Gating of voltage-dependent K(+) channels involves movements of membrane-spanning regions that control the opening of the pore. Much less is known, however, about the contributions of large intracellular channel domains to the conformational changes that underlie gating. Here, we investigated the functional role of intracellular regions in Kv4 channels by probing relevant cysteines with thiol-specific reagents. We find that reagent application to the intracellular side of inside-out patches results in time-dependent irreversible inhibition of Kv4.1 and Kv4.3 currents. In the absence or presence of Kv4-specific auxiliary subunits, mutational and electrophysiological analyses showed that none of the 14 intracellular cysteines is …

Developmental Systems Theory: A Search For Human Nature, Erin Kenzie Jul 2005

Developmental Systems Theory: A Search For Human Nature, Erin Kenzie

Honors Theses

When trying to identify the driving forces behind the development of traits, 'nature' and 'nurture' are presented as the sole answers to a seemingly inevitable philosophical question. There must be reasons behind why an individual has a certain trait, so the rhetoric goes, and the line of reasoning has to go in one of two directions. Either factors in the individual's environment - such as parental care or life experience - are responsible, or the trait is an immutable characteristic of the individual's biological makeup. Although this dichotomy has permeated the public consciousness, it is dismissed by those who study …

The Presidency And The Meaning Of Citizenship, Malinda L. Seymore Jul 2005

The Presidency And The Meaning Of Citizenship, Malinda L. Seymore

Faculty Scholarship

This Article uses the issue of presidential qualification as a vehicle to examine the meaning of citizenship today, arguing that the Natural-Born Citizen Clause perpetuates a second-class citizenship that is inappropriate and inapposite in modern American society. Upon this premise, this Article proposes that a constitutional amendment may be necessary since the argument that the Fourteenth Amendment serves as an implicit repeal of the Natural-Born Citizen Clause has proved historically insufficient. Part II of this Article examines the origins of the constitutional requirement that the President be a "natural born Citizen" and discusses the unsuccessful attempts to amend this requirement. …

Managing Susceptibility To Hereditary Breast And Ovarian Cancer, Cynthia E. Perry Phd, Msn, Rn

Dissertations

The recent identification of Breast Cancer 1 (BRCA1) and BRCA2 genes offers an opportunity for high-risk individuals to learn whether they may be genetically predisposed to develop breast and/or ovarian cancer. The purpose of this study was to examine how unaffected women, identified as BRCA positive and variant of uncertain significance (VUS) mutation carriers, managed their susceptibility to hereditary breast and ovarian cancer (HBOC). Thirty North American women ranging in age from 22 to 60 years responded to open-ended interviews. These interviews were analyzed using constant comparative method to generate a grounded theory. Managing Susceptibility was identified as the basic …

Notch Signaling From Tumor Cells: A New Mechanism Of Angiogenesis, J-L Li, Al Harris Jul 2005

Notch Signaling From Tumor Cells: A New Mechanism Of Angiogenesis, J-L Li, Al Harris

School of Biomedical Sciences

Notch signaling is an evolutionarily conserved pathway and plays key roles in embryonic vascular development and angiogenesis. Multiple components of the Notch pathway are expressed in vasculature, and mice deficient for a variety of these components display embryonic lethality with vascular remodeling defects. Alteration of Notch signaling in various endothelial cells generates profound effects on angiogenesis in vitro. New evidence shows that Notch signaling from tumor cells is able to activate endothelial cells and trigger tumor angiogenesis in vitro and in a xenograft mouse tumor model. Selective interruption of Notch signaling within tumors may provide an antiangiogenic strategy.

Time Invariance And Liquid State Machines, Eric Goodman, Dan A. Ventura

Faculty Publications

Time invariant recognition of spatiotemporal patterns is a common task of signal processing. Liquid state machines (LSMs) are a paradigm which robustly handle this type of classification. Using an artificial dataset with target pattern lengths ranging from 0.1 to 1.0 seconds, we train an LSM to find the start of the pattern with a mean absolute error of 0.18 seconds. Also, LSMs can be trained to identify spoken digits, 1-9, with an accuracy of 97.6%, even with scaling by factors ranging from 0.5 to 1.5.

Faculty Publications

A sequence-to-structure library has been created based on the complete PDB database. The tetrapeptide was selected as a unit representing a well-defined structural motif. Seven structural forms were introduced for structure classification. The early-stage folding conformations were used as the objects for structure analysis and classification. The degree of determinability was estimated for the sequence-to-structure and structure-to-sequence relations. Probability calculus and informational entropy were applied for quantitative estimation of the mutual relation between them. The structural motifs representing different forms of loops and bends were found to favor particular sequences in structure-to-sequence analysis. © 2005 Hindawi Publishing Corporation.

