volume and/or issue numbering is irregular.

Increases in P_i combined with decreases in myoplasmic Ca2+ are believed to cause a significant portion of the decrease in muscular force during fatigue. To investigate this further, we determined the effect of 30 mM P_ion the force-Ca²⁺ relationship of chemically skinned single muscle fibers at near-physiological temperature (30°C). Fibers isolated from rat soleus (slow) and gastrocnemius (fast) muscle were subjected to a series of solutions with an increasing free Ca²⁺ concentration in the presence and absence of 30 mM P_iat both low (15°C) and high (30°C) temperature.
