The Orphan Drug Act of 1983 is an attempt at solving an important problem: how to induce a market-driven pharmaceutical industry to develop new therapeutics for diseases affecting relatively small numbers of persons. In the case of protein therapeutics produced by biotechnology, the FDA’s administration of the Act has repeatedly led to litigation and controversy. The principal and recurrent problem has been determining when a second applicant for the approval of a drug to treat an orphan disease (or “indication”) is seeking approval for the “same” drug as a previously approved orphan drug. Signaling Switches And Bistability Arising From Multisite Phosphorylation In Protein Kinase Cascades., Nick I Markevich, Jan B. Hoek, Boris N. Kholodenko

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

Mitogen-activated protein kinase (MAPK) cascades can operate as bistable switches residing in either of two different stable states. MAPK cascades are often embedded in positive feedback loops, which are considered to be a prerequisite for bistable behavior. Here we demonstrate that in the absence of any imposed feedback regulation, bistability and hysteresis can arise solely from a distributive kinetic mechanism of the two-site MAPK phosphorylation and dephosphorylation. Importantly, the reported kinetic properties of the kinase (MEK) and phosphatase (MKP3) of extracellular signal-regulated kinase (ERK) fulfill the essential requirements for generating a bistable switch at a single MAPK cascade level. We Assessed the Energetic and Aerodynamic Limits of Hovering Flight in the Carpenter Bee Xylocopa Varipuncta. using Normoxic, Variable-Density Mixtures of O2, N2 and He, We Were Able to Elicit Maximal Hovering Performance and Aerodynamic Failure in the Majority of Bees Sampled. Bees Were Not Isometric Regarding Thorax Mass and Wing Area, Both of Which Were Disproportionately Lower in Heavier Individuals. the Minimal Gas Density Necessary for Hovering (MGD) Increased with Body Mass and Decreased with Relative Thoracic Muscle Mass. Only the Four Bees in Our Sample with the Highest Body Mass-Specific Thorax Masses Were Able to Hover in Pure …

An important objective of science is to find global theories, those that explain/predict what happens in a wide variety of circumstances. Along the way, scientists usually encounter local theories which are either discarded or embedded in a more general theory. Statistical hypothesis tests provide two tools for this scientific method: (a) Tests for theory significance, regardless of local/global distinction; and (b) Tests for global-ness versus local-ness. The present work takes pieces of information from each method and builds some new tests, with power focused on global theories. In practice, paternity rulings are remarkably unimportant. With the exception of state welfare authorities pursuing mostly impoverished biological fathers, few paternity actions are brought, few mothers want to bring them and (even with state-sponsored pursuit) very few dollars get transferred to children as a result of them. In theory, however, paternity judgments are very and perniciously important because they keep alive the biological fatherhood ideal, an ideal that has never been reflected in law or fact and that is inconsistent with the emerging law of parental rights and responsibilities. Sensitivity Analysis Of Hill Muscle Parameters, Janet Brelin-Fornari, Paras Shah, Mohamed El-Sayed

A computational, rigid body model of a 50th percentile male head and neck utilizing 15 Hill Muscle pairs is used to study the sensitivity of Hill Muscle Model parameters. A 15g linear acceleration is applied within the transverse plane at the lowest vertebral level of the neck (T1). The resultant linear acceleration of the head is analyzed. In response is minimally affected. The peak accelerations did change, and in the case of varying muscle activation, the peak acceleration changed significantly, 36%. Each of the other parameter variations affected the peak acceleration of the head by less than 5%. Overall, …

Health Policy and Management Issue Briefs

This Policy Brief reviews judicial decisions construing state laws that permit insurers to include intoxication exclusionary clauses in their insurance policies. The cases and state laws examined in this analysis span health, life, disability, accidental death and dismemberment (AD&D), workers compensation, and unemployment insurance. Some variant of the intoxication exclusionary clause appears across all of these products, depending on the state. The widespread nature of such laws has its roots in the 1947 Uniform Accident and Sickness Policy Provision Law (UPPL), a model statute whose broader public policy purpose was to avoid the use of insurance to protect against the …

