Hierarchy Of Protein Assembly At The Vertex Ring Domain For Yeast Vacuole Docking And Fusion, Li Wang, Alexey J. Merz, Kevin M. Collins, William Wickner

Vacuole tethering, docking, and fusion proteins assemble into a “vertex ring” around the apposed membranes of tethered vacuoles before catalyzing fusion. Inhibitors of the fusion reaction selectively interrupt protein assembly into the vertex ring, establishing a causal assembly hierarchy: (a) The Rab GTPase Ypt7p mediates vacuole tethering and forms the initial vertex ring, independent of t-SNAREs or actin; (b) F-actin disassembly and GTP-bound Ypt7p direct the localization of other fusion factors; (c) The t-SNAREs Vam3p and Vam7p regulate each other’s vertex enrichment, but do not affect Ypt7p localization. The v-SNARE Vti1p is enriched at vertices by a distinct pathway that …

We examined the influence of predator odor on reproductive output of Norway rats (Rattus norvegicus). Naive laboratory rats responded to predator chemical cues with reduced litter sire and skewed sex ratio. We found that exposure to predator urine had its greatest effect on implantation and maintenance of implantation when predator urine was applied to the bedding of rats during the first third of gestation. Based on the physical appearance of corpora lutea and uterine implantation scars, we found that the reduction in litter sire was due to resolution of the embryos during the early part of gestation. Sexuality: A Queer Omission In U.S. Latino/A Theology, James B. Nickoloff
Journal of Hispanic / Latino Theology
U.S. Latino/a theologies have been prophetically strident in calling out oppression of every sort for scrutiny — all, that is, except for sexuality: particularly homosexuality, around which they has been an embarrassing and stultifying silence. Nickoloff calls for a “teología homo de conjunto,” calling attention to questions of gender and sexuality, and the crossings between them, in contemporary critical theory and Latino/a theology.

‘One Is Not Born A Latina, One Becomes One’: The Construction Of The Latina Feminist Theologian In Latino/A Theology, Michelle A. González
Journal of Hispanic / Latino Theology
The author argues that feminism and feminist theory are not taken seriously enough as critical resources in Latina/o theology. She interrogates the politics of identity as practiced &mdash overtly and covertly by Latinx theologian &mdash and how that identity discourse refracts the production of theology in the academy. She calls for greater collaboration among feminist theologians, as well as women who do not consciously use a feminist hermeneutic. Finally, she urges Latino theology to train a hermeneutics of suspicion on every essentialism, particularly figurings of "the people," as they often appear in Latinx theologies.

Evolutionary indeterminists argue that, in addition to any indeterminism introduced by quantum events, at least some evolutionary processes are themselves fundamentally indeterministic. That is, they maintain that the chance element in evolutionary processes results from indeterminism in the processes themselves, rather than simply from our cognitive limitations. Not everyone has been persuaded. A number of philosophers have argued that claims for evolutionary indeterminism are premature at best and deeply confused at worst. They maintain that evolutionary processes can and should be understood as deterministic processes. According to them, "chance" is merely a word denoting our ignorance of causes. This controversy …

Disruption of the growth hormone/insulin-like growth factor-1 (GH/IGF-1) axis has been reported and studied in menopause, hypothalamic amenorrhea, and anorexia nervosa, but not in weight-stable amenorrheic athletes. We investigated the effects of short-term transdermal estradiol on basal and exercise-stimulated serum GH, IGF-1, and associated binding proteins (IGFBP-1 and IGFBP-3) in seven weight-stable female amenorrheic athletes with percentage body fats greater that 12%. Each subject received a 72h placebo patch followed by 144h of transdermal estradiol. Serum samples for GH, IGF-1, IGFBP-1, and IGFBP-3 were obtained at baseline (t1), 72hr (0), 144hr (6), and during three 90-minute trials of aerobic exercise. …

In the presence of proliferating cell nuclear antigen, yeast DNA polymerase delta (Pol delta) replicated DNA at a rate of 40-60 nt/s. When downstream double-stranded DNA was encountered, Pol delta paused, but most replication complexes proceeded to carry out strand-displacement synthesis at a rate of 1.5 nt/s. In the presence of the flap endonuclease FEN1 (Rad27), the complex carried out nick translation (1.7 nt/s). The Dna2 nuclease/helicase alone did not efficiently promote nick translation, nor did it affect nick translation with FEN1. To address the different functions of Pol δ and FEN1 (Rad27) in Okazaki fragment maturation, exonuclease-deficient polymerase Pol δ-01 and Pol δ-5DV (corresponding to alleles pol3–01-(D321A, E323A) and pol3–5DV-(D520V), respectively) were purified and characterized in this process. In the presence of the replication clamp PCNA, both wild-type and exo− Pol δ carried out strand displacement synthesis with similar rates; however, initiation of strand displacement synthesis was much more efficient with Pol δ-exo−. When Pol δ-exo− encountered a downstream primer, it paused with 3–5 nucleotides of the primer displaced, whereas the wild type carried out precise gap filling. Consequently, in the …

Obesity is simply understood as an imbalance between energy intake and expenditure in favor of weight accretion. However, the human biological interface between food consumption and energy dissipation results in broad individual differences in eating behavior, physical activity, and efficiency of fuel storage and metabolism. In particular, the basal metabolic rate, which accounts for the greatest portion of overall energy expenditure, can vary almost twofold among individuals. Classically, three major biochemical systems are believed to contribute to basal thermogenesis: futile cycles, Na+/K+ATPase activity, and mitochondrial proton leak. Several techniques are known for non-destructive testing of aerospace structures, such as pulse echo, Eddy current, magnetic resonance, etc. Each of these techniques detects some faults but misses others, so it is desirable to combine (fuse) the results of these techniques. Several methods of data fusion are known. To improve the quality of fault detection, we modified the straightforward statistical method as follows: (1) we computed mean and variance iteratively: detected faults are excluded form the computation on the next iteration; (2) we treated the plate's edge and the inside separately; (3) we dismissed measurements in which only one technique …

Housing and Confinement of Farm Animals Collection
One important criterion in choosing appropriate housing systems for dairy cattle is that the freestall provides a comfortable surface for the cow. This paper describes two experiments testing the effects of commonly used lying surfaces on stall preference and stall usage by Holstein cows. In both experiments, 12 cows were housed individually in separate pens. Each pen contained three free stalls with a different surface: deep-bedded sawdust, deep-bedded sand, and a geotextile mattress covered with 2 to 3 cm of sawdust.
