Characterization Of Metallothionein Expression In Response To Cadmium In The 267-B1, Normal Prostate Cell Line, Madhuri G. Neriyanuri

The objective of the study was to characterize the mRNA and protein expression of the MT gene in response to Cd2+ exposure in the 267-B1 immortalized normal human neonatal prostate cell line. The cells were exposed to 18muM Cd2+ for 4hr and rescued for 48hr in P4-8F normal medium. Samples were obtained at 0, 1, 2, 4hr exposure and 4, 8, 12, 16, 24, 36, and 48hr recovery. Basal levels of mRNA for MT-1E, MT-1X, and MT-2A genes in 267-B1 cells were similar to the expression of the G3PDH where as MT-3 had a low basal level expression. Exposure to …

The risk based sport fish consumption advisory protocol, West Virginia Sport Fish Consumption Advisory Guide (the Guide), was drafted for the state. Fish were collected from ten West Virginia sites (5 previously identified as contaminated, 5 previously identified as uncontaminated). Fish samples were collected, processed and analyzed for total mercury concentration and Aroclors (PCBs). Recommended meal frequencies were calculated from contaminant levels and based on the Guide. Mean fillet sample contaminant levels were compared to composite sample levels and found to require the same level or in one case a more protective of consumption advisory. Tall fescue (Lolium arundinaceum), an agronomically important forage grass, is typically associated with a mutualistic asexual fungus Neotyphodium coenophialum. Endophytic fungal growth is confined to the intercellular spaces of the plant and induces no symptoms. The endophyte partner enhances its host's fitness by improving nutrient acquisition as well as by providing protection from various abiotic and biotic stresses. However, grass-endophyte partnerships are detrimental to grazing livestock, due to the production of an array of fungal alkaloids. The Eifelian stage of eastern North America has produced the remains of a substantial number of fish taxa, mostly from the Onondaga Limestone of New York and the Columbus and Delaware limestones of Ohio. Fish specimens in rock matrix from the Buffalo Science Museum and the Cleveland Museum of Natural History were compared to rock samples taken from quarries in Cheektowaga, New York and Warrensburg, Ohio, in order to determine the units from which the fish specimens originally came. B lymphocytes are continually produced in bone marrow from pluripotential hematopoietic stem cells. Lymphopoiesis is characterized by a series of highly regulated genotypic and phenotypic changes resulting in immunocompetent effector cells which express cell surface immunoglobulin. Our laboratory has focused on defining extracellular signals regulating lymphoid progenitor cell survival, proliferation, and differentiation. These studies have demonstrated that pro-B cell survival, proliferation, and differentiation are regulated by interactions with fibroblastic stromal cells in the hematopoietic microenvironment. However, specific molecular mechanisms by which stromal cells regulate B lymphoid development are largely unknown. In an attempt to better understand molecular mechanisms regulating maturation …

Proton Coupled Electron Transfer (PCET) reactions play a vital rote in a variety of biological and chemical processes. Prior work by E. Laviron developed the theory of proton coupled electron transfer kinetics based on a stepwise mechanism with the transfer coefficient for electron transfer remaining constant at all overpotentials. The present work explores the consequences of a potential-dependent transfer coefficient as applied to redox couples attached to electrodes. Specifically, this work focuses on the 1-electron, 1-proton (1e1H) system. The successes of the unmanned aerial vehicle (UAV) in Afghanistan most likely will accelerate the introduction of UAVs into the military force structure. What emerged in Afghanistan was a tiered observation-detection-targeting system consisting of spacecraft, the Global Hawk UAV, the Predator UAV, and often a spotter on the ground. However, in complex terrain, as in Afghanistan, and in urban situations, airborne assets may be needed much closer to the ground. A low-altitude tier of UAVs—mini UAVs—could serve such a purpose.

Despite the tolerance of common reed grass to environmental extremes, mid-water stands in the Winnebago pool lakes of central Wisconsin appear to be diminishing. Formerly occupying shoreline locations, water level manipulations subsequent to dam construction beginning in the 1850's have isolated reed stands off shore. These stands have persisted but casual observations indicate that stand size has been declining. To address this perception we obtained an approximately decadal series of aerial photographs dating back to 1937 for four stands in Lake Poygan. Annual records were available for 1986–94. Using image analysis software, we determined shape and size metrics. Changes in …

The most commonly used system of writing by and for blind persons is called Braille, named after its French inventor, Louis Braille (1809-1952). Braille is comprised of a rectangular six-dot cell on its end, with up to 63 possible combinations using one or more of the six dots. Braille is the only reliable method of literacy for blind persons, because it enables them to read and write by touch¹.

