The present invention provides a method and apparatus adapted to facilitate the entry of a preselected molecule into the intracellular space of a cellular sample through the use of ions generated by a corona charge source. With the present method and apparatus, molecules are manipulated within cells and in the extracellular space surrounding the cells. Manipulation enhances the permeability of cell barriers to allow the subsequent introduction of molecules of interest into the interior of a cell.

The primary purpose of this study was to determine the relationship of self-reported resistance training to lipid profiles. The study consisted of 10 subjects between the ages of 19 and 35. Participants were assigned to an exercise group or control group based on self-reported resistance training and according to ACSM standards. After means were determined for the data gathered, the exercise group had lower averages of weight, percent body fat, total cholesterol, low-density lipoprotein and triglycerides, as well as, a higher average of high-density lipoproteins. This research focused on the cytotoxic properties of RRR-alpha-tocopheryl polyethylene glycol 1000 succinate (TPGS) in transformed and cancerous cell lines. We used RAW264.7 macrophage and prostate cancer (LNCaP) cell lines in this study. TPGS caused cell death and decreased cell viability in a dose and time dependent manner. Cell death was evaluated fluorimetrically by employing the nucleic acid-binding fluorophore; propidium iodide. A colorimetric 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) assay was used to evaluate cell viability. Cell death can occur through necrosis or apoptosis. This research focused on investigating and comparing the cytotoxicity and cellular uptake of RRR-alpha-tocopheryl polyethylene glycol succinate (TPGS, with that of alpha-tocopheryl succinate (α-TS). Both TPGS and α-TS are water-soluble forms of vitamin E with important clinical applications. Cytotoxicity assays with RAW 264.7 and LNCaP cells incubated overnight with TPGS or α-TS at concentrations ≥ 12.4 μM suggest that α-TS is more cytotoxic than TPGS. Macrophages were found to be more sensitive than LNCaP cells when treated with similar concentrations of α-TS. For both cell lines, most of the TPGS or α-TS taken up remained esterified after 24 hours. Our …

Sex, gender-role identity, and gender-role stress were assessed in terms of their relationship to observed gender differences in self-reported aggression. Physical and verbal aggression were explored, as well as the affective component of anger and cognitive component of hostility. The role of emotional intelligence in these relationships was also evaluated, as a possible correlate to the gender-related variables. The results indicated that both gender-role stress and gender-role identification were significantly associated with all components of aggression; however, only physical aggression was related to sex. Emotional intelligence was linked to sex and gender-role identity but not with gender-role stress. The results …

The D-alanine racemase gene (alrA) is involved in the systhesis of D-alanine, a basic component of peptidoglycan that forms the backbone of the bacterial cell wall. The present invention is also directed to methods of making live-attenuated vaccines against pathogenic mycobacteria using such alrA mutants and to the vaccines made according to such methods. A cross-sectional study was conducted to evaluate a company’s online H & S training program. The ABC Company’s online 8-hr HAZWOPER refresher course is based on current OSHA standard topics required for certification and principles flexible to fit site- or task-specific situations and conditions.

An 18-question survey was used to evaluate employee perceptions of online training compared to traditional classroom training. Perceptions were compared by demographic variables (e.g. gender, position, race, age and longevity with the ABC Company), which suggested that only age had an influence on employee response. Iron is essential to the majority of microorganisms; it is an important cofactor in many cellular processes and enzymes. However in an aerobic environment and at biological pH, iron is primarily found as insoluble oxyhydroxides and is unavailable to microorganisms. Many bacteria have the ability to produce siderophores, low molecular weight compounds that have a high affinity for Fe³⁺. Siderophores are part of a multi-component system that actively transports the iron-siderophore complex into the cytoplasm. A perennial question in the philosophy of history is whether history is a science or an art. This thesis contests that this question constitutes a false dichotomy, limiting the discussion in such a way as to exclude other possibilities of understanding the nature of the historical task.

