Characterization Of Human Udp-Glucose Dehydrogenase: Cys-276 Is Required For The Second Of Two Successive Oxidations, Brandi J. Sommer, Joseph J. Barycki, Melanie A. Simpson

UDP-glucose dehydrogenase (UGDH) catalyzes two oxidations of UDP-glucose to yield UDP-glucuronic acid. Pathological overproduction of extracellular matrix components may be linked to the availability of UDP-glucuronic acid; therefore UGDH is an intriguing therapeutic target. Specific inhibition of human UGDH requires detailed knowledge of its catalytic mechanism, which has not been characterized. In this report, we have cloned, expressed, and affinity-purified the human enzyme and determined its steady state kinetic parameters. The human enzyme is active as a hexamer with values for Km and Vmax that agree well with those reported for a bovine homolog. We used crystal coordinates …

Eugenol Stimulates Lactate Accumulation Yet Inhibits Volatile Fatty Acid Production And Eliminates Coliform Bacteria In Cattle And Swine Waste, Vincent H. Varel, D. L. Miller

Aim: To determine how eugenol affects fermentation parameters and faecal coliforms in cattle and swine waste slurries stored anaerobically. Methods and Results: Waste slurries (faeces : urine : water, 50 : 35 : 15) were blended with and without additives and aliquoted to triplicate 1-l flasks. Faecal coliforms were eliminated in cattle and swine waste slurries within 1 or 2 days with additions of eugenol at 10·05 mM (0·15%) and 16·75 mM (0·25%). At these concentrations volatile fatty acids (VFA) were reduced ca 70 and 50% in cattle and swine waste, respectively, over 6–8 weeks. Additionally, in cattle waste, eugenol …

The purpose of this study was to identify the variables that influence the frequency of personal respiratory protection (PRP) use by workers in swine confinement buildings (SCBs). The study was designed to answer four research questions: 1). What is the validity and reliability of the Personal Respiratory Protection Survey (PRPS)? 2). How frequently do workers in SCBs wear PRP? 3). How do demographics, occupational histories, and self-reported respiratory health histories influence the use of PRP in SCBs? and 4). The Influence Of Amyloid-Beta, A Major Pathological Marker In Alzheimer's Disease, On Molecular Cognitive Processes Of App+Ps1 Transgenic Mice, Chad Anthony Dickey

Alzheimer's disease (AD) is characterized by anterograde amnesia followed by a progressive decline in cognitive function. Post mortem examination of forebrain tissue from sufferers reveals the presence of extracellular amyloid-beta plaques, intracellular neurofibrillary tangles, activation of glial cells and massive neuron loss. Transgenic mice expressing mutated forms of the amyloid precursor protein (APP) and presenilin-1 (PS1) genes develop neuritic amyloid plaques, glial cell activation and memory deficits, without the formation of intracellular tangles and neurodegeneration.
The mechanisms by which these transgenic mice develop mnemonic deficiencies are unclear. Gene expression of aged memory-deficient APP+PS1 mice compared with non-transgenic littermates measured by …

Digital technology has vastly increased the ability of individuals to copy, produce and distribute information, making the behavior of individuals a far more significant factor in the enforcement of copyright and fair use laws than in the past. This research investigates the resources used by university faculty and their knowledge of fair use regarding web-based course materials. Aspects Of The Ecology Of A Distinct Population Of Xenosaurus Platyceps From Querétaro, México, Julio A. Lemos-Espinal, Geoffrey R. Smith, Royce E. Ballinger
Papers in Herpetology
Lizards in the genus Xenosaurus (Xenosauridae) show a flattened morphology, which is presumably an adaptation for their crevice-dwelling habits. Despite their general morphological similarities (Ballinger et al., 2000a), there is considerable variation among species and populations in ecological traits, and even populations of nominally the same species (e.g., X. grandis grandis and X. g. agrenon; Ballinger et al., 1995; Lemos-Espinal et al., 2003a) show variation, sometimes as much as between different species.
Populations of Xenosaurus are often geographically isolated (e.g., Pérez Ramos et al., 2000; Nieto Montes de Oca, 2001), and movement appears to be minimal (Lemos-Espinal et al., …

Each Psalm states a basis for the Psalmist's appeal of integrity, i.e., a reason why he should have some sort of standing before Yahweh. Each appeal is constructed around one word that summarizes the appeal. Psalm 17 bases its appeal on righteousness, or that which Yahweh requires to be acceptable to him. Psalm101 is a song to integrity, or blamelessness, the lack of any transgression or impurity. In Psalm 26, the Psalmist addresses Yahweh the Judge and challenges him to find a verdict of innocence, or no basis for any charges of wrongdoing against him. PV1 is an endothelial-specific integral membrane glycoprotein associated with the stomatal diaphragms of caveolae, transendothelial channels, and vesiculo-vacuolar organelles and the diaphragms of endothelial fenestrae. Multiple PV1 homodimers are found within each stomatal and fenestral diaphragm. We investigated the function of PV1 within these diaphragms and their regulation and found that treatment of endothelial cells in culture with phorbol myristate acetate (PMA) led to upregulation of PV1. This correlated with de novo formation of stomatal diaphragms of caveolae and transendothelial channels as well as fenestrae upon PMA treatment. The newly formed diaphragms could be labeled with anti-PV1 antibodies. The upregulation …

Barbituric acids have been historically classified as compounds that act on the central nervous system, and as such provide therapeutic uses as anxiolytics, sedatives, hypnotics, and anticonvulsants. Recent investigations of barbituric acid derivatives have provided scientists with information that barbituric acids may have applications in antibacterial, anti-chlamydial, anti-viral, as well as anti-cancer treatments. Additionally, recent literature accounts have indicated that barbituric acid derivatives may also act as immune modulators. The recent explorations of barbiturates and their potential anti-cancer and immune modulating properties are the subject of this work. "The Bear Went Over the Mountain" is a memoir that marks the people, events, landscape, and era that shapes a women's identity as she journeys from adolescence to adulthood. The story evolves through accretion with the use of a variety of writing strategies such as third person limited omniscient narrator, auto-fiction, mosaic, and disrupted narrative. Other conventions of Creative Non-fiction are used such as dialogue, characterization and plot. Autotopography (photographs) are used to create a motif of ancestral ghosts. They haunt the lives of these characters as they act and react to plots that began long before they were born. …

Music has always played a large role in human life. The technology behind the art has progressed and grown over time in many areas, for instance the instruments themselves, the recording equipment used in studios, and the reproduction through digital signal processing. One facet of music that has seen very little attention over time is the ability to transcribe audio files into musical notation. In this thesis, a method of multipitch analysis is used to track multiple simultaneous notes through time in an audio music file. This thesis illustrates the production of the narrative short film Neil and the Nun. The various chapters included in this written work delineate the development, preproduction, production, and post-productions stages of this thesis film. In supplement to these four larger chapters, the script, shot list, budget, and script breakdown are included. Neil and the Nun, written, directed, produced, and edited by me, articulates life through the perceptions of a child. The story centers on the volatile relationship of two fourth graders and their teacher, a possibly closeted lesbian nun. Because they are children, they have different notions than adults of …

In the modern geographic information systems, COTS software has been playing a major role. However, deploying heterogeneous GIS software has the tendency to form fragmented data sets and to cause inconsistency. To accomplish data consolidation, we must achieve interoperability between different GIS tools. In my thesis project, I developed Vector and Raster Data Adapters to implement the spatial data consolidation. I deployed ArcIMS to publish the spatial data and metadata onto Internet.