Sustainable Development And Private Global Governance, Douglas A. Kysar Jun 2005

Sustainable Development And Private Global Governance, Douglas A. Kysar

Cornell Law Faculty Publications

This Article utilizes recent controversy over Coca-Cola's alleged depletion of groundwater resources in India as a vehicle for exploring competing conceptions of global environmental governance and the role of private actors within them. Initially, it uses the Coca-Cola groundwater situation to identify core substantive and procedural meanings that lurk within the otherwise ingeniously ambiguous concept of sustainable development. Through this exercise, it is shown that - when properly understood - the sustainable development paradigm stands in considerable tension with the premises of market liberalism that drive such political and economic trends as global market integration; privatization and commodification of water …

The Cell Cycle–Regulated Genes Of Schizosaccharomyces Pombe, Anna Oliva, Adan Rosebrock, Francisco Ferrezuelo, Haiying Chen, Saumyadipta Pyne, Steve Skiena, Bruce Futcher, Janet Leatherwood

Department of Molecular Genetics and Microbiology Faculty Publications

Many genes are regulated as an innate part of the eukaryotic cell cycle, and a complex transcriptional network helps enable the cyclic behavior of dividing cells. This transcriptional network has been studied in Saccharomyces cerevisiae (budding yeast) and elsewhere. To provide more perspective on these regulatory mechanisms, we have used microarrays to measure gene expression through the cell cycle of Schizosaccharomyces pombe (fission yeast). The 750 genes with the most significant oscillations were identified and analyzed. There were two broad waves of cell cycle transcription, one in early/mid G2 phase, and the other near the G2/M transition. The early/mid G2 …

Cross-Validated Bagged Learning, Mark J. Van Der Laan, Sandra E. Sinisi, Maya L. Petersen Jun 2005

Cross-Validated Bagged Learning, Mark J. Van Der Laan, Sandra E. Sinisi, Maya L. Petersen

U.C. Berkeley Division of Biostatistics Working Paper Series

Many applications aim to learn a high dimensional parameter of a data generating distribution based on a sample of independent and identically distributed observations. For example, the goal might be to estimate the conditional mean of an outcome given a list of input variables. In this prediction context, Breiman (1996a) introduced bootstrap aggregating (bagging) as a method to reduce the variance of a given estimator at little cost to bias. Bagging involves applying the estimator to multiple bootstrap samples, and averaging the result across bootstrap samples. In order to deal with the curse of dimensionality, typical practice has been to …

Phylogeny And Biogeography Of Ice Crawlers (Insecta: Grylloblattodea): Evidence From Six Molecular Loci, Karl J. Jarvis Jun 2005

Phylogeny And Biogeography Of Ice Crawlers (Insecta: Grylloblattodea): Evidence From Six Molecular Loci, Karl J. Jarvis

Theses and Dissertations

Ice crawlers (Insecta: Grylloblattodea) are rarely encountered insects that consist of five genera representing 26 species from North America and Asia. Asian grylloblattids are the most diverse, but North American ice crawlers (genus Grylloblatta) are known for their adaptation to cold conditions. Phylogenetic relationships among grylloblattid species and genera are not known. Late Pleistocene glaciations had a major effect on the current Grylloblatta distribution, because their specific habitat requirements restrict them to small geographical areas. Six genes were sampled in 37 individuals for 18S rRNA, 28S rRNA, histone 3, 12S rRNA, 16S rRNA, and cytochrome oxidase II (COII) from 27 …

Gravin Targets Pka Rii To Subcellular Sites In Cultured Cells, Xiaohong Yan Jun 2005

Gravin Targets Pka Rii To Subcellular Sites In Cultured Cells, Xiaohong Yan

Theses and Dissertations

Gravin, an A Kinase Anchoring Protein, has been shown through biochemical studies to bind to the regulatory subunit of type II PKA and to PKCα. Preliminary studies also show that gravin localizes to the cell periphery but redistributes to an intracellular vesicular compartment in response to PKC activation using PMA. Although the intracellular targeting mechanisms of some AKAPs have been explored, the targeting domains and possible targeting mechanisms of gravin remain unknown. In addition, the capacity for gravin to affect the subcellular distribution of PKA has not been investigated in vivo. To identify the domains critical in determining the subcellular …
