High-Temperature Fluorescent In Situ Hybridization For Detecting Escherichia Coli In Seawater Samples, Using Rrna-Targeted Oligonucleotide Probes And Flow Cytometry, Ying Zhong Tang, Karina Yew Hoong Gin, Tok Hoon Lim

OES Faculty Publications

Fluorescence in situ hybridization (FISH) is a widely used method to detect environmental microorganisms. The standard protocol is typically conducted at a temperature of 46°C and a hybridization time of 2 or 3 h, using the fluorescence signal intensity as the sole parameter to evaluate the performance of FISH. This paper reports our results for optimizing the conditions of FISH using rRNA-targeted oligonucleotide probes and flow cytometry and the application of these protocols to the detection of Escherichia coli in seawater spiked with E. coli culture. We obtained two types of optimized protocols for FISH, which showed rapid results with …

The Effect Of Growth Rate, Phosphorus Concentration, And Temperature On N-2 Fixation, Carbon Fixation, And Nitrogen Release In Continuous Cultures Of Trichodesmium Ims101, Margaret R. Mulholland, Peter W. Bernhardt

OES Faculty Publications

With the use of continuous culture systems, rates of dinitrogen (N2) and carbon (C) fixation and nitrogen (N)- and C-based doubling times were assessed in Trichodesmium IMS101 growing exponentially at steady state dilution rates of 0.10, 0.20, and 0.33 d-1 (doubling times of 10, 5, and 3 d - within the range reported for natural populations). Rates of C fixation, N2 fixation, and N release were examined in replicate culture systems with several techniques. Biomass-specific C uptake varied little with population doubling time, but N2 fixation and N release varied markedly among treatments. Total daily …

Do Viruses Affect Fecundity And Survival Of The Copepod Acartia Tonsa Dana?, Lisa A. Drake, Fred C. Dobbs Jan 2005

Do Viruses Affect Fecundity And Survival Of The Copepod Acartia Tonsa Dana?, Lisa A. Drake, Fred C. Dobbs

OES Faculty Publications

Naturally occurring viruses are extremely abundant in aquatic systems, and they infect bacteria, cyanobacteria, prokaryotic and eukaryotic phytoplankton, heterotrophic nanoflagellates, fish and mammals. Viral infections of single-celled organisms have been studied intensively in the past decade, but little is known about the effects of viruses on aquatic metazoans, other than for some economically important species. Because zooplankton assemblages are often dominated in number and biomass by copepods, we used them as model organisms to study the effects of naturally occurring viruses on higher trophic levels. We attempted to induce viral infection in laboratory-reared cultures of the estuarine copepod Acartia tonsa …

Ridding Ships' Ballast Water Of Microorganisms, Fred C. Dobbs, Andrew Rogerson Jan 2005

Ridding Ships' Ballast Water Of Microorganisms, Fred C. Dobbs, Andrew Rogerson

OES Faculty Publications

( First paragraph) A complex assemblage of microorganisms exists in nearly every aquatic system on earth. In lakes and oceans, every milliliter of water contains about 102 protists (single-celled eukaryotes), 106 bacteria, and 107–109 viruses. Therefore, billions of microorganisms inevitably enter ships’ ballast tanks during normal operations. It has been argued that microorganisms must certainly be frequent invaders of coastal ecosystems, given the high densities of bacteria and viruses in ballast water—108 and 109 organisms per liter, respectively (1)—their potentially high reproductive rates, broad tolerances to physical conditions, and ability to form resting …

Variability Of Sea Ice Cover In The Chukchi Sea (Western Arctic Ocean) During The Holocene, Anne De Vernal, Claude Hillaire-Marcel, Dennis A. Darby Jan 2005

Variability Of Sea Ice Cover In The Chukchi Sea (Western Arctic Ocean) During The Holocene, Anne De Vernal, Claude Hillaire-Marcel, Dennis A. Darby

