Lumpy skin disease (LSD) is an economically crippling disease of cattle with epizootic occurrence particularly after the onset of the rainy season. Biting arthropods have been implicated as vectors of lumpy skin disease virus (LSDV) due to observations that there is no transmission of LSD when cattle are confined to insect-proof houses. Nevertheless, the method by which LSDV is transmitted under field conditions is not clear and no specific vector has been conclusively pinpointed. Comparative Studies On The Adult Dispersal Behaviour Of Busseola Fusca (Fuller) (Lepidoptera:Noctuidae) And Chilo Partellus Swinhoe (Lepidoptera:Pyralidae), Jenard Patrick Mbugi

Two oil soluble dyes, Calco Red N-1700 and Sudan Blau-670 were added to the artificial diet of Chilo partellus and Busseola fusca. The two dyes effectively marked the larvae, adults, eggs and first stage F1 larvae. The dye treatments did not have any effects on mean fecundity, longevity, pupal weights and flight duration of mated female moths of the two stem borer species. Further experiments showed that, B fusca reared on a normal diet for 18 days and transferred to the Calco Red dyed could still lay marked eggs. The dye was found to be more appropriate for further marking …

Recruitment Of The Puf3 Protein To Its Mrna Target For Regulation Of Mrna Decay In Yeast, Wendy Olivas, J. Jackson, S. Houshmandi, Leban Lopez

The Puf family of RNA-binding proteins regulates mRNA translation and decay via interactions with 3' untranslated regions (3' UTRs) of target mRNAs. In yeast, Puf3p binds the 3' UTR of COX17 mRNA and promotes rapid deadenylation and decay. We have investigated the sequences required for Puf3p recruitment to this 3' UTR and have identified two separate binding sites. These sites are specific for Puf3p, as they cannot bind another Puf protein, Puf5p. Both sites use a conserved UGUANAUA sequence, whereas one site contains additional sequences that enhance binding affinity. In vivo, presence of either site partially stimulates COX17 mRNA decay, …

Developmental Regulation Of Apoptosis In Zebrafish Oocytes, 64-Cell Stage Blastomeres, And Germ-Ring Stage Embryos, Javier Francisco Negrón

The zebrafish early embryo acquires a general capability for apoptosis as a consequence of passage through the maternal-zygotic transition, the time that marks the beginning of zygotic transcription and the degradation of inherited maternal factors. We describe in detail the origin and regulation of the programmed cell death machinery found in the zebrafish early embryo. To elicit an apoptotic response from zebrafish early embryos, gastrulating germ-ring stage embryos were chronically exposed to the protein synthesis inhibitor cycloheximide. Chronic exposure of germ-ring stage embryos to cycloheximide resulted in the rapid activation of an apoptotic response by the embryos, which was evident …

Two subspecies of cricket frogs, Acris crepitans blanchardi and Acris crepitans crepitans, historically have occurred in West Virginia. The range of Acris c. blanchardi, Blanchard’s Cricket Frog, begins along the Ohio River extending into West Virginia. The Eastern Cricket Frog, Acris c. crepitans, is found in counties located in the eastern panhandle. The Eastern Cricket Frog is a species of great concern. Two sites where Acris c. crepitans historically occurred, two sites that never contained A. c. crepitans, one historical site that currently has A. c. crepitans, and two new sites that have A. c. crepitans were studied by auditory …

Divorce rates in Japan have increased since the mid-1960s, and even more rapidly since the 1990s. Divorce rates decreased throughout the period of industrialization, although modernization theory has argued that economic development brings pervasive cultural changes (including higher divorce rates). However, values regarding family are also influenced by the persistence of traditional values. Before WWII in Japan, a decreasing divorce rate was influenced by political ideology, which deliberately intended to change traditional ways of marriage and divorce. After WWII, however, this ideology diminished, and material affluence has led to an individualistic view that in turn has led to higher divorce …

Chronic wasting disease (CWD) status and PrP genotypes were determined for a group of 133 wild white-tailed deer in a 780 acre enclosure in western Nebraska, USA. Approximately half of the deer tested showed evidence of PrPd in the brainstem or lymphoid tissues. Four PRNP alleles encoding amino acid substitutions were identified, with substitutions at residues 95 (Q→H), 96 (G→S) or 116 (A→G), each with serine (S) at residue 138. In addition, a processed pseudogene with two alleles encoding five or six copies of the octapeptide repeat was identified in 26% of the deer. Both alleles encoded asparagine (N) …

