Human immunodeficiency virus type 1 (HIV-1) subtype C constitutes a majority of the newly transmitted cases of HIV-1 in many developing countries. There is growing evidence suggesting that subtype C viruses may display characteristics that make them distinct from subtype B and other subtypes, and such differences may affect its transmission and pathogenesis in infected individuals. In this study, HIV-1 sequences from the C2–V4 region of the envelope gene were analyzed from four infected mother–infant pairs (MIPs) after perinatal transmission in Zambia. Molecular chaperone complexes containing heat shock protein (Hsp) 70 and Hsp90 are regulated by cochaperones, including a subclass of regulators, such as Hsp70 interacting protein (Hip), C-terminus of Hsp70 interacting protein (CHIP), and Hsp70-Hsp90 organizing factor (Hop), that contain tetratricopeptide repeats (TPRs), where Hsp70 refers to Hsp70 and its nearly identical constitutive counterpart, Hsc70, together. These proteins interact with the Hsp70 to regulate adenosine triphosphatase (ATPase) and folding activities or to generate the chaperone complex. The relationships between wetland water conditions and breeding numbers of Mallard (Anas platyrhynchos), Northern Pintail (A. acuta), Blue-winged Teal (A. discors), and Northern Shoveler (A. clypeata) during May of 1992-1995, were examined on twelve study areas in the eastern Prairie Pothole Region. Data were collected on water levels (by wetland class [temporary, seasonal, semipermanent]), pond density (density of wet basins), and numbers of indicated pairs for each species from weekly roadside transect surveys. The diet of shovelnose sturgeon (Scaphirhynchus platorhynchus) in the Missouri River downstream from Gavins Point Dam was characterized in 1993 (N=100 fish) and 1994 (N=103 fish) for comparison with similar data collected in 1971-72 (N=130 fish) by Modde and Schmulbach (1977, hereafter Modde). The primary difference among years in the South Dakota studies was river discharge, which was low in 1993 (9-22 kcfs; kcfs=thousand cubic feet per second), moderate in 1994 (18-31 kcfs), and high in 1971-72 (26-49 kcfs). We evaluated spatiotemporal variation in clutch sizes of Northern Pintails (pintails; Anas acuta) nesting in California (1985 to 1996), North Dakota (1982 to 1985), Saskatchewan (1982 to 1985) and Alaska (1991 to 1993) to determine whether seasonal declines in clutch size varied in ways that were consistent with a controlling influence of increasing day length. Pintails began nesting in mid-March in California, mid-April in North Dakota and Saskatchewan, and mid-May in Alaska. There is an increasing need to develop field immobilization techniques that allow researchers to handle safely swift foxes (Vulpes velox) with minimal risk of stress or injury. We immobilized captive swift foxes to determine the safety and effectiveness of ketamine hydrochloride and xylazine hydrochloride at different dosages. We attempted to determine appropriate dosages to immobilize swift foxes for an adequate field-handling period based on three anesthesia intervals (induction period, immobilization period, and recovery period) and physiologic responses (rectal temperature, respiration rate, and heart rate). The United States Fish and Wildlife Service has determined that the black-tailed prairie dog (Cynomys ludovicianus) warrants listing as a threatened species under the Endangered Species Act. Central to any conservation planning for the black-tailed prairie dog is an appropriate detection and monitoring technique. Because coarse-resolution satellite imagery is not adequate to detect black-tailed prairie dog colonies, we examined the usefulness of recently available high-resolution (1-m) satellite imagery. In 6 purchased scenes of national grasslands, we were easily able to visually detect small and large colonies without using image-processing algorithms. The ability of Pseudomonas syringae pv. tomato DC3000 to be pathogenic on plants depends on the Hrp (hypersensitive response and pathogenicity) type III protein secretion system and the effector proteins it translocates into plant cells. Through iterative application of experimental and computational techniques, the DC3000 effector inventory has been substantially enlarged. Five homologs of known avirulence (Avr) proteins and five effector candidates, encoded by genes with putative Hrp promoters and signatures of horizontal acquisition, were demonstrated to be secreted in culture and/or translocated into Arabidopsis in a Hrp-dependent manner. Citrus tristeza virus (CTV), a member of the Closteroviridae, has a 19.3-kb positive-stranded RNA genome that is organized into 12 open reading frames (ORFs) with the 10 3' genes expressed via a nested set of nine or ten 3'-coterminal subgenomic mRNAs (sgRNAs). Relatively large amounts of negative-stranded RNAs complementary to both genomic and sgRNAs accumulate in infected cells. As is characteristic of RNA viruses, wild-type CTV produced more positive than negative strands, with the plus-to-minus ratios of genomic and sgRNAs estimated at 10 to 20:1 and 40 to 50:1, respectively. The Iterated Prisoner’s Dilemma (IPD) is a central paradigm in the study of animal cooperation. According to the IPD framework, repeated play (repetition) and reciprocity combine to maintain a cooperative equilibrium. However, experimental studies with animals suggest that cooperative behavior in IPDs is unstable, and some have suggested that strong preferences for immediate benefits (that is, temporal discounting) might explain the fragility of cooperative equilibria. We studied the effects of discounting and strategic reciprocity on cooperation in captive blue jays. Our results demonstrate an interaction between discounting and reciprocity. Taurine efflux occurs in association with cell swelling in both hyposmotic and isosmotic conditions and during cell shrinkage in apoptotic death. Release occurs through a leak pathway, is largely Ca2+-independent and is sensitive to Cl– channel blockers. Taurine efflux elicited by hyposmolarity is reduced or suppressed by tyrosine kinase blockers and increased by tyrosine phosphatase inhibitors. The specific kinases involved are still unknown and may be different in the various cell types. Non-receptor and scr-related protein kinases have been identified in some cells as elements that directly phosphorylate the taurine efflux pathway. Possible tyrosine kinase targets are …

