Sulfolipid Deficiency Does Not Affect The Virulence Of Mycobacterium Tuberculosis H37rv In Mice And Guinea Pigs, Cécile Rousseau, Oliver C. Turner, Erik Rush, Yann Bordat, Tatiana D. Sirakova, Pappachan E. Kolattukudy, Shannon Ritter, Ian M. Orme, Brigitte Gicquel, Mary Jackson Jan 2003

Sulfolipid Deficiency Does Not Affect The Virulence Of Mycobacterium Tuberculosis H37rv In Mice And Guinea Pigs, Cécile Rousseau, Oliver C. Turner, Erik Rush, Yann Bordat, Tatiana D. Sirakova, Pappachan E. Kolattukudy, Shannon Ritter, Ian M. Orme, Brigitte Gicquel, Mary Jackson

Faculty Bibliography 2000s

Lipids that are found only in the cell envelope of pathogenic mycobacteria, such as those containing multiple methyl-branched fatty acids, have long been thought to play a role in pathogenesis. Among these complex lipids, sulfolipids have been the most extensively studied over the last 50 years. The numerous biological effects exhibited by purified sulfolipids on phagocytic cells led to the idea that these molecules are probably important virulence factors facilitating the intracellular survival of Mycobacterium tuberculosis. However, definitive evidence to support this concept has been lacking. The recent construction of an isogenic sulfolipid-deficient mutant of M. tuberculosis H37Rv (Sirakova et …

Use Of Population Viability Analysis And Reserve Selection Algorithms In Regional Conservation Plans, Carlos Carroll, Reed F. Noss, Paul C. Paquet, Nathan H. Schumaker

Faculty Bibliography 2000s

Current reserve selection algorithms have difficulty evaluating connectivity and other factors necessary to conserve wide-ranging species in developing landscapes. Conversely, population viability analyses may incorporate detailed demographic data, but often lack sufficient spatial detail or are limited to too few taxa to be relevant to regional conservation plans. We developed a regional conservation plan for mammalian carnivores in the Rocky Mountain region using both a reserve selection algorithm (SITES) and, a spatially explicit population model (PATCH). The spatially explicit population model informed reserve selection and network design by producing data on the locations of population sources, the degree of threat …

Biochemical Function Of Msl5 (Pks8 Plus Pks17) In Mycobacterium Tuberculosis H37rv: Biosynthesis Of Monomethyl Branched Unsaturated Fatty Acids, Vinod S. Dubey, Tatiana D. Sirakova, Michael H. Cynamon, Pappachan E. Kolattukudy

Faculty Bibliography 2000s

We show that the disruption of one of the mycocerosic acid synthase (mas)-like genes, msl5 (pks8 plus pks17) in Mycobacterium tuberculosis H37Rv generates a mutant incapable of producing monomethyl branched unsaturated C-16 to C-20 fatty acids that are minor constituents of acyltrehaloses and sulfolipids. The msl5 mutation did not cause any significant change in the acyl lipid composition and also did not affect growth in culture, in mouse alveolar macrophage cell line MH-S, or in the murine lung.

Potential Involvement Of Extracellular Signal-Regulated Kinase 1 And 2 In Encystation Of A Primitive Eukaryote, Giardia Lamblia - Stage-Specific Activation And Intracellular Localization, John G. Ellis Iv, Monica Davila, Ratna Chakrabarti Jan 2003

Potential Involvement Of Extracellular Signal-Regulated Kinase 1 And 2 In Encystation Of A Primitive Eukaryote, Giardia Lamblia - Stage-Specific Activation And Intracellular Localization, John G. Ellis Iv, Monica Davila, Ratna Chakrabarti

Faculty Bibliography 2000s

Mitogen-activated protein kinase (MAPK) pathways are major signaling systems by which eukaryotic cells convert environmental cues to intracellular events such as proliferation and differentiation. We have identified Giardia lamblia homologues of two members of the MAPK family ERK1 and ERK2. Functional characterization of giardial ERK1 and ERK2 revealed that both kinases were expressed in trophozoites and encysting cells as 44- and 41-kDa polypeptides, respectively, and were catalytically active. Analysis of the kinetic parameters of the recombinant proteins showed that ERK2 is similar to5 times more efficient than ERK1 in phosphorylating myelin basic protein as a substrate, although the phosphorylating efficiency …