To determine the transmissibility of transmissible mink encephalopathy (TME) agent to raccoons and to provide information about clinical course, lesions, and suitability of currently used diagnostic procedures for detection of transmissible spongiform encephalopathies (TSEs) in raccoons, 4 raccoon kits were inoculated intracerebrally with a brain suspension from mink experimentally infected with TME. One uninoculated raccoon kit served as a control. All 4 animals in the TME-inoculated group showed clinical signs of neurologic disorder and were euthanized between 21 and 23 weeks postinoculation (PI). Necropsy examinations revealed no gross lesions. Spongiform encephalopathy was observed by light microscopy, and the presence of …

Sections of medulla oblongata. taken at the level of the obex, palatine tonsil and medial retropharyngeal lymph node from 10,269 captive Rocky Mountain elk (Cervus elaphus nelsoni), were examined by immunohistochemical staining with monoclonal antibody for the prion protein associated with the transmissible spongiform encephalopathy of cervids, chronic wasting disease (PrPCWD). The protein was detected in 226 of them. On the basis of the anatomical location of the deposits in the brainstem of 183 elk, four distinct patterns of distribution of PrPCWD within the parasympathetic region of the dorsal motor nucleus of the vagus nerve …

Human serum paraoxonase (HuPON1) is a calcium-dependent enzyme that hydrolyzes esters, including organophosphates and lactones, and exhibits anti-atherogenic properties. A few amino acids have been shown to be essential for the enzyme’s arylesterase and organophosphatase activities. Until very recently, a three-dimensional model was not available for HuPON1, so functional roles have not been assigned to those residues. Based on sequence-structure alignment studies, we have folded the amino acid sequence of HuPON1 onto the sixfold β-propeller structure of squid diisopropylfluorophosphatase (DFPase). We tested the validity of this homology model by circular dichroism (CD) spectroscopy and site-directed mutagenesis. Consistent with predictions from …

In oviparous vertebrates, estrogen stimulates the liver to produce vitellogenin. Vitellogenin induction is a useful biomarker for exposure to endocrine disruptors, specifically environmental estrogens. The objective of this project was to design in vivo bioassays for environmental estrogens based on induction of vitellogenin in male Xenopus laevis. Frogs were exposed to ethinyl-estradiol by injection or immersion for different times and doses, than vitellogenin induction was assessed. An ELISA was developed to measure serum vitellogenin. Additionally, a vitellogenin rt-PCR assay was developed using species-specific vitellogenin primers, to detect mRNA induction. The use of non-invasive DNA-based sampling is becoming increasingly popular. However, the misidentification of individuals due to genotyping error (primarily allelic dropout) is a critical problem, especially when using individuals in the capture–mark–recapture (CMR) approach to estimate population size. We propose 2 simple and cost-effective tests, Examining Bimodality (EB) and Difference in Capture History (DCH), to determine whether a sample contains genotyping errors and the relative magnitude of the problem. These tests formalize currently used approaches for identifying genotyping errors. We evaluate the efficacy of these tests using simulated CMR data. Neurologically Plausible Distinctions In Cognition Relevant To Drug Use Etiology And Prevention, Alan W. Stacy, Susan L. Ames, Barbara J. Knowlton

This article outlines several distinctions in cognition and related topics in emotion that receive support from work in cognitive neuroscience and have important implications for prevention: implicit cognition, working memory, nonverbal memory, and neurobiological systems of habit. These distinctions have not been widely acknowledged or applied in drug use prevention research, despite their neural plausibility and the availability of methods to make this link. The authors briefly review the basis for the distinctions and indicate general implications and assessment possibilities for prevention researchers conducting large-scale field trials. Subsequently, the article outlines a connectionist framework for specific applications in prevention interventions. …

This article introduces the scope of the Special Issue. A variety of scientific disciplines are brought together to establish theoretical integration of the arenas of drug use, misuse, “abuse,” and drug misuse prevention. Transdisciplinary scientific collaboration (TDSC) is utilized as a process of integration. Introductory comments regarding the strengths and limitations of TDSC are presented. Then, the relevance of genetics to substance misuse and substance misuse prevention is presented. Next, the relevance of cognition for prevention is discussed. Specifically, neurologically plausible distinctions in cognition and implicit cognition and their relevance for prevention are discussed. At a relatively molar social-level of …