The survival of E. coli O157:H7 in acid foods for weeks and its prolonged survival in refrigerated acid foods are well documented. This prompted the study to evaluate survival of E. coli O157:H7 in tomato and processed tomato products. The pH of the various products ranged from 4.2 to 4.8 and some products contained preservatives such as vinegar. Samples were separately inoculated with a mixture of four E. coli O157:H7 strains previously isolated from hamburger meat at lower and higher initial inoculum levels, incubated at 4 and 25 °C and assayed for survival at regular intervals. Two methods of decomposition of probabilistic relations are presented in this paper. They consist of splitting relations (blocks) into pairs of smaller blocks related to each other by new variables generated in such a way so as to minimize a cost function which depends on the size and structure of the result. The decomposition is repeated iteratively until a stopping criterion is met. Topology and contents of the resulting structure develop dynamically in the decomposition process and reflect relationships hidden in the data.

During late spring through summer of 1994 and 1995, 290 randomly selected stream sites in Nebraska, Kansas, and Missouri were sampled once for several parameters including conductivity, turbidity, total phosphorus, nitrate-nitrite nitrogen, the index of biotic integrity, and a habitat index. Based on landscape data from watersheds that were delineated for each sampling location, interrelationships were examined between these water quality parameters and land use/land cover, the normalized difference vegetation index (NDVI), and vegetation phenological metrics derived from the NDVI. Heritabilities, commonly used to predict evolutionary potential, are notoriously low for behaviors. Apart from strong contributions of environmental variance in reducing heritabilities, the additive genetic components can be very low, especially when they are camouflaged by nonadditive genetic effects. We first report the heritabilities of courtship traits in founder‐flush and control populations of the housefly (Musca domestica L.). We estimated the heritability of each male and female display through the regression of the courtships involving daughters and sons (with randomly selected mates) onto the “midparental” courtship values of their parents. During animal development, organ size is determined primarily by the amount of cell proliferation, which must be tightly regulated to ensure the generation of properly proportioned organs. However, little is known about the molecular pathways that direct cells to stop proliferating when an organ has attained its proper size. We have identified mutations in a novel gene, shar-pei, that is required for proper termination of cell proliferation during Drosophila imaginal disc development. Clones of shar-pei mutant cells in imaginal discs produce enlarged tissues containing more cells of normal size. We show that this phenotype is the result of both …

Reviews of the literature have identified few consistent theoretical approaches in the assessment of death anxiety and death attitudes for patients diagnosed with a potentially life threatening illness, such as cancer. This illness experience forces patients to contemplate their mortality. The present study employed a portion of the Death Anxiety Model to examine the relationships between death salience, beliefs about the self, beliefs about the world, death meaning, and death anxiety. Three groups (N = 121) who differed on death salience were examined: cancer survivors (intrapersonal - high salience), spouses of cancer survivors (interpersonal - moderate salience), and healthy patients …

1. The degree to which the RhoA kinase (ROK) blockers, Y-27632 (1 µM) and HA-1077 (10 µM), and the PKC blocker, GF-109203X (1 µM), reduced force produced by carbachol, a muscarinic receptor agonist, and phenylephrine, an α-adrenoceptor agonist, was examined in rabbit stomach fundus smooth muscle.

2. When examining the effect on cumulative carbachol concentration-response curves (CRCs), ROK and PKC blockers shifted the potency (~EC50) to the right but did not reduce the maximum response.

3. In a single-dose carbachol protocol using moderate (~EC50) and maximum carbachol concentrations, Y-27632 and HA-1077 reduced peak force, but GF-109203X had no effect. By …

This study explored an individual's need for control and the level of violence within a dating relationship.

This was a self-report study. Subjects consisted of 175 students from a university in the southern Appalachian region of the U.S. Questionnaires were combined with a scenario depicting violent behavior. Subjects were asked to rate their level of control on the Fundamental Interpersonal Relations Inventory (FIRO-B), to complete the Conflict Tactics Scale - revised (CTS-2), and to rate the acceptability of the scenario. Data were analyzed using an ANOVA.

Because of increasing numbers in wildlife, immunocontraceptives are being developed to decrease these animals and other birds. This study was conducted in conjunction with the United States Geological Survey Midcontinent Science Center in Fort Collins, Colorado (USGS), to determine whether mammalian GnRH (m-GnRH) can be used as a potential immunocontraceptive to decrease reproductive fertility in mature Japanese quail.

Eighty male and female Japanese quail were injected with either mGnRH, chicken GnRH-I (c-GnRH-I), chicken GnRH-II (c-GnRH-II), or vehicle (controls). Insect herbivory is common, but the conditions under which it reduces the fitness and population size of plants remain poorly understood. We quantified population-level impacts of floral herbivory by specialized insects on cobweb thistle (Cirsium occidentale var. occidentale) in a California coastal dune ecosystem, and then compared these demographic effects to those published for Platte thistle (Cirsium canescens) in similarly designed exclusion experiments in the continental sand dune ecosystem in the Great Plains.
