Intracellular transport and egress of alphaherpesviruses require the coordinate function of multiple proteins and glycoproteins. Recently, we showed that gK is expressed on infected cell surfaces and that gK cell-surface expression required the presence of the UL20 protein [J. Virol. 77 (2003), 499]. Overexpression of gK by gK-transformed cells blocked transport of enveloped virions from perinuclear spaces and inhibited virus-induced cell fusion caused by gK syncytial mutants [J. Virol. 69 (1995), 5401]. Therefore, we investigated whether altered synthesis and transport of gK was responsible for the observed gK-mediated interference phenomena. Marginal Modeling Of Multilevel Binary Data With Time-Varying Covariates, Diana Miglioretti, Patrick Heagerty

We propose and compare two approaches for regression analysis of multilevel binary data when clusters are not necessarily nested: a GEE method that relies on a working independence assumption coupled with a three-step method for obtaining empirical standard errors; and a likelihood-based method implemented using Bayesian computational techniques. Implications of time-varying endogenous covariates are addressed. The methods are illustrated using data from the Breast Cancer Surveillance Consortium to estimate mammography accuracy from a repeatedly screened population.

The predictive accuracy of a survival model can be summarized using extensions of the proportion of variation explained by the model, or R^2, commonly used for continuous response models, or using extensions of sensitivity and specificity which are commonly used for binary response models.

In this manuscript we propose new time-dependent accuracy summaries based on time-specific versions of sensitivity and specificity calculated over risk sets. We connect the accuracy summaries to a previously proposed global concordance measure which is a variant of Kendall's tau. It is common in longitudinal studies to collect information on the time until a key clinical event, such as death, and to measure markers of patient health at multiple follow-up times. One approach to the joint analysis of survival and repeated measures data adopts a time-varying covariate regression model for the event time hazard. Using this standard approach the instantaneous risk of death at time t is specified as a possibly semi-parametric function of covariate information that has accrued through time t. Semiparametric Estimation Of Time-Dependent: Roc Curves For Longitudinal Marker Data, Yingye Zheng, Patrick Heagerty

One approach to evaluating the strength of association between a longitudinal marker process and a key clinical event time is through predictive regression methods such as a time-dependent covariate hazard model. For example, a time-varying covariate Cox model specifies the instantaneous risk of the event as a function of the time-varying marker and additional covariates. In this manuscript we explore a second complementary approach which characterizes the distribution of the marker as a function of both the measurement time and the ultimate event time. Thermodynamic Characterization Of The Binding Of Activator Of G Protein Signaling 3 (Ags3) And Peptides Derived From Ags3 With G Alpha I1, Anirban Adhikari, Stephen R. Sprang

Activator of G protein signaling 3 (AGS3) is a guanine nucleotide dissociation inhibitor (GDI) that contains four G protein regulatory (GPR) or GoLoco motifs in its C-terminal domain. The entire C-terminal domain (AGS3-C) as well as certain peptides corresponding to individual GPR motifs of AGS3 bound to G alpha i1 and inhibited the binding of GTP by stabilizing the GDP-bound conformation of G alpha i1. The stoichiometry, free energy, enthalpy, and dissociation constant for binding of AGS3-C to G alpha i1 were determined using isothermal titration calorimetry. There have been few studies that have attempted to understand the world of parents raising a child with depression. This study introduces the magnitude of the problem of childhood depression and explores what it means to be the mother of an adolescent with depression. For this study, three mothers of adolescents wit.h depression were questioned interviews were in unstructured interviews. This thesis focuses on the impact of a mega-store on the existing businesses in a specific area--Magazine Street in New Orleans. The potential mega-store is a 200,000 square foot Wal-Mart Supercenter that is part of the retail component of the St. Thomas housing re-development. The focus of this project is the retail competition that might be generated by this mega-store. Businesses were studied and placed into categories as either direct or indirect competitors to the mega-store. Biofiltration For Control Of H2s From Wastewater Treatment Plant Gases, Vivian Bermudez

A low-cost and efficient methodology was used to test the performance of a biofilter removing gaseous hydrogen sulfide generated in the headworks and a primary clarifier of a local Wastewater Treatment Plant. The contaminated gas stream is distributed upward through 1,718 m3 of filter material. With a flow rate varying between 3,503.0 m3/h and 4,587.3 m3/h and hydrogen sulfide inlet concentrations between 0.8 and 146 ppm, hydrogen sulfide was efficiently eliminated by the wood bark biofilter. The removal efficiencies ranged from 97.5% to 99.9%. The mean water content of the filter material was determined to be 67.1%. The excess water …

While people all over the world are vulnerable to natural disasters, the available data clearly demonstrate a great deal of cross-country variance in the impact of catastrophic events. For example, while Hurricane Mitch took an estimated 13,000 lives when it struck Honduras and Nicaragua, the stronger Hurricane Andrew took only 26 lives when it impacted the United States. What factors explain this difference? Thus far, disaster researchers have emphasized economic and social vulnerability as determinants of disaster impact; the conventional wisdom accepts that poor and underdeveloped countries are more vulnerable than wealthy, developed countries. This study examined factors that may protect African-American young adults in college from early onset of cigarette smoking, and factors that may place them at risk for cigarette smoking, using the framework of the Social Cognitive Theory. It was hypothesized that existing regulation of smoking, positive attitude toward academics and school connectedness would be related to low cigarette-smoking rates, while exposure to pro-tobacco promotions would be related to high cigarette smoking rates. A convenience sample of 576 undergraduates at Dillard University of New Orleans completed a Harvard College Alcohol Study survey. Forty six percent (n=269) completed the survey. Ninetyone percent …

In an effect to develop for more selective neuronal nicotinic acetylcholine receptor analgesics that have less toxicity and adverse side effects relative to epibatidine, three new classes of epibatidine analogues were synthesized and evaluated in vitro as potential potent selective nAChR ligands. Specifically, three analogues of epibatidine were synthesized to explore the structure-activity relationships of epibatidine relative to neuromuscular blocking activity as well as nAChRs. Both quaternary epibatidine analogues 2 and bis-epibatidine derivative 3 exhibited high binding affinity relative to nicotine. In addition, a new series of 2-(hydroxyalkylpyridyl)-7-azabicyclo[2.2.1]heptane derivatives were synthesized and evaluated as potential ligands for nicotinic acetylcholine receptors. …
