Chromosome-Wide Control Of Meiotic Crossing Over In C. Elegans, Kenneth J. Hillers, Anne M. Villeneuve

A central event in sexual reproduction is the reduction in chromosome number that occurs at the meiosis I division. Most eukaryotes rely on crossing over between homologs, and the resulting chiasmata, to direct meiosis I chromosome segregation, yet make very few crossovers per chromosome pair [1] and [2]. This indicates that meiotic recombination must be tightly regulated to ensure that each chromosome pair enjoys the crossover necessary to ensure correct segregation. Here, we investigate control of meiotic crossing over in Caenorhabditis elegans, which averages only one crossover per chromosome pair per meiosis [3] and [4], by constructing genetic maps …

Long-term (20th century) and recent (1991–2003) trends in South Dakota’s agricultural land values are the main topics of this report. It is written for farmers and ranchers, landowners, agricultural professionals (lenders, rural appraisers, Extension educators, and agribusiness persons) and policymakers interested, for various decision- making purposes, in agricultural land market trends. Topics covered in the first section are: (1) historical trends in South Dakota farm real estate values from 1910 to 2000, and (2) the distribution of current agricultural land use by region. Cultivation Of Tropheryma Whipplei From Cerebrospinal Fluid, Matthias Maiwald, Axel Von Herbay, David N. Fredricks, Cleber C. Ouverney, Jon C. Kosek, David A. Relman

Whipple disease (WD) is a systemic disorder caused by the bacterium Tropheryma whipplei. Since the recognition of a bacterial etiology in 1961, many attempts have been made to cultivate this bacterium in vitro. It was eventually isolated, in 2000, from an infected heart valve, in coculture with human fibroblasts. Here we report the isolation of 2 new strains of T. whipplei from cerebrospinal fluid (CSF) of 2 patients with intestinal WD but no neurological signs or symptoms. One culture-positive specimen was obtained before treatment; the other was obtained 12 months after discontinuation of therapy, at a time of intestinal remission. …

A Null Mutation For Tissue Inhibitor Of Metalloproteinases-3 (Timp-3) Impairs Murine Bronchiole Branching Morphogenesis., Sean E Gill, M Cynthia Pape, Rama Khokha, Andrew J Watson, Kevin J Leco

Tissue inhibitors of metalloproteinases (TIMPs) regulate extracellular matrix (ECM) degradation by matrix metalloproteinases (MMPs). We have examined the role of TIMP-3 on ECM homeostasis and bronchiole branching morphogenesis during murine embryogenesis. Employing an in vitro organ culture system, we found decreased bronchiolar branching in null lungs when compared with wild type (WT) counterparts after 2 days in culture. When a synthetic inhibitor of MMPs at low dose was added to the culture system, branching was augmented regardless of genotype. Gelatin and in situ zymography revealed that null lungs exhibited enhanced activation of MMPs throughout lung development. We analysed the impact …

Comparative Thermal Denaturation Of Thermus Aquaticus And Escherichia Coli Type 1 Dna Polymerases, Irene Karantzeni, Carmen Ruiz, Chin Chi Liu, Vince J. Licata

Thermal denaturations of the type 1 DNA polymerases from Thermus aquaticus (Taq polymerase) and Escherichia coli (Pol 1) have been examined using differential scanning calorimetry and CD spectroscopy. The full-length proteins are single-polypeptide chains comprising a polymerase domain, a proofreading domain (inactive in Taq) and a 5′ nuclease domain. Removal of the 5′ nuclease domains produces the 'large fragment' domains of Pol 1 and Taq, termed Klenow and Klentaq respectively. Although the high temperature stability of Taq polymerase is well known, its thermal denaturation has never been directly examined previously. Habitat loss from human settlement and intensified farming has brought about a rapid loss of grassland and wetland habitats. Cultivated wild rice (Zizania palustris) paddies and associated uplands could help counteract this loss of habitat by providing nesting and feeding habitat for a variety of wildlife. Waterfowl have been the focus of many management activities designed to combat habitat loss. Although it is generally believed that the overall effects of waterfowl habitat management and enhancement are beneficial to other game and non-game birds, many direct effects have not been investigated. Papers in Virology

A tobacco mosaic virus (TMV)-based vector was utilized for expression of a cytosolic form of the bovine herpesvirus type 1 (BHV-1) protein glycoprotein D (gDc). Nicotiana benthamiana plants were harvested 7 days after inoculation with RNA transcripts derived from the TMV-gDc recombinant virus. Recombinant gDc protein of expected electrophoretic mobility accumulated in inoculated leaves to a concentration of about 20 μg/g of fresh leaf tissue. Oil-based vaccines were formulated with crude foliar extracts to immunize mice parentally. After a single injection, animals developed a sustained and specific response to both the isolated gD and native virus particles. Cattle vaccinated with …

The Fauna of Madagascar series has been around for almost 50 years. In that time authors have published on a variety of taxa that occur on this remarkable island. Throughout the history of the series there has been a heavy emphasis on beetles. The latest volume by Jean-Bernard Huchet is no exception. Huchet is an expert on Chironidae (or Chironinae depending on which scarab classification scheme you ascribe to). He is in the process of giving a much-needed taxonomic overhaul to the group.
