Anisomycin Activates Jnk And Sensitises Du 145 Prostate Carcinoma Cells To Fas Mediated Apoptosis, James Curtin, Thomas Cotter

Treatment of the hormone refractory prostate cancer cell line DU 145 with sublethal concentrations of chemotherapeutic drugs has been reported to sensitise these cells to Fas mediated apoptosis. However, the mechanism by which this occurs has not been determined. Our group has shown that inhibition of JNK activity completely abrogates the effects of chemotherapeutic drugs. Using anisomycin, a potent JNK agonist, we have demonstrated a role for JNK in Fas mediated apoptosis in DU 145 cells. Inhibition of Caspase 8 and Caspase 9 completely inhibits this process which suggests that DU 145 cells require mitochondrial amplification of the Fas apoptotic …

This paper presents a novel computational approach, the discrete singular convolution (DSC) algorithm, for analysing plate structures. The basic philosophy behind the DSC algorithm for the approximation of functions and their derivatives is studied. Approximations to the delta distribution are constructed as either bandlimited reproducing kernels or approximate reproducing kernels. Unified features of the DSC algorithm for solving differential equations are explored. It is demonstrated that different methods of implementation for the present algorithm, such as global, local, Galerkin, collocation, and finite difference, can be deduced from a single starting point. The debate over Title IX and athletics has neglected the impact that Title IX could have on high school athletic programs. An increased focus on high school athletic opportunity is important because the benefits for girls that participate in athletics as they grow up, and for society, are so significant. In this article, the author proposes that compliance with Title IX in high school athletics is only appropriately measured by way of a test that requires schools maintain athletic participation rates for each sex in proportion to the population of each sex within the school. Partial ion yield spectroscopy provides a very detailed picture of fragmentation processes following core excitation in isolated molecules. We exploit this potential in the analysis of decay processes following C1s→π and C1s→Rydberg excitations in ethylene and acetylene. We show that the relative intensity of spectral features related to the excitation to empty molecular orbitals or to Rydberg states is a function of the time variation of the fragmentation process. Namely, we see an intensity increase in the Rydberg states compared to the molecular orbitals as the fragmentation process becomes more extensive, a result attributable to the diffuse nature of the …

Ground corn and soybean meal are common ingredients in feed mixes. The knowledge of their mechanical properties is important to the feed manufacturer and consumer. Changes in these properties can lead to abnormally high or low levels of active ingredients in finished feed, thus decreasing its quality. Mechanical properties of wheat, corn meal, and soybean meal were investigated using a modified direct shear apparatus. The moisture content (wet basis), uncompacted bulk density, and particle density were: 10.4%, 733 kg/m3, and 1410 kg/m3 for soft red winter wheat; 11.4%, 583 kg/m3, and 1350 kg/m3 for …

In a game theoretic framework it can be argued that a gender sensitive institution is an offshoot of certain social conditions, which in most cases need to be acted upon by some anti-establishment catalytic agent. Given the fact that among about half of the population there is a need for such an institution, the main function of a catalytic agent is to engineer a conversion of that need into an active demand. In a society characterized by gender exploitation, catalytic agent can only come exogenously. For a sub-society it is easier to come across such an exogenous catalytic agent. The …

Bovine immunodeficiency virus (BIV) and Jembrana disease virus (JDV) are closely related bovine lentiviruses that are difficult to distinguish by presently available diagnostic methods. Recently, in our laboratory, a monoclonal antibody (MAb; MAb 10H1) against the BIV Gag protein identified a differential epitope, located at the 6.4-kDa N terminus of a 29-kDa Gag capsid protein, which was absent in JDV. To define the essential amino acids of the epitope, a series of primers within the 163 bp of DNA corresponding to the 6.4-kDa protein were designed. In order to apply manure or compost to fulfill the nutrient requirements of a crop, knowledge of the amount of nutrients mineralized following application is needed. Nutrient mineralization from applied manure depends on temperature, soil moisture, soil properties, manure characteristics, and microbial activity. Since these factors cannot be accurately predicted, nutrient mineralization from applied manure can only be approximated. Nitrogen (N) availability from applied manure includes the inorganic N (NO3-N and NH4-N) in manure plus the amount of organic N mineralized following application. Ageing (senescence) has never been demonstrated convincingly in any insect in the wild, where mean life-spans are probably much shorter than in the laboratory1, and most evidence for senescence in other wild animals (such as mammals) is limited to their reduced survival with age2. Here we show that ageing is detectable in wild populations of a very short-lived insect, the antler fly (Protopiophila litigata), and causes debilitating and costly effects that force a decline not only in survival probability, but also in the reproductive rate of males. Objective - This study examined the association between cardiorespiratory fitness and C-reactive protein (CRP), with adjustment for weight and within weight categories.
Methods and Results - We calculated median and adjusted geometric mean CRP levels, percentages of individuals with an elevated CRP (≥2.00 mg/L), and odds ratios of elevated CRP across 5 levels of cardiorespiratory fitness for 722 men. CRP values were adjusted for age, body mass index, vitamin use, statin medication use, aspirin use, the presence of inflammatory disease, cardiovascular disease, and diabetes, and smoking habit. Cachexia is commonly seen in cancer and is characterized by severe muscle wasting, but little is known about the effect of cancer cachexia on expression of contractile protein isoforms such as myosin. Other causes of muscle atrophy shift expression of myosin isoforms toward increased fast (type II) isoform expression. We injected mice with murine C-26 adenocarcinoma cells, a tumor cell line that has been shown to cause muscle wasting. Mice were killed 21 days after tumor injection, and hindlimb muscles were removed. Myosin heavy chain (MHC) and myosin light chain (MLC) content was determined in muscle homogenates by SDS-PAGE. Dead reckoning is an on-line form of spatial navigation used by an animal to identify its present location and return directly to a starting location, even after circuitous outward trips. At present, it is not known which of several self-movement cues (efferent copy from movement commands, proprioceptive information, sensory flow, or vestibular information) are used to compute homeward trajectories. To determine whether vestibular information is important for dead reckoning, the impact of chemical labyrinthectomy was evaluated in a test that demanded on-line computation of a homeward trajectory. Human neutrophils are rescued from apoptosis following incubation with once-washed, fibroblast-derived Toxoplasma gondii tachyzoites. Both infected and uninfected neutrophils are rescued, implicating a soluble mediator. In this study we investigated the origin and identity of this soluble mediator. Neutrophils were incubated either with purified tachyzoites or with conditioned medium derived from T. gondii-infected human fibroblasts. Conditioned medium was found to be a potent stimulus that delayed neutrophil apoptosis up to 72 h, whereas purified and extensively washed tachyzoites had no effect. Delayed apoptosis correlated with up-regulation of the neutrophil antiapoptotic protein, Mcl-1, and the neutrophil interleukin 3 receptor alpha subunit …

This thesis examines data on gender and the incidence and prevalence of delinquent and criminal offending, as well as gender differences in the context of offending for a sample of high school students in Omaha, Nebraska. Context refers to the specific attributes of a particular offense, whose interrelationship describes both the features and the circumstances of the offense. A focus on gender differences in the context of offending highlights how gender impacts the structural and social conditions that are related to commission of delinquent and criminal acts, and the findings of this study underscore the importance of this research.