The present invention relates to methods and compositions for eliciting an immune response against bovine viral epitopes. The methods comprise combining at least one heat shock protein with at least one bovine viral epitope to form a purified epitope/heat shock protein complex and administration of an immune system stimulating amount of the purified epitope/heat shock protein complex. The compositions comprise, a purified epitope/heat shock protein complex comprising at least one bovine viral epitope complexed with at least one heat shock protein, and a pharmaceutically acceptable carrier, diluent or excipient.

The longitudinal evolution of HIV-1 phenotypes was studied in a cohort of six vertically infected children with early onset and rapid progression of clinical disease. Among 30 viral isolates obtained from peripheral blood, tropisms for both human blood-derived cells (macrophages, T-lymphocytes), and for human neural (brain-derived) cells (microglia, astrocytes) were determined, as was chemokine co-receptor usage. All children harbored from birth macrophage-tropic isolates using the CCR5 co-receptor. Two children later developed T-cell tropic isolates with CXCR4 and CCR3 usage. While all six patients developed neurological abnormalities, only three produced neural cell tropic isolates, which used CCR5. However, early and persistent …

In avian species, spermatozoa reside in the oviduct for prolonged periods in specialized structures known as sperm storage tubules, but little is known about the relative distribution of spermatozoa in these tubules after successive inseminations by different males. The staining efficacies of various fluorescent dyes for fowl and turkey spermatozoa were evaluated to investigate one proposed mechanism of sperm competition. Hens were then inseminated at different intervals with stained and unstained spermatozoa to observe the spatial distribution of spermatozoa within the storage tubules. Several novel fluorescent lipophilic tracers that successfully stain mammalian spermatozoa either did not stain fowl or turkey …

Brucellosis is endemic in free-ranging elk (Cervus elaphus) and bison (Bison bison) in the Greater Yellowstone Area (GYA; USA). It is possible that an oral brucellosis vaccine could be developed and disseminated in the GYA to reduce disease transmission. In 1999, an interdisciplinary research team at the University of Nebraska established a field experiment to (1) quantify and understand the yield potential of corn and soybean under irrigated conditions, (2) identify efficient crop management practices to achieve yields that approach potential levels, and (3) determine the energy use efficiency, global warming and soil C-sequestration potential of intensively managed corn systems. The experiment compares systems that represent different levels of management intensity expressed as combinations of crop rotation (continuous corn, corn-soybean), plant density (low, medium, high) and nutrient management (recommended best management vs. intensive management). Understanding long-term management effects on soil properties is necessary to determine the relative sustainability of cropping systems. Soil physical, chemical, and biological properties were measured in a long-term cropping system study in the Western Corn Belt. Properties were evaluated after 16 yr in four crop sequences [continuous corn (Zea mays L.) (CC), corn–soybean [Glycine max. (L.)] (C–SB), corn–oat (Avena sativa L.) + clover (80% sweet clover [Melilotus officinalis L.] and 20% red clover [Trifolium pratense L.])–grain sorghum [(Sorghum bicolor (L.)
