Customizing Conception: A Survey Of Preimplantation Genetic Diagnosis And The Resulting Social, Ethical, And Legal Dilemmas, Jason Christopher Roberts Jul 2002

Customizing Conception: A Survey Of Preimplantation Genetic Diagnosis And The Resulting Social, Ethical, And Legal Dilemmas, Jason Christopher Roberts

Duke Law & Technology Review

One in six American couples experience difficulties conceiving a child. With fertility rates at an all time low, the business of treating infertility is booming. However, due to the United States prohibition on government funding for embryonic research, the $4 billion industry of assisted reproductive technologies (ART) has been incompletely monitored and largely removed from oversight. Additionally, due to the fervent abortion debate, in vitro fertilization (IVF) was introduced in the United States without a research phase and procedures have been forced to evolve in the private sector. Thus, the checks and balances on medical innovation that are generally imposed …

Efficient Synthesis Of Secondary Amines By Selective Alkylation Of Primary Amines, Kyung Woon Jung Jul 2002

Efficient Synthesis Of Secondary Amines By Selective Alkylation Of Primary Amines, Kyung Woon Jung

USF Patents

A method for selective mono-N-alkylation of primary amines to produce secondary amines that are substantially free of overalkylated tertiary amines and quaternary ammonium salts, under mild reaction conditions without the necessity of protecting groups. Compounds of the class of secondary amines are produced by reacting an alkyl halide with an alkyl amine in anhydrous solvent, preferably dimethyl sulfoxide or N,N-dimethylformamide, in the presence of 0.1 to 3 molar equivalents of a cesium base. Optionally, the extent and selectivity of mono-N-alkylation is enhanced by addition to the reaction mixture of a powdered molecular sieve material for removal of water produced by …

Regulation Of Retinoic Acid Receptor Α By Protein Kinase C In B16 Mouse Melanoma Cells, Goran Boskovic, Dinakar Desai, Richard M. Niles Jul 2002

Regulation Of Retinoic Acid Receptor Α By Protein Kinase C In B16 Mouse Melanoma Cells, Goran Boskovic, Dinakar Desai, Richard M. Niles

Biochemistry and Microbiology

We have previously found that retinoic acid stimulates the expression of protein kinase Cα (PKC) in B16 mouse melanoma cells. Because it has been reported that PKC can phosphorylate retinoic acid receptor (RAR) and alter its function, we determined whether changes in the level and/or activity of PKC could affect the expression or function of the RAR in B16 melanoma. Using in vivophosphorylation and band shift techniques, we could not demonstrate that altering PKC activity and/or protein level changed thein vivo phosphorylation of RARα. However activation of PKC resulted in increased RARα protein. Increased receptor protein correlated with …

Tolerance, Performance, And Aerobic Metabolism Of Redband Trout (Oncorhynchus Mykiss Ssp.) During Acute Exposure To Elevated Temperature, Karen Reed Lizars Jul 2002

Tolerance, Performance, And Aerobic Metabolism Of Redband Trout (Oncorhynchus Mykiss Ssp.) During Acute Exposure To Elevated Temperature, Karen Reed Lizars

Dissertations and Theses

The objective of this thesis was to determine whether the redband trout (Oncorhynchus mykiss ssp.) of Oregon's inland desert basins are negatively affected by the high peak temperatures (> 20 °C) often observed in their native streams. Small (40 - 140 g) redband trout from three streams (Bridge, Rock, and Twelve-mile creeks) were collected and tested on-site for aspects of thermal physiology, including: critical swimming velocity (Ucrit), maximum metabolic rate (MO2), metabolic power, and total cost of transport (TCT) at 24 °C; routine MO2 at 14 to 2: 28 °C: and critical thermal maximum (CTM). At …

Beaver Herbivory And Its Effect On Cottonwood Trees: Influence Of Flooding Along Matched Regulated And Unregulated Rivers, Stewart W. Breck, Kenneth R. Wilson, Douglas C. Andersen

United States Department of Agriculture Wildlife Services: Staff Publications

We compared beaver (Castor canadensis) foraging patterns on Fremont cottonwood (Populus deltoides subsp. wislizenii ) saplings and the probability of saplings being cut on a 10 km reach of the flow-regulated Green River and a 8.6 km reach of the free-flowing Yampa River in northwestern Colorado. We measured the abundance and density of cottonwood on each reach and followed the fates of individually marked saplings in three patches of cottonwood on the Yampa River and two patches on the Green River. Two natural floods on the Yampa River and one controlled flood on the Green River between …

Role Of Lipid Peroxidation And Antioxidant Enzymes In Omega 3 Fatty Acids Induced Suppression Of Breast Cancer Xenograft Growth In Mice, W. Elaine Hardman, Jesus Munoz Jr., Ivan L. Cameron Jul 2002

Role Of Lipid Peroxidation And Antioxidant Enzymes In Omega 3 Fatty Acids Induced Suppression Of Breast Cancer Xenograft Growth In Mice, W. Elaine Hardman, Jesus Munoz Jr., Ivan L. Cameron

Biochemistry and Microbiology

Background:

