Eosinophils are often dominant inflammatory cells present in the lungs of asthma patients. Nonetheless, the role of these leukocytes remains poorly understood. We have created a transgenic line of mice (PHIL) that are specifically devoid of eosinophils, but otherwise have a full complement of hematopoietically derived cells. Allergen challenge of PHIL mice demonstrated that eosinophils were required for pulmonary mucus accumulation and the airway hyperresponsiveness associated with asthma. The development of an eosinophi-less mouse now permits an unambiguous assessment of a number of human diseases that have been linked to this granulocyte, including allergic diseases, parasite infections, and tumorigenesis.

Differential Regulation Of Matrix Metalloproteinase-2 And -9 Expression And Activity In Adult Rat Cardiac Fibroblasts In Response To Interleukin-1Β, Zhonglin Xie, Mahipal Singh, Krishna Singh

Matrix metalloproteinases (MMPs), a family of endoproteinases, are implicated in cardiac remodeling. Interleukin-1β (IL-1β), which is increased in the heart following myocardial infarction, increases expression and activity of MMP-2 (gelatinase A) and -9 (gelatinase B) in cardiac fibroblasts. Previously, we have shown that IL-1β activates ERK1/2, JNKs, and protein kinase C (PKC). However, signaling pathways involved in the regulation of MMP-2 and -9 expression and activity are not yet well understood. Using adult rat cardiac fibroblasts, we show that inhibition of ERK1/2 and JNKs inhibits IL-1β-stimulated increases in MMP-9, not MMP-2, expression and activity. Plant parameters are critical inputs in crop simulation models and allow a general set of algorithms to represent features of specific cultivars. A subset of plant parameters is often referred to as ‘‘genetic coefficients’’. However, these genetic coefficients are developed from phenotypic observations, usually have a weak genetic basis, and are at best ‘‘genotypic’’ coefficients because they consider the genotype from a very integrative perspective and likely include some impact of environment on the trait or characteristic described. With increased understanding of crop genomes, we believe models can be improved by incorporating genetic coefficients that accurately describe the action of …

BACKGROUND: Hemin, the oxidized form of heme, accumulates in intracranial hematomas and is a potent oxidant. Growing evidence suggests that it contributes to delayed injury to surrounding tissue, and that this process is affected by the heme oxygenase enzymes. In a prior study, heme oxygenase-2 gene deletion increased the vulnerability of cultured cortical astrocytes to hemin. The present study tested the effect of HO-2 gene deletion on protein oxidation, reactive oxygen species formation, and cell viability after mixed cortical neuron/astrocyte cultures were incubated with neurotoxic concentrations of hemin. Over 90 percent of all microprocessors are now used for real-time and embedded applications. The behavior of these applications is often constrained by the physical world. It is therefore important to devise higher-level languages and middleware that meet conventional functional requirements, as well as dependably and productively enforce real-time constraints. Real-Time Java is emerging as a safe, real-time environment. In this thesis we use it as our experimentation platform; however, our findings are easily adapted to other similar platforms. This thesis provides the following contributions to the study of safe and efficient real-time middleware. First, it identifies potential bottlenecks and …

Production of structural proteins from foot-and-mouth disease virus (FMDV) and bovine herpes virus (BHV-1) in Nicotiana benthamiana through the use of a tobacco mosaic virus-based vector (TMV-30B) has been reported previously. The development of the TMV-30B-HISc vector, a new version that adds a C-terminal histidine (His) sequence to the foreign protein expressed is described. Coding sequences from the FMDV VPl protein and the core protein, p24, from a clade C HIV-1 isolate from Zambia were cloned into the new vector and infective RNAs were generated for each construct to inoculate N. benthamiana plants.
