Mechanisms Of Resistance To Halogenated And Non-Halogenated Ahr Ligands In Chronically Contaminated Killifish Populations, Xabier Arzuaga

University of Kentucky Doctoral Dissertations

Chronically contaminated killifish from Newark Bay (NB) NJ, and New Bedford Harbor (NBH) MA, have developed resistance to halogenated aromatic hydrocarbons that bind to and activate the aryl hydrocarbon receptor (AHR). To study the mechanisms of resistance, adult killifish were exposed to halogenated and non-halogenated AHR ligands and enzymatic and toxicological endpoints were measured in adult and embryonic fish.

The chlorinated and non-chlorinated AHR ligands 3,34,4-tetrachlorobiphenyl (PCB77) and benzo-a-pyrene (B[a]P) induced cytochrome P450 1A (CYP1A) in reference site, but not in NB killifish. Expression of CYP3A (not part of the AHR gene battery) was inducible only in Flax Pond killifish. …

Analysis Of Demographic Variables And Levels Of Stress In Law Enforcement Officers, Kathleen M. Jansen Jan 2004

Analysis Of Demographic Variables And Levels Of Stress In Law Enforcement Officers, Kathleen M. Jansen

PCOM Psychology Dissertations

Employment in a law enforcement profession exposes an individual to potential stressors not experienced in most other professions. Sporadic efforts have been made to design stress management programs to assist officers in coping with the demands of their jobs. Much of the previous research on law enforcement stress has focused on examining the effectiveness of various coping skills; however, the training has focused on teaching a healthy lifestyle. This study seeks to add to the existing research by examining what elements of the job are perceived as the most stressful and what demographic variables, if any, may influence an individual's …

Determination Of Dimethylselenide And Dimethyldiselenide By Gas Chromatography–Photoionization Detection, William J. Hunter, L. David Kuykendall

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

A simple method for the determination of volatile selenium compounds employing a gas chromatograph equipped with a photoionization detector is described. The method involves the direct injection of dimethylselenide (DMS) or dimethyldiselenide (DMDS) into the gas chromatograph; no derivatization of the sample was required. The photoionization detector was capable of detecting 60 pg (0.55 pmol) of DMS and 150 pg (0.80 pmol) DMDS. Sensitivity was 10–50 times greater with DMS and 4–20 times greater with DMDS when the photoionization detector was employed than when the flame ionization detector was employed.

Detection And Characterization Of Amitraz Resistance In The Southern Cattle Tick, Boophilus Microplus (Acari: Ixodidae), Andrew Y. Li, Ronald B. Davey, Robert J. Miller, John E. George

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Amitraz, a formamidine acaricide, plays an important role in the control of the southern cattle tick, Boophilus microplus (Canestrini), and other tick species that infest cattle, dogs, and wild animals. Although resistance to amitraz in B. microplus was previously reported in several countries, the actual measurement of the level of amitraz resistance in ticks has been difficult to determine due to the lack of a proper bioassay technique. We conducted a survey, by using a newly reported technique that was a modification of the standard Food and Agriculture Organization larval packet test, to measure the levels of resistance to amitraz …

Temperature-Dependent Oxygen Limitation In Insect Eggs, H. Arthur Woods, Ryan I. Hill Jan 2004

Temperature-Dependent Oxygen Limitation In Insect Eggs, H. Arthur Woods, Ryan I. Hill

College of the Pacific Faculty Articles

Most terrestrial insect embryos support metabolism with oxygen from the environment by diffusion across the eggshell. Because metabolism is more temperature sensitive than diffusion, embryos should be relatively oxygen-limited at high temperatures. We tested whether survival, development time and metabolism of eggs of a moth, Manduca sexta, were sensitive to experimentally imposed variation in atmospheric oxygen availability (5–50 kPa; normoxia at sea level is 21 kPa) across a range of biologically realistic temperatures. Temperature–oxygen interactions were apparent in most experiments. Hypoxia affected survival more strongly at warmer temperatures. Metabolic rates, measured as rates of CO2 emission, were virtually insensitive …