Supplementing mice with high levels of dietary n-3 polyunsaturated fatty acids (PUFAs) increases the n-3 PUFAs in cell membranes, increases the susceptibility of the cells for lipid peroxidation (LPO) and decreases the growth rate of mammary and other tumors. However, the results of an earlier study indicated that a factor in addition to LPO was involved in the reduction in tumor growth in n-3 PUFAs fed mice. Athymic mice bearing MDA-MB-231 human breast carcinoma xenografts, were fed fish oil concentrate (FOC) or control diets, with and without supplemental Vitamin E (2000 IU /kg diet) and were sacrificed both before …

Appropriate Aims: Setting Boundaries For Reprogenetic Technology, Dana Ziker Jul 2002

Appropriate Aims: Setting Boundaries For Reprogenetic Technology, Dana Ziker

Duke Law & Technology Review

Not too long ago, ten fingers and ten toes defined a successful birth. Not too far from now, ten fingers and ten toes will be just the beginning. Parents always hope for a healthy baby, and medical advances continue to help secure the fulfillment of this hope. But reprogenetics, a new combination of technology and science that allows us to choose the genes, and thus the traits, of the children we create, is raising new questions about what it means to have a healthy baby.

Genetic Evidence For Long-Term Population Decline In A Savannah Dwelling Primate: Inferences From A Hierarchical Bayesian Model, Jay F. Storz, Mark A. Beaumont, Susan C. Alberts Jul 2002

Genetic Evidence For Long-Term Population Decline In A Savannah Dwelling Primate: Inferences From A Hierarchical Bayesian Model, Jay F. Storz, Mark A. Beaumont, Susan C. Alberts

Jay F. Storz Publications

The purpose of this study was to test for evidence that savannah baboons (Papio cynocephalus) underwent a population expansion in concert with a hypothesized expansion of African human and chimpanzee populations during the late Pleistocene. The rationale is that any type of environmental event sufficient to cause simultaneous population expansions in African humans and chimpanzees would also be expected to affect other codistributed mammals. To test for genetic evidence of population expansion or contraction, we performed a coalescent analysis of multilocus microsatellite data using a hierarchical Bayesian model. Markov chain Monte Carlo (MCMC) simulations were used to estimate …

Method For Analyzing Molecular Expression Or Function In An Intact Single Cell, Philip W. Landfield, Olivier Thibault, Eric M. Blalock, Kuey-Chu Chen, Patrick Kaminker Jul 2002

Method For Analyzing Molecular Expression Or Function In An Intact Single Cell, Philip W. Landfield, Olivier Thibault, Eric M. Blalock, Kuey-Chu Chen, Patrick Kaminker

Pharmacology and Nutritional Sciences Faculty Patents

A method for obtaining molecules from a substantially intact single cell is disclosed. The method differs from prior methods that obtain a variable fraction of a cell's contents and therefore cannot quantitatively estimate the number of molecules in the cells. The present method comprises isolating and harvesting a substantially intact single cell from its organ tissue comprising the steps of subjecting a tissue mass to a dissociation method so that the cells are dissociated from the tissue to expose cell bodies or cell processes, contacting a dissociated cell with a device capable of collecting the cell from the tissue substantially …

Regulation Of Flagellar Dynein By Calcium And A Role For An Axonemal Calmodulin And Calmodulin-Dependent Kinase, Elizabeth F. Smith Jul 2002

Regulation Of Flagellar Dynein By Calcium And A Role For An Axonemal Calmodulin And Calmodulin-Dependent Kinase, Elizabeth F. Smith

Dartmouth Scholarship

Ciliary and flagellar motility is regulated by changes in intraflagellar calcium. However, the molecular mechanism by which calcium controls motility is unknown. We tested the hypothesis that calcium regulates motility by controlling dynein-driven microtubule sliding and that the central pair and radial spokes are involved in this regulation. We isolated axonemes from Chlamydomonasmutants and measured microtubule sliding velocity in buffers containing 1 mM ATP and various concentrations of calcium. In buffers with pCa > 8, microtubule sliding velocity in axonemes lacking the central apparatus (pf18 and pf15) was reduced compared with that of wild-type axonemes. In contrast, at …

D1 Dopamine Receptor Regulation Of Microtubule-Associated Protein-2 Phosphorylation In Developing Cerebral Cortical Neurons, Z.-M. Song, Ashiwel S. Undieh, Y.-Y. Fang, L. Zhang, S. Dracheva, S.C. Sealfon, M.S. Lidow

Publications and Research

This study addresses the hypothesis that the previously described capacity of D1 dopamine receptors (D1Rs) to regulate dendritic growth in developing cortical neurons may involve alterations in the phosphorylation state of microtubuleassociated protein-2 (MAP2). The changes in phosphorylation of this protein are known to affect its ability to stabilize the dendritic cytoskeleton. The study involved two systems: primary cultures of mouse cortical neurons grown in the presence of the D1R agonists, SKF82958 or A77636, and the cortex of neonatal transgenic mice overexpressing the D1A subtype of D1R. In both models, a decrease in dendritic extension corresponded with an elevation in …
