Characterization Of Prokaryotic Diversity In A Chlorinated Solvent-Contaminated Bedrock Aquifer Using Denaturing Gradient Gel Electrophoresis, Ribosomal Dna Sequencing, And Real Time Pcr, Walid Naser

Doctoral Dissertations

A culture-independent molecular phylogenetic approach was implemented to investigate spatial and temporal variations in microbial diversity and abundance in a chlorinated solvent-contaminated bedrock aquifer. Core and groundwater samples were collected from several boreholes and included the following samples: (1) groundwater that was associated with the core, (2) open fractures and (3) partially mineralized sealed fractures (PMSF) exposed by striking the core with a surface-sterilized geology hammer. DNA representing the attached and unattached microbial communities was extracted from these samples, and the 16S rRNA gene was amplified with primers specific for Bacteria, Archaea, sulfate reducers, Geobacteraceae family, Dehalorespirers (Dehalococcoides sp. and …

Factors Affecting Plant Community Composition And Dynamics In The Ossipee Pine Barrens, New Hampshire, Lauren Fredrick Howard Jan 2003

Factors Affecting Plant Community Composition And Dynamics In The Ossipee Pine Barrens, New Hampshire, Lauren Fredrick Howard

Doctoral Dissertations

Forty-one 0.25 ha sites were sampled in the Ossipee Pine Barrens to identify and describe tree community types and investigate factors controlling forest composition and dynamics. Every site had three site-time assemblages (STA's) representing past, present, and future trees. Past (1952) vegetation was calculated based on reverse growth estimates of current stems and stumps. Future (2052) vegetation was predicted by current sapling (<10 cm dbh and ≥1 m tall) relative densities.

Cluster analysis produced three community types from 121 STA's: pitch pine, mixed pine-hardwoods, and red maple. Pitch pine communities comprised 63% of sites in 1952, but declined since. Mixed pine-hardwoods peaked at 58% in 2002, but were …

Developing Methods For The Rapid Molecular Assessment Of Aquatic Microbial Communities, Traci D. Hudson Jan 2003

Developing Methods For The Rapid Molecular Assessment Of Aquatic Microbial Communities, Traci D. Hudson

Theses, Dissertations and Capstones

Molecular investigations of microbial community structure and dynamics involve costly and time-consuming methods. This approach is limiting when rapid assessment and detection of microbial organisms are needed. In aquatic environments, especially freshwater environments which may be used as a water source, rapid detection of pathogenic microbes is essential. Likewise, monitoring for the presence or absence of various functional genes can be used to indicate the type of microbial community present in the environment of interest. Therefore, the aim of this study was to develop and optimize methods necessary for the rapid assessment of freshwater microbial community structure and dynamics. Primers …

A Key To The Eggs And Nests Of Iowa Turtles, James L. Christiansen, Jeffrey R. Parmelee Jan 2003

A Key To The Eggs And Nests Of Iowa Turtles, James L. Christiansen, Jeffrey R. Parmelee

Journal of the Iowa Academy of Science: JIAS

Biologists often need to identify destroyed turtle nests where a predator has excavated and eaten the eggs. We present a dichotomous key to the eggs and nests of Iowa's turtles based on egg or nest morphology and known range of the species. Egg and nest morphology clearly separated most nests of the families Chelydridae and Kinosternidae and many of the Emydidae. However, egg morphology must be combined with known range to distinguish wood turtles (Glyptemys insculpta) from other emydids. These factors will also separate most false map turtles (Graptemys pseudogeographica) from other species. Similarities among egg morphologies, nest morphologies, and …

Department of Plant Pathology: Faculty Publications

Plant pathology has made significant progress over the years, a process that involved overcoming a variety of conceptual and technological hurdles. Descriptive mycology and the advent of chemical plant-disease management have been followed by biochemical and physiological studies of fungi and their hosts. The later establishment of biochemical genetics along with the introduction of DNA-mediated transformation have set the stage for dissection of gene function and advances in our understanding of fungal cell biology and plant–fungus interactions. Currently, with the advent of high-throughput technologies, we have the capacity to acquire vast data sets that have direct relevance to the numerous …

Implementation Plan For The Conservation Of Nebraska’S Eastern Saline Wetlands, Ted Lagrange, Terry Genrich, Glenn Johnson, Dan Schulz, Brent Lathrop

Nebraska Game and Parks Commission: Publications

Nebraska’s Eastern Saline Wetlands are the most limited and endangered wetland type and vegetation community in the State and are considered critically imperiled in Nebraska. These wetlands provide habitat for a variety of native plant and animal species that depend on a saline environment, including two endangered species.