OES Faculty Publications

Dinocysts from cores collected in the Chukchi Sea from the shelf edge to the lower slope were used to reconstruct changes in sea surface conditions and sea ice cover using modern analogue techniques. Holocene sequences have been recovered in a down-slope core (B15: 2135 m, 75°44'N, sedimentation rate of ~1cm kyr-1) and in a shelf core (P1: 201 m, 73°41'N, sedimentation rate of ~22 cm kyr-1). The shelf record spanning about 8000 years suggests high-frequency centennial oscillations of sea surface conditions and a significant reduction of the sea ice at circa 6000 and 2500 calendar (cal) …

Determination Of Trace Copper By Faas After Solid Phase Extraction And Preconcentration Onto Amberlite Xad-2 Loaded With Nitroso-R Salt, Mohammad Ali Taher, Sayed Ziae Mohammadi Mobarakeh, Ali Reza Mohadesi Jan 2005

Determination Of Trace Copper By Faas After Solid Phase Extraction And Preconcentration Onto Amberlite Xad-2 Loaded With Nitroso-R Salt, Mohammad Ali Taher, Sayed Ziae Mohammadi Mobarakeh, Ali Reza Mohadesi

Turkish Journal of Chemistry

A procedure for the separation and preconcentration of trace amounts of copper is proposed. It is based on the adsorption of copper(II) ions solution onto a column of Amberlite XAD-2 resin loaded with disodium 1-nitroso-2-naphthol-3, 6-disulfonate (nitroso-R salt). Copper is quantitatively retained on the column in the pH range 5.5-7.8 at a flow rate of 2 mL min^{-1}. The copper complex eluted with 5 mL of dimethylformamide (DMF) and copper was measured by flame atomic absorption spectrometry (FAAS) at 324.8 nm. In this case, 0.10 \mu g of copper can be concentrated in the column from 800 mL of aqueous …

Hydrographic And Biological Changes In The Taiwan Strait During The 1997-1998 El Niño Winter, S. Shang, C. Zhang, H. Hong, Q. Liu, G. T. F. Wong, C. Hu, B. Huang Jan 2005

Hydrographic And Biological Changes In The Taiwan Strait During The 1997-1998 El Niño Winter, S. Shang, C. Zhang, H. Hong, Q. Liu, G. T. F. Wong, C. Hu, B. Huang

OES Faculty Publications

During the 1997–1998 El Niño event, the average sea surface temperature (SST) in the Taiwan Strait (TWS) in the winter was ~1.4°C higher than that of the winter climatological mean. The areal ratio of the warm water (≥2°C above the regional mean) to the cold water (≥2°C below the regional mean) in the TWS increased by 25% while the area of the eutrophic water (chlorophyll a >1 mg m-3) was halved. Field observations also indicate that the mixed layer in the TWS became more nutrient-poor during this winter. These observations are consistent with a diminished advection of the …

An Advanced Specular And Diffuse Bidirectional Reflectance Distribution Function Target Model For A Synthetic Aperture Ground Penetrating Radar, Robert M. Kapfer Jan 2005

An Advanced Specular And Diffuse Bidirectional Reflectance Distribution Function Target Model For A Synthetic Aperture Ground Penetrating Radar, Robert M. Kapfer

Theses

Specific radars that are designed to radiate electromagnetic (EM) energy into the ground for the purpose of detecting and identifying underground targets are called ground penetrating radars (GPR). High resolution three-dimensional images of the underground environment can be produced using a bistatic, synthetic aperture radar (SAR) processing technique. Information pertaining to the underground scenario can be extracted from the three-dimensional images through methodical post data analysis.

