Structural Studies On A Mitochondrial Glyoxalase Ii, Gishanthi P. K. Marasinghe, Ian M. Sander, Brian Bennett, Gopal R. Periyannan, Ke-Wu Yang, Christopher A. Makaroff, Michael W. Crowder

Glyoxalase 2 is a β-lactamase fold-containing enzyme that appears to be involved with cellular chemical detoxification. Although the cytoplasmic isozyme has been characterized from several organisms, essentially nothing is known about the mitochondrial proteins. As a first step in understanding the structure and function of mitochondrial glyoxalase 2 enzymes, a mitochondrial isozyme (GLX2-5) from Arabidopsis thaliana was cloned, overexpressed, purified, and characterized using metal analyses, EPR and 1H NMR spectroscopies, and x-ray crystallography. The recombinant enzyme was shown to bind 1.04 ± 0.15 eq of iron and 1.31 ± 0.05 eq of Zn(II) and to exhibit kcat and …

Discovery And Characterization Of The Cryptic Psi Subunit Of The Pseudomonad Dna Replicase, Thale C. Jarvis, Amber A. Beaudry, James M. Bullard, Urs Ochsner, H. Garry Dallmann, Charles S. Mchenry

We previously reconstituted a minimal DNA replicase from Pseudomonas aeruginosa consisting of α and ϵ (polymerase and editing nuclease), β (processivity factor), and the essential τ, δ, and δ′ components of the clamp loader complex (Jarvis, T., Beaudry, A., Bullard, J., Janjic, N., and McHenry, C. (2005) J. Biol. Chem. 280, 7890-7900). In Escherichia coli DNA polymerase III holoenzyme, χ and Ψ are tightly associated clamp loader accessory subunits. The addition of E. coli χΨ to the minimal P. aeruginosa replicase stimulated its activity, suggesting the existence of χ and Ψ counterparts in P. aeruginosa. The P. aeruginosa χ …

The Effects Of Fluency Training On Performance, Maintenance, And Generalization Of Parenting Skills, Gertie Williams

The effects of fluency training on performance, maintenance, and generalization of parent training skills were examined within the context of a classroom and home setting. Three foster parents attended a 24-hour Parenting Tools for Positive Behavior Change (PBC) course. Participants completed timed fluency drills using flash cards to increase learning and performance of PBC tools. A non-concurrent multiple baseline design across participants was used to assess participant performance on flash card drills and PBC tools during in-class, pre-test, and post-test role plays, and in novel situations with children in the home before, during and after the course. Legalizes parents or guardians providing alcoholic beverages to children. Legalizes transport of open containers of alcoholic beverages purchases within preceding 24 hours. Repeals state prohibitions on importation of alcoholic beverages for personal consumption. Exempts such imports from state sales taxes. Repeals state authority to confiscate alcoholic beverages. In the event this initiative conflicts with existing federal regulations, requires Californian Congressional delegation to propose new federal regulations. Summary of estimate by Legislative Analyst and Director of Finance of fiscal impact on state and local governments: Unknown, but potentially minor changes in alcohol-related regulatory costs and revenues. Demographics And Motivation Of Adult Group Piano Students, Brenda Wristen

Adults are continuing to pursue a greater quantity and variety of educational opportunities. As evidenced by this study, adults typically have compelling goals from the outset of piano study and maintain clear expectations regarding in¬struction. They found the group environment helpful, underscoring findings from aforementioned studies regarding the success of choir, band, and other group musical activities for adults. More research concerning the social motivation of group music participation is warranted. Further investigation into what factors motivate adults to seek out and continue to pursue music study would help teachers better address the needs and concerns of adult learners. Likewise, …