Because of their location in and around the city of Lincoln, saline wetlands are ideally located to provide recreational opportunities and flood protection. Past impacts have resulted in the degradation of nearly 90% of the saline wetlands and those remaining face continued threats.

Although several programs are in existence to address saline …

Public Health And The Built Environment: Historical, Empirical, And Theoretical Foundations For An Expanded Role, Wendy Collins Perdue, Lawrence O. Gostin, Lesley A. Stone

Law Faculty Publications

In 2000, the Center for Disease Control and Prevention's National Center for Environmental Health issued a report that explored some of the ways in which "sprawl" impacts public health. The report has generated great interest, and state health officials are beginning to discuss the relationship between land use and public health. The CDC report has also produced a backlash. For example, the Southern California Building Industry Association labeled the report "a ludicrous sham" and argued that the CDC should stick to "fighting physical diseases, not defending political ones."

In this environment, it is understandable if the CDC looks to such …

The Ecology Of Two Estuarine-Dependent Tonguefishes, The Blackcheek Tonguefish (Symphurus Plagiusa) And The Offshore Tonguefish (S. Civitatium), In Coastal Louisiana, Theodore Scott Switzer Jan 2003

The Ecology Of Two Estuarine-Dependent Tonguefishes, The Blackcheek Tonguefish (Symphurus Plagiusa) And The Offshore Tonguefish (S. Civitatium), In Coastal Louisiana, Theodore Scott Switzer

LSU Doctoral Dissertations

The pattern and process of wetland loss in coastal Louisiana has the potential to influence the distribution and abundance of fishes and macroinvertebrates that utilize inshore estuarine habitat as nurseries during their early life histories. Current management plans designed to stabilize marsh elevations, such as freshwater diversions, may also influence the distribution and abundance of estuarine-dependent nekton. Given the importance of nursery function of inshore estuaries in coastal Louisiana, it is of interest to explore the response of estuarine-dependent organisms to environmental gradients. To this end I examined the ecology of two sympatric tonguefishes, the blackcheek tonguefish (Symphurus plagiusa) and …

Conservation Versus Survival: A Cultural Ecological Study Of Changing Settlement Patterns, Cultures, And Land Use In The Rio Platano Biosphere Reserve Of Northeast Honduras, Elizabeth Ann Fraser Jan 2003

Conservation Versus Survival: A Cultural Ecological Study Of Changing Settlement Patterns, Cultures, And Land Use In The Rio Platano Biosphere Reserve Of Northeast Honduras, Elizabeth Ann Fraser

LSU Doctoral Dissertations

Conserving ecologically valuable areas is proclaimed a priority by governments, institutions, and citizens throughout the world. Preventing the erosion of the remaining indigenous cultures also receives widespread support. In response to these desires, numerous protected areas now exist; spaces that theoretically should attain both ecological and cultural preservation. However, many of these areas are found within a larger setting plagued by ongoing struggles to meet basic needs. Often these larger problems create a challenge to, if not work in opposition to, the original aims of protected areas. This study of nine communities looks at these and related issues in the …

Second Natures: Media, Masculinity And The Natural World In Twentieth-Century American Literature And Film., Jeanne E. Hamming Jan 2003

Second Natures: Media, Masculinity And The Natural World In Twentieth-Century American Literature And Film., Jeanne E. Hamming

Graduate Theses, Dissertations, and Problem Reports (ETD)

This multidisciplinary dissertation examines the cultural anxieties associated with masculine identity in relation to both technology and nature in key literary texts and contemporary films of the twentieth-century: Ray Bradbury's “The Veldt” and The Martian Chronicles, Philip K. Dick's Do Androids Dream of Electric Sheep?, James Dickey's Deliverance , Don DeLillo's White Noise, Fight Club, American Psycho, Brazil, 12 Monkeys, and Dark City. Exploring the theoretical intersections of technoculture, ecocriticism, and gender studies, this dissertation analyzes the relationship between masculine identity and nature, as it is mediated by American technoculture. This relationship is marked by a discernible cultural malaise—a sense …

Wedge Veg : Gradient Analysis Of Impacts To Coastal Heath Vegetation At Wedge, Western Australia, Steven O'Dwyer Jan 2003

Wedge Veg : Gradient Analysis Of Impacts To Coastal Heath Vegetation At Wedge, Western Australia, Steven O'Dwyer

Theses : Honours

A reliable technique to assess impacts to coastal zones is required as coastal areas across the world are under pressure from an increasing human presence. The result is a greater level of disturbance to the coast, both marine and terrestrial. This study addresses a worldwide problem but is focused to a local scale. Wedge, on the central coast of Western Australia, supports a squatter shack settlement and has done so for approximately 50 years. Over time, the number of shacks, campers, tourists, 4WD's and motorbikes has increased. In recent years this growth has been significant. Management of this area and …