Pupfishes in the Death Valley region of California and Nevada comprise a monophyletic group of populations that became isolated in remote streams and springs over the past 20,000 years. These aquatic habitats show considerable ecological diversity, and allopatric populations have evolved differences in morphology and behavior. Here we investigated whether the divergence of pupfish populations in Death Valley might be associated with changes in arginine vasotocin (AVT). We used immunocytochemistry to compare the expression of AVT in the brain of Amargosa pupfish (Cyprinodon nevadensis) from two Death Valley populations: (1) the Amargosa River—a highly variable desert stream containing …

Questions concerning available products, effectiveness, and safety, especially for children, are being raised. Most species of mosquito are most active during the evening shortly after sundown and again before sunrise. Popular word puzzle lore has it that there are only two relatively common words having the five vowels AEIOU appearing once each and in order: ABSTEMIOUS and FACETIOUS. Over a dozen other such words and terms were logged in Language on Vacation and pre-1998 issue of Word Ways. The number of such AEIOU words jumped dramatically in February 1998, though, with the publication of SUsan Thorpe's article, "AEIOU in Order". Susan's article brought together the previously published AEIOU words, many new finds in Webster's Second Edition and the Oxford English Dictionary, plus many culled from the pages of biological references. …

Oxalate Metabolism By The Acetogenic Bacterium Moorella Thermoacetica, Steven L. Daniel, Christine Pilsl, Harold L. Drake

Whole-cell and cell-extract experiments were performed to study the mechanism of oxalate metabolism in the acetogenic bacterium Moorella thermoacetica. In short-term, whole-cell assays, oxalate consumption was low unless cell suspensions were supplemented with CO2, KNO3, or Na2S2O3. Cell extracts catalyzed the oxalate-dependent reduction of benzyl viologen. Oxalate consumption occurred concomitant to benzyl viologen reduction; when benzyl viologen was omitted, oxalate was not appreciably consumed. Based on benzyl viologen reduction, specific activities of extracts averaged 0.6 μmol oxalate oxidized min−1 mg protein−1. Extracts also catalyzed the formate-dependent reduction of NADP+; however, oxalate-dependent reduction of NADP+ was negligible. Oxalate- or formate-dependent reduction …

A Model Of Animal Selfhood: Expanding Interactionist Possibilities, Leslie Irvine

Human and Animal Bonding Collection

Interaction between people and companion animals provides the basis for a model of the self that does not depend on spoken language. Drawing on ethnographic research in an animal shelter as well as interviews and autoethnography, this article argues that interaction between people and animals contributes to human selfhood. In order for animals to contribute to selfhood in the ways that they do, they must be subjective others and not just the objects of anthropomorphic projection. Several dimensions of subjectivity appear among dogs and cats, constituting a “core” self consisting of agency, coherence, affectivity, and history. Conceptualizing selfhood in this …

Decommissioning Of Research Reactors At Brookhaven National Laboratory: Status, Future Options And Hazards, Gordon Thompson, Standing For Truth About Radiation, Inc.

Standing for Truth About Radiation, Inc. (STAR Foundation)

Three nuclear research reactors operated at the Brookhaven National Laboratory (BNL) during the period 1950-2000. The Graphite Research Reactor (BGRR) operated from 1950-1968, the High Flux Beam Reactor (HFBR) from 1965-1996, and the Medical Research Reactor (BMRR) from 1959-2000. Decommissioning of the BGRR is under way. Limited decommissioning has occurred at the HFBR, and no decommissioning has occurred at the BMRR. This report describes the reactors, reviews the planning and status of their decommissioning, summarizes available knowledge about each reactor’s radioactive inventory and the associated risks, and reviews BNL’s management of decommissioning. Conclusions and recommendations are provided.