Theoretical models and proof-of-concept designs are used to validate and advance deep GPR research and development efforts. The theoretical modeling of a deep GPR system is a lucrative method to obtain realistic deep GPR …

Chemistry Of Surface Waters: Distinguishing Fine-Scale Differences In Sea Grass Habitats Of Chesapeake Bay, Emmanis Dorval, Cynthia M. Jones Jan 2005

Chemistry Of Surface Waters: Distinguishing Fine-Scale Differences In Sea Grass Habitats Of Chesapeake Bay, Emmanis Dorval, Cynthia M. Jones

OES Faculty Publications

We tested the hypothesis that the physical and chemical processes acting in sea grass habitats of the lower Chesapeake Bay are spatially structured and that dissolved elemental chemistry of sea grass-habitat surface waters have their own unique identity. We sampled surface waters from July to September 2001 in five sea grass habitats of the lower bay: Potomac, Rappahannock, York, Island (Tangier-Bloodsworth), and Eastern Shore. Dissolved Mg, Mn, Sr, and Ba concentrations were measured by sector field inductively coupled plasma-mass spectrometry. As expected, Mg, Sr, and Ba exhibited conservative behavior, but Mn exhibited nonconservative behavior along the salinity gradient. Spatial differences …

Biophysical Mechanisms Of Larval Fish Ingress Into Chesapeake Bay, Jonathan A. Hare, Simon Thorrold, Harvey Walsh, Christian Reiss, Arnoldo Valle-Levinson, Cynthia Jones Jan 2005

Biophysical Mechanisms Of Larval Fish Ingress Into Chesapeake Bay, Jonathan A. Hare, Simon Thorrold, Harvey Walsh, Christian Reiss, Arnoldo Valle-Levinson, Cynthia Jones

OES Faculty Publications

Selective tidal stream transport is hypothesized as a dominant mechanism by which larvae of marine animals move through estuarine openings. For larvae moving from the shelf to estuarine habitats, selective tidal stream transport proposes that larvae are higher in the water column during flood tide and lower in the water column during ebb tide. Although a number of studies conclude that selective tidal stream transport is the mechanism responsible for larval ingress, few studies consider alternative mechanisms or consider passive explanations for tidal patterns in larval distributions. We examined the biophysical mechanisms responsible for larval ingress into Chesapeake Bay using …

Built-In-Self-Test Of Rf Front-End Circuitry, Anand Gopalan Jan 2005

Built-In-Self-Test Of Rf Front-End Circuitry, Anand Gopalan

Theses

Fuelled by the ever increasing demand for wireless products and the advent of deep submicron CMOS, RF ICs have become fairly commonplace in the semiconductor market. This has given rise to a new breed of Systems-On-Chip (SOCs) with RF front-ends tightly integrated along with digital, analog and mixed signal circuitry. However, the reliability of the integrated RF front-end continues to be a matter of significant concern and considerable research. A major challenge to the reliability of RF ICs is the fact that their performance is also severely degraded by wide tolerances in on-chip passives and package parasitics, in addition to …

Optimization Of An Acrylamides-Based Photopolymer For Reflection Holographic Recording, Raghavendra Jallapuram Jan 2005

Optimization Of An Acrylamides-Based Photopolymer For Reflection Holographic Recording, Raghavendra Jallapuram

Doctoral

Photopolymers have been the subject of special attention in the last two decades. The advantage of being self-developing makes them a practical alternative to silver halide photographic emulsions in holographic interferometry applications. This thesis is aimed at understanding diffusion properties and optimization of an acrylamide-based green sensitized photopolymer material for reflection holographic recording. The composition of the photopolymer includes a green sensitive dye (erythrosine B), acrylamide (monomer), electron donor (triethanolamine) a cross linking monomer (NN’methylenebisacrylamide) and a binder (polyvinyl alcohol). On illumination with light of the appropriate wavelength the dye absorbs a photon and enters into an excited state. The …

Testing Whiteness: No Child Or No School Left Behind?, Helen A. Moore Jan 2005

Testing Whiteness: No Child Or No School Left Behind?, Helen A. Moore

Department of Sociology: Faculty Publications

I began my study of schooling and critical pedagogy as a research assistant for a program called PRIME: Program Research in Multicultural Education. One of our challenges in the mid-1970s was to assist the Principal Investigator, Dr. Jane Mercer, in constructing educational materials for her role as an expert witness in Larry P. v. Riles. Through that project, I learned a great deal about testing issues as they relate to the whiteness of educational, political, legal and policy perspectives. Today, as a sociologist, I commit my research to an understanding of the processes and consequences of testing policy and the …