A Chloroplast Transgenic Approach To Hyper-Express And Purify Human Serum Albumin, A Protein Highly Susceptible To Proteolytic Degradation, Alicia Fernández-San Millán, Angel Mingo-Castel, Michael Miller, Henry Daniell

Faculty Bibliography 2000s

Human Serum Albumin (HSA) accounts for 60% of the total protein in blood serum and it is the most widely used intravenous protein in a number of human therapies. HSA, however, is currently extracted only from blood because of a lack of commercially feasible recombinant expression systems. HSA is highly susceptible to proteolytic degradation in recombinant systems and is expensive to purify Expression of HSA in transgenic chloroplasts using Shine-Dalgarno sequence (SD), which usually facilitates hyper-expression of transgenes, resulted only in 0.02% HSA in total protein (tp). Modification of HSA regulatory sequences using chloroplast untranslated regions (UTRs) resulted in hyper-expression …

Attenuation Of Mycobacterium Tuberculosis By Disruption Of A Mas-Like Gene Or A Chalcone Synthase-Like Gene, Which Causes Deficiency In Dimycocerosyl Phthiocerol Synthesis, Tatiana D. Sirakova, Vinod S. Dubey, Michael H. Cynamon, Pappachan E. Kolattukudy Jan 2003

Kolattukudy

Faculty Bibliography 2000s

Tuberculosis is one of the leading preventable causes of death. Emergence of drug-resistant tuberculosis makes the discovery of new targets for antimyrobacterial drugs critical. The unique mycobacterial cell wall lipids are known to play an important role in pathogenesis, and therefore the genes responsible for their biosynthesis offer potential new targets. To assess the possible role of some of the genes potentially involved in cell wall lipid synthesis, we disrupted a mas-like gene, msl7, and a chalcone synthase-like gene, pks10, with phage-mediated delivery of the disruption construct, in which the target gene was disrupted by replacement of an internal segment …

The Largest Open Reading Frame (Pks12) In The Mycobacterium Tuberculosis Genome Is Involved In Pathogenesis And Dimycocerosyl Phthiocerol Synthesis, Tatianna D. Sirakova, Vinod S. Dubey, Hwa-Jung Kim, Michael H. Cynamon, Pappachan E. Kolattukudy Jan 2003

Faculty Bibliography 2000s

The cell wall lipids in Mycobacterium tuberculosis are probably involved in pathogenesis. The largest open reading frame in the genome of M. tuberculosis H37Rv, pks12, is unique in that it encodes two sets of domains needed to produce fatty acids. Apks12-disrupted mutant was produced, and disruption was confirmed by both PCR analysis and Southern blotting. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis showed that a 430-kDa protein band present in the wild type was missing in the mutant. Matrix-assisted laser desorption ionization-time of flight mass spectrometry (MS) and liquid chromatography (LC)-MS analysis of tryptic peptides showed that 54 peptides distributed …

Public Relations For Small Business: A Communication Application To Business Management, Diane J. Morton Jan 2003

Public Relations For Small Business: A Communication Application To Business Management, Diane J. Morton

Theses

Large corporations and businesses in the public eye employ public relations professionals or commission public relations agencies to manage efforts that protect business reputation and mediate relationships between a business and its publics. To succeed, small businesses need similar relationships with their publics, but many business owners do not dedicate time or resources to public relations functions. Relatively little information is available in the public relations literature specifically for small businesses. The purpose of this thesis is to identify the applicability of corporate public relations functions for small businesses, and to formulate a public relations plan for entrepreneurs and small …

Gender Differences Of College Students And Pathological Gambling, Richard Clyde Vantreece Jan 2003

Gender Differences Of College Students And Pathological Gambling, Richard Clyde Vantreece

Theses

Gender differences among college students and pathological gambling have been studied in the past by various researchers. The purpose of this study was to investigate which gender was more likely to be at risk for a pathological gambling disorder. This population is interesting for the purpose of this study because the majority of pathological gamblers report beginning this habit in adolescence. Fifty college students were selected from the Human Subject Research Pool Department. Students completed a demographic sheet and the South Oaks Gambling Screen. T-tests were done to compare the differences among the genders. The results showed no significant difference …