Quantitative Assessment Of Habitat Preferences For The Puerto Rican Terrestrial Frog, Eleutherdoctylus Coqui, Karen H. Beard, S. Mccullough, A. K. Eschtruth

Wildland Resources Faculty Publications

We conducted a quantitative analysis of adult and juvenile Eleutherodactylus coqui (coquí) habitat preferences in Puerto Rico. The analysis consisted of two surveys: one to quantify potential habitat and another to quantify habitat use. Coquís were found to use most habitats available to them; however, adults and juveniles preferred different plant species, habitat structural components, and heights from the forest floor. Adult and juvenile coquís had opposite associations with many important plant species in the forest (e.g., Prestoea montana and Heliconia carabea) and habitat structural components. Adults had a negative association with leaves and a positive association with leaf litter. …

Differences In Habitat Use By Blanding’S Turtles, Emydoidea Blandingii, And Painted Turtles, Chysemys Picta, In The Nebraska Sandhills, R. Bruce Bury, David J. Germano

United States Geological Survey: Staff Publications

We sampled a variety of wetlands in the Nebraska sandhills at Valentine National Wildlife Refuge. Significantly more individuals of painted turtles (Chrysemys picta) occurred in lakes and open waters than in marshes or small ponds, and the opposite was true for Blanding’s turtles (Emydoidea blandingii). Besides this marked difference in habitat use, 46% of the captured E. blandingii in pond/marsh habitat were juveniles, but only 31.6% in lakes and open water. Current information suggests that marshes and small ponds are important habitat for juvenile turtles, especially Emydoidea blandingii.

Impact Of Fire Management On The Ecology Of Collared Lizard (Crotaphytus Collaris) Populations Living On The Ozark Plateau, Jennifer A. Brisson, Jared L. Strasburg, Alan R. Templeton Jan 2003

Impact Of Fire Management On The Ecology Of Collared Lizard (Crotaphytus Collaris) Populations Living On The Ozark Plateau, Jennifer A. Brisson, Jared L. Strasburg, Alan R. Templeton

School of Biological Sciences: Faculty Publications

Eastern collared lizards (Crotaphytus collaris collaris) on the Ozark Plateau of southern Missouri are restricted to islands of rocky glade habitat located throughout the oak–hickory forests of the region. Human mediated suppression of fire negatively impacts collared lizard populations by permitting the overgrowth and consequent disappearance of this glade habitat and also leads to a dense forest understorey that impedes movement of lizards between glades. We present data examining the effects of a fire management regime on collared lizard populations we monitored via mark/recapture from 1998 to 2001. We demonstrate increased glade-to-glade dispersal, colonization of previously unoccupied glades, …

Invasiveness Of Some Biological Control Insects And Adequacy Of Their Ecological Risk Assessment And Regulation, Svata M. Louda, Amy E. Arnett, Tatyana A. Rand, F. L. Russell Jan 2003

Invasiveness Of Some Biological Control Insects And Adequacy Of Their Ecological Risk Assessment And Regulation, Svata M. Louda, Amy E. Arnett, Tatyana A. Rand, F. L. Russell

School of Biological Sciences: Faculty Publications

The problem of invasive species has reignited interest in biological control as a management tool. Classical biological control involves deliberate release of exotic natural enemies into new environments in an attempt to limit the density of an invasive species. Persistent, sustained limitation of invasive species by coevolved natural enemies is a seductive concept. Evidence now suggests, however, that biological control through the release of natural enemies can carry unanticipated ecological risks. There have been ecological side effects of distributing a deliberately introduced weevil (Rhinocyllus conicus) and an adventitious weevil (Larinus planus) for the biological control of …

Improving Precision And Reducing Bias In Biological Surveys: Estimating False-Negative Error Rates, Andrew J. Tyre, Brigitte Tenhumberg, Scott A. Field, Darren Niejalke, Kirsten Parris, Hugh P. Possingham

School of Biological Sciences: Faculty Publications

The use of presence/absence data in wildlife management and biological surveys is widespread. There is a growing interest in quantifying the sources of error associated with these data. We show that false-negative errors (failure to record a species when in fact it is present) can have a significant impact on statistical estimation of habitat models using simulated data. Then we introduce an extension of logistic modeling, the zero-inflated binomial (ZIB) model that permits the estimation of the rate of false-negative errors and the correction of estimates of the probability of occurrence for false-negative errors by using repeated visits to the …