Family Economic Pressure And Adolescent Suicidal Ideation: Application Of The Family Stress Model, Kevin A. Yoder, Dan R. Hoyt Jan 2005

Family Economic Pressure And Adolescent Suicidal Ideation: Application Of The Family Stress Model, Kevin A. Yoder, Dan R. Hoyt

Department of Sociology: Faculty Publications

This study used a sample of 501 families from the Mississippi Delta region to examine the feasibility of the Family Stress Model for understanding adolescent suicidal ideation. The results indicated that family economic pressure was related to parental depressive symptoms, which, in turn, was related to parental hostile behavior and physical abuse. These parental behaviors were related to adolescent self-esteem and depressive symptoms, which, in turn, were related to adolescent suicidal ideation. Therefore, family economic pressure and parental depressive symptoms and behaviors were indirectly related to adolescent suicidal ideation. The results indicate the applicability of the Family Stress Model for …

Voltammetric, Uv Absorption And Viscometric Studies Of The Interaction Of Norepinephrine With Dna, Mehmet Aslanoğlu, Necla Öge Jan 2005

Voltammetric, Uv Absorption And Viscometric Studies Of The Interaction Of Norepinephrine With Dna, Mehmet Aslanoğlu, Necla Öge

Turkish Journal of Chemistry

Voltammetric, UV-vis spectroscopic and viscometric methods were used to investigate the interaction between norepinephrine and DNA. The results of square wave voltammetric and UV-absorption measurements have shown that norepinephrine was bound to DNA. Besides the electrostatic interaction between DNA and norepinephrine, hydrogen bonds and intercalation may also exist. Viscosity measurements also suggest that NE is bound to DNA via an electrostatic mode of interaction at lower norepinephrine concentrations. However, at higher concentrations of norpinephrine, the increase in the relative viscosity of DNA indicated that the intercalation is also involved in the binding. The binding constant and the binding site size …

Development Of Gold Porous Plate Electrode And Examination Of Its Performance For Iodine Determination, Ülkü Anik Kirgöz, Hüseyi̇n Tural

Turkish Journal of Chemistry

A novel procedure for the construction of a home-made gold porous plate electrode is described. A plate of silver and gold alloy was treated with nitric acid or alternatively by anodic oxidation to remove the silver component, leaving a porous surface having active sites for adsorption. The performance of the electrode developed was tested using iodine solution as analyte material. The parameters related to the working conditions were optimized before the examination of analytical characteristics. Under the optimal conditions, the voltammetric results show that the gold porous plate electrode offers the detection limit of 2.18 x 10^{-5} M and reproducibility …

A New Approach To The Ring Opening Reactions Of 2,7,7-Trimethyl-3,3-Dibromotricyclo[4.1.1.0^{2,4}]Octane, Zeki̇ Şenol, Ömer Zai̇m, Meti̇n Balci

Turkish Journal of Chemistry

The reaction of \alpha-pinene and dibromocarbene produced a fairly unstable product, 2,7,7-trimethyl-3,3-dibromotricyclo[4.1.1.0^{2,4}]octane, from which 3-bromo-7,7-dimethyl-2-methylenebicyclo[4,1,1]oct-3-ene and 3-bromo-2,7,7-trimethylbicyclo [4,1,1]octa-2,4-diene were obtained in chloroform at room temperature and in various other media and conditions. Two new compounds, 3-bromo-2,7,7-trimethylbicyclo [4.1.1]oct-3-en-2-ol and 2-(4-bromo-5-methylcyclohepta-3,5-dien-1-yl) propan-2-ol, were observed in aqueous acetone in addition to the previous two. The stability and formation mechanism of the formed products are discussed
