Stat5a Promotes Adipogenesis In Nonprecursor Cells And Associates With The Glucocorticoid Receptor During Adipocyte Differentiation, Z. Elizabeth Floyd, Jacqueline M. Stephens

Faculty Publications

The differentiation of adipocytes is regulated by the activity of a variety of transcription factors, including peroxidase proliferator-activated receptor (PPAR)-γ and C/EBPα. Our current study demonstrates that ectopic expression of STAT5A, such as that of PPAR-γ and C/EBPα, promotes adipogenesis in two nonprecursor fibroblast cell lines. Using morphologic and biochemical criteria, we have demonstrated that STAT5A and the combination of STAT5A and STAT5B are sufficient to induce the expression of early and late adipogenic markers in BALB/c and NIH-3T3 cells. Yet, the ectopic expression of STAT5B alone does not induce the expression of adipocyte genes, but enhances the induction of …

Methylation-Sensitive Binding Of Transcription Factor Yy1 To An Insulator Sequence Within The Paternally Expressed Imprinted Gene, Peg3, Joomyeong Kim, Angela Kollhoff, Anne Bergmann, Lisa Stubbs Feb 2003

Methylation-Sensitive Binding Of Transcription Factor Yy1 To An Insulator Sequence Within The Paternally Expressed Imprinted Gene, Peg3, Joomyeong Kim, Angela Kollhoff, Anne Bergmann, Lisa Stubbs

Faculty Publications

The 5-ends of two paternally expressed mouse genes, Peg3 and Usp29, are jointly associated with a CpG island that exhibits allele-specific methylation. Sequence comparison of the regions derived from human, mouse and cow revealed the presence of two evolutionarily conserved sequence motifs including one that is repeated multiple times within the first intron of Peg3 in all three mammals. DNA mobility shift and chromatin immunoprecipitation (ChlP) assays clearly demonstrated that this motif is an in vivo binding site for the Gli-type transcription factor YY1. The YY1-binding site contains one CpG dinucleotide, and methylation of this CpG site abolishes the binding …

Transcriptional Initiation Under Conditions Of Anoxia-Induced Quiescence In Mitochondria From Artemia Franciscana Embryos, Brian D. Eads, Steven C. Hand Feb 2003

Transcriptional Initiation Under Conditions Of Anoxia-Induced Quiescence In Mitochondria From Artemia Franciscana Embryos, Brian D. Eads, Steven C. Hand

Faculty Publications

In response to anoxia, embryos of the brine shrimp Artemia franciscana are able coordinately to downregulate metabolism to levels low enough to permit survival for several years at room temperature. In addition to dramatic decreases in free ATP levels and heat production, intracellular pH drops from 7.8 to 6.3 overnight. Use of isolated mitochondria to study transcriptional responses to anoxia offers several advantages: (1) the localized nature of transcript initiation, processing and degradation, all of which may be followed in organello; (2) the relatively simple cis- and trans-machinery involved and (3) the ability to provide relevant physiological treatments in vitro. …

Localization And Characterization Of Phenamil-Sensitive Na+ Influx In Isolated Rainbow Trout Gill Epithelial Cells, Scott D. Reid, G. S. Hawkings, F. Galvez, G. G. Goss

Faculty Publications

Percoll density-gradient separation, combined with peanut lectin agglutinin (PNA) binding and magnetic bead separation, was used to separate dispersed fish gill cells into sub-populations. Functional characterization of each of the sub-populations was performed to determine which displayed acid-activated phenamil- and bafilomycin-sensitive Na+ uptake. Analysis of the mechanism(s) of 22Na+ influx was performed in control and acid-activated (addition of 10 mmoll-1 proprionic acid) cells using a variety of Na+ transport inhibitors (ouabain, phenamil, HOE-694 and bumetanide) and a V-type ATPase inhibitor (bafilomycin). We found that cells migrating to a 1.03-1.05 g ml-1 Percoll interface [pavement cells (PVCs)] possessed the lowest rates …

Background: Four multicenter Networks (GenNet, GENOA, HyperGEN, SAPPHIRe) form the National Heart, Lung and Blood Institute Family Blood Pressure Program (FBPP), to search for hypertension/blood pressure (BP) genes. The networks used different family designs and targeted multiple ethnic groups, using standardized protocols and definitions. Linkage genome scans were done on samples within each network (N = 6245 relatives). Methods: The evidence was synthesized using meta-analysis. Results: Combining ethnic groups, no region reached LOD >2, but several small peaks were identified, including chromosome 2p where two other recent reports find hypertension linkage. Conclusions: No regions show uniformly large effects on BP/hypertension …

Effect Of Integrating A Sportfishing Curriculum Into A Camp Program On The Knowledge, Awareness, And Attitudes Of Participating Youth, Keith Koupal, Marianne Krasny Feb 2003

Effect Of Integrating A Sportfishing Curriculum Into A Camp Program On The Knowledge, Awareness, And Attitudes Of Participating Youth, Keith Koupal, Marianne Krasny

Journal of Extension

We evaluated the effect of incorporating a sportfishing and environmental curriculum into a short-term summer camp program on participating youths' knowledge of fishing and biology/ecology, awareness of ethical behavior, and attitudes concerning fishing and saving the environment. Using a pre-/post-survey of camp participants, we determined that the program was successful in developing youth knowledge related to fishing skills and biology/ecology, but did not affect ethical behavior awareness or attitudes of participants. Although longer-term programs may be needed to affect youth attitudes, camp programs offer the opportunity to increase knowledge among a large number of youth with a minimal investment in …

Energetic Adaptations Along A Broad Latitudinal Gradient: Implications For Widely Distributed Assemblages, James E. Garvey, Dennis R. Devries, Russell A. Wright, Jeffrey G. Miner