In June 2001 we initiated a Raptor Strike Avoidance Program to monitor raptor activity and develop a biologically-based approach to reduce raptor densities at Seattle-Tacoma International Airport (SEA), King County, WA. Weekly surveys established the species, sighting frequencies, and distributions of raptors. Red-tailed Hawks (RTHAs), our most common raptor, occur as both resident and migrant/wintering hawks. We attached colored leg bands, backpack radio-transmitters and wing-tags to 8 of 12 resident RTHAs to identify individuals, locate nests and to assist in defining territories. These adult RTHAs were released onsite with the expectation that they would act as “sentinel” hawks by driving …

This contribution compares all known collisions of the Czech Air Force’s aircraft with birds, from 2000 to 2004. In the course of this period we have assayed 122 cases altogether. Unfortunately, it hasn’t been possible to obtain all needed data due to whether objective or subjective reasons. The Vancouver International Airport (YVR) is the second busiest airport in Canada. YVR is located on Sea Island in the Fraser River Estuary - a world-class wintering and staging area for hundreds of thousands of migratory birds. The Fraser Delta supports Canada’s largest wintering populations of waterfowl, shorebirds, and raptors. The large number of aircraft movements and the presence of many birds near YVR pose a wide range of considerable aviation safety hazards. Until the late 1980s when a full-time Wildlife Control Program (WCP) was initiated, YVR had the highest number of bird strikes of any Canadian commercial airport. Although …

Reporting of wildlife strikes with civil aircraft in the USA is voluntary but strongly encouraged by the Federal Aviation Administration (FAA) through Advisory Circulars and FAA publications. The National Wildlife Strike Database contained 59,196 strike reports for civil aircraft, 1990-2004. An initial analysis of independent strike data from an eastern USA airport in 1994 indicated that less than 20% of strikes were actually reported to the FAA for inclusion in the National Wildlife Strike Database. To obtain an improved estimate of the percent of strikes reported, we obtained 14 sets of wildlife strike data maintained by three airlines and three …

Bacterial histone-like DNA-binding proteins are best known for their role in compacting the genomic DNA. Of these proteins, HU is ubiquitous and highly conserved across the eubacterial kingdom. Using the HBsu (Bacillus subtilis-encoded HU homologue) as a model, we explore here the molecular basis for the ability of some HU homologues to engage a longer approx. 35 bp DNA site as opposed to the much shorter sites reported for other homologues. Using electrophoretic mobility-shift assays, we show that the DNA site size for HBsu is approx. 10-13 bp and that a specific surface salt bridge limits the DNA site size …

Fluorescent protein chromatin tagging as achieved by the lac operator/lac repressor system is useful to trace distinct chromatin domains in living eukaryotic nuclei. To interpret the data correctly, it is important to recognize influences of the tagging system on nuclear architecture of the host cells. Within an Arabidopsis line that carries lac operator/lac repressor/GFP transgenes, the transgene loci frequently associate with each other and with heterochromatic chromocenters. Accumulation of tagged fusion protein further enhances the association frequency. Independent experiments with a transgenic plant carrying another multi-copy transgene also revealed, independent of its transcriptional state, unusually high frequencies of association with …

Several primary murine and human B lymphomas and cell lines were found to constitutively express high levels of the activated form of c-jun N-terminal kinase (JNK), a member of the mitogen-activated protein (MAP) kinase family. Proliferation of murine B lymphomas CH31, CH12.Lx, BKS-2, and WEHI-231 and the human B lymphomas BJAB, RAMOS, RAJI, OCI-Ly7, and OCI-Ly10 was strongly inhibited by SP600125, an anthrapyrazolone inhibitor of JNK, in a dose-dependent manner. The lymphoma cells underwent apoptosis and arrested at the G2/M phase of cell cycle. Furthermore, JNK-specific small interfering RNA (siRNA) inhibited the growth of both murine and human B lymphomas. …

During the past four decades, several species of aphidophagous Coccinellidae became established in North America, including Coccinella septempunctata, Harmonia axyridis, Hippodamia variegata, and Propylea quatuordecimpunctata. After their establishment, unknown circumstances favoured a rapid increase in population densities and distribution of H. axyridis and C. septempunctata at localities hundreds and thousands of kilometers from their release sites. Propylea quatuordecimpunctata and Hippodamia variegata have spread more slowly after becoming established in northeastern North America. Comparative studies based upon allozyme variation in these four introduced species and in six native North American species of ladybird beetles revealed …

The community of predators within agroecosystems has the potential to restrict aphid populations, especially early in the season before exponential increases in density and prior to the arrival of specialist natural enemies. Although direct observations of predation, laboratory feeding trials and manipulative field studies have been used to estimate levels of biological control exerted by different species (or potentially negative interactions between them), it is often difficult to extrapolate results to naturally occurring interactions in the field.
