Merlin Phosphorylation By P21-Activated Kinase 2 And Effects Of Phosphorylation On Merlin Localization, Joseph L. Kissil, Kristen C. Johnson, Matthew S. Eckman, Tyler Jacks

Life Sciences Faculty Scholarship

The Nf2 tumor suppressor gene product merlin is related to the membrane-cytoskeleton linker proteins of the band 4.1 superfamily, including ezrin, radixin, and moesin (ERMs). Merlin is regulated by phosphorylation in a Rac/cdc42-dependent fashion. We report that the phosphorylation of merlin at serine 518 is induced by the p21-activated kinase PAK2. This is demonstrated by biochemical fractionation, use of active and dominant-negative mutants of PAK2, and immunodepletion. By using wild-type and mutated forms of merlin and phospho-directed antibodies, we show that phosphorylation of merlin at serine 518 leads to dramatic protein relocalization.

OLD-GROWTH TREE PRESERVATION. INITIATIVE STATUTE. Protects California's old growth trees by prohibiting cutting or significant harm to heritage trees in California except as permitted in statute and by creating buffer zones. Requires that timber and other operations be conducted consistent with statute. Creates Heritage Tree Preservation Fund administered by Wildlife Conservation Board. Provides for civil penalties and injunctive relief against any person who performs, conducts, orders, or directs timber operations in violation of statute. Also provides for investigation and disciplinary action against any registered professional forester for material misstatement by failing to designate heritage trees in timber harvesting plan. The isolation and characterization of cDNAs encoding poly(ADP-ribose) glycohydrolase (PARG) enzymes and the amino acid sequences of PARGs from several species are described. PARG is involved in the cellular response to DNA damage and its proper function is associated with the body's response to neoplastic disorder inducing agents and oxidative stress. Expression vectors containing the cDNAs and cells transformed with the vectors are described. Probes and primers that hybridize with the cDNAs are described. Expression of the cDNA in E. coli results in an enzymatically active protein of about 111 kDa and an active fragment of about 59 kDa. Methods …

The present invention provides materials and methods related to causing insect resistance in plants. Specifically, the present invention provides nucleic acid compounds which encode a teratocyte secreted protein (TSP) which is capable of causing inhibition of growth and developmental arrest in Lepidoptera larvae. Brucella melitensis is a facultative intracellular bacterial pathogen that causes abortion in goats and sheep and Malta fever in humans. The genome of B. melitensis strain 16M was sequenced and found to contain 3,294,935 bp distributed over two circular chromosomes of 2,117,144 bp and 1,177,787 bp encoding 3,197 ORFs. By using the bioinformatics suite ERGO, 2,487 (78%) ORFs were assigned functions. The origins of replication of the two chromosomes are similar to those of other α-proteobacteria. Housekeeping genes, including those involved in DNA replication, transcription, translation, core metabolism, and cell wall biosynthesis, are distributed on both chromosomes. Type I, II, …

The extensive ab initio quantum chemical study of ten stationary point on the water dimer potential energy surface (PES) was discussed. The geometry optimizations were carried out using the CCSD(T) method in conjunction with the [TZ2P(f,d) + dif] basis set. The study showed that the optimized structures of (H2O)2 PES were quite similar to those obtained at the MP2 level with much smaller basis sets and the number of imaginary vibrational frequencies or hessian index of each stationary point was unchanged despite fairly significant geometrical differences for some of the optimized structures.

A new species and genus of digenetic trematode of the family Dicrocoeliidae is described from 2 species of grass mice, Akodon fumeus and Akodon mimus, both collected from an isolated area in the eastern part of the Andes mountains or Yungas of Bolivia. This species differs from all known dicrocoeliids in the unique disposition of the vitelline fields, the much less extensive uterus through the body of the worm (extending posteriad only just past the midline), and cecal branches that are generally conspicuous and lined with a thick layer of glandular epithelial tissue.

This paper examines the historical allocation of materials funds at Providence College and describes a study to develop an allocation formula based on circulation patterns. With selection of materials shared by faculty and librarians, it also considers whether faculty or librarians make better selectors and raises questions about re-allocation of funds based on those findings.

HIV-1HXB2 5′LTR region, most of BIVR29 gag-pol segment and HIV-1HXB2 pol IN-3′LTR region were respectively amplified. A chimeric clone, designated as pHBIV3753, was constructed by cloning three fragments sequentially into pUC18. MT4 cells were transfected with pHBIV3753. The replication and expressions of the chimeric virus (HBIV3753) were monitored by RT activity and IFA. The results firstly demonstrated that it is possible to generate a new type of the BIV/HIV-1 chimeric virus containing BIV gag-pol gene.

Phosphoglycan messengers (PGMs) which are carbohydrate derivatised phosphatidyl cyclitols are disclosed, together with the finding that these substances are biologically active, e.g. in lowering blood glucose levels. These compounds are distinct from GPI-anchors disclosed in the prior art as these GPI-anchors are protein linked and are not biologically active.

PPhosphoglycan messengers (PGMs) which are carbohydrate derivatised phosphatidyl cyclitols are disclosed, together with the finding that these substances are biologically active, especially in modulating plasma cholesterol, plasma triglycerides and/or high density lipoprotein levels and/or modulating the LDL:HDL ratio, and for the treatment of lipodystrophy or dyslipidemia. These compounds are distinct from GPI-anchors disclosed in the prior art as these GPI-anchors are protein linked and are not biologically active.