Most community-based models in ecology assume that all individuals within a species respond similarly to environmental conditions and thereby exert identical effects as consumers or prey. Rather, individuals differ among systems, with important implications for population demographics and community interactions. For widely distributed assemblages made up of poikilotherms with high first-year mortality, species-specific differences in growth reaction norms as affected by both temperature and genotype will influence biotic interactions. For a broadly distributed fish assemblage, first-year growth does not vary with latitude for a planktivorous prey species, but declines with increasing latitude for a terminal piscivore. Size-based competitive interactions between …

Rhamnolipid Surfactant Production Affects Biofilm Architecture In Pseudomonas Aeruginosa Pao1, Mary E. Davey, Nicky C. Caiazza, George A. O'Toole

Dartmouth Scholarship

In response to certain environmental signals, bacteria will differentiate from an independent free-living mode of growth and take up an interdependent surface-attached existence. These surface-attached microbial communities are known as biofilms. In flowing systems where nutrients are available, biofilms can develop into elaborate three-dimensional structures. The development of biofilm architecture, particularly the spatial arrangement of colonies within the matrix and the open areas surrounding the colonies, is thought to be fundamental to the function of these complex communities. Here we report a new role for rhamnolipid surfactants produced by the opportunistic pathogen Pseudomonas aeruginosa in the maintenance of biofilm architecture. …

Uterine Neoplasm Resembling An Ovarian Sex Cord Tumor, Sarah Kuruvila, Dushyanti Samarasinghe, Sosamma Thomas

Saudi Medical Journal

Uterine neoplasm resembling an ovarian sex cord tumor is a rare neoplasm. Patients are generally of reproductive age, although a few may be perimenopausal or post menopausal. This neoplasm produces abnormal vaginal bleeding and uterine enlargement, suggesting leiomyoma or polyps. Based on the histology and clinical outcome, this tumor can be classified into 2 groups, namely one with focal areas resembling sex cord elements with a tendency to recur or metastasize and another with exclusive sex cord elements which runs a benign course. Immunohistochemical stains of the sex cord elements may show positively for vimentin, cytokeratin, actin and desmin in …

Genetic Variation In The Midcontinental Population Of Sandhill Cranes, Grus Canadensis, Jessica L. Petersen, Richard Bischof, Gary L. Krapu, Allen L. Szalanski

Nebraska Game and Parks Commission: Staff Research Publications

Three subspecies of sandhill crane (Grus canadensis) are recognized in the Midcontinental population, the lesser (Grus c. canadensis), Canadian (G. c. rowani), and greater (G. c. tabida). Blood samples collected on the population's primary spring staging area in Nebraska, U.S.A., were used to resolve the genetic relationship among these subspecies. Phylogenetic analysis of 27 G. canadensis, by DNA sequencing of a 675 bp region of the mtDNA, supports the subspecies designations of G. c. canadensisand G. c. tabida. G. c. rowani individuals were intermediate with each of the other two …

The Impact Of Future Orientation On The Avoidance Of Teen Pregnancy, Cheryl L. Somers, Traci A. Gizzi, Stephanie A. Johnson Feb 2003

The Impact Of Future Orientation On The Avoidance Of Teen Pregnancy, Cheryl L. Somers, Traci A. Gizzi, Stephanie A. Johnson

Michigan Journal of Counseling: Research, Theory and Practice

This study explores the relationship between future orientation and teens’ attitudes about pregnancy and avoidance behaviors. Participants were 91 adolescents, ages 15-19, surveyed using measures developed for this study that examined future orientation, attitudes to avoiding pregnancy, child-rearing responsibility, and perceptions about teen parenting. The results suggested that finishing high school was important in predicting a desire to avoid pregnancy. The perception that a teen pregnancy would make it difficult to achieve post-high school educational goals, specifically a college education, was also found to be important. Implications for school counselors and mental health professionals are discussed. Issues specific to non …

Multiple Roles For Hedgehog Signaling In Zebrafish Pituitary Development, Jennifer L. Sbrogna, Michael J.F. Barresi, Rolf O. Karlstrom

Biological Sciences: Faculty Publications

The endocrine-secreting lobe of the pituitary gland, or adenohypophysis, forms from cells at the anterior margin of the neural plate through inductive interactions involving secreted morphogens of the Hedgehog (Hh), fibroblast growth factor (FGF), and bone morphogenetic protein (BMP) families. To better understand when and where Hh signaling influences pituitary development, we have analyzed the effects of blocking Hh signaling both pharmacologically (cyclopamine treatments) and genetically (zebrafish Hh pathway mutants). While current models state that Shh signaling from the oral ectoderm patterns the pituitary after placode induction, our data suggest that Shh plays a direct early role in both pituitary …

Efficient Incorporation Of Coa, Nad And Fad Into Rna By In Vitro Transcription, Faqing Huang Feb 2003

Efficient Incorporation Of Coa, Nad And Fad Into Rna By In Vitro Transcription, Faqing Huang

Faculty Publications

Protein enzymes frequently recruit small molecule coenzymes to perform a variety of biochemical reactions. While the catalytic activities of RNA have been expanding rapidly, a similar strategy for RNA to utilize coenzymes and to increase its functional capabilities has yet to be demonstrated. A general in vitro transcription procedure has been developed to efficiently prepare RNA with coenzymes CoA, NAD and FAD covalently attached to the 5' end. These adenosine-containing coenzymes initiate transcription under the T7 class II promoter by T7 RNA polymerase. In addition to the three coenzymes, other adenosine-containing molecules may be incorporated into the first nucleotide position …
