Leptin And The Control Of Body Weight And Metabolism: Role For Stearoyl Coa Desaturase-1 And Other Leptin-Regulated Genes In The Liver, Paul Cohen

Leptin is an adipocyte-derived hormone that regulates energy balance, metabolism, and the neuroendocrine response to altered nutrition. Mice lacking leptin (ob/ob) or its receptor (db/db) are massively obese and hyperphagic. They also have increased triglyceride deposits in multiple peripheral organs, including the liver, massively enlarged and steatotic. Leptin elicits a metabolic response that cannot be explained by its anorectic effects alone. This response is characterized by weight loss, depletion of lipid from peripheral tissues, and enhanced insulin sensitivity. The mechanisms underlying these actions were largely unknown. Nova-1, an autoantigen in paraneoplastic opsoclonus myoclonus ataxia (POMA), a disorder associated with gynecologic cancer and CNS motor dysfunction, is a neuronspecific nuclear RNA binding protein. The work presented here confirms that Nova-1 acts as a regulator of alternative splicing in vivo. In addition to the known target Nova-dependent splicing regulation, exon 3A of glycine receptor α2 subunit (GlyRα2), two new targets are identified, which are the alternative exon, E9 in the inhibitory neurotransmitter receptor subunit, GABAARγ2, and the alternatively spliced exon H in Nova-1's own message. Nova acts to enhance alternative exon inclusion in GlyRα2 and GABAARγ2 mRNAs and …

Zipro 1 is a zinc finger transcription factor that has a restricted pattern of expression in several tissues. The main cells expressing Zipro1 are proliferating cells from epidermal lineages. This includes the cerebellar granule cells (GCs), nasal epithelia and dermal papilla. Through gene dosage experiments we have shown that Zipro1 is involved in proliferation GCs, Hair follicles and the Cranio-facial process. Overexpression of Zipro1 results an increase in the number of GCs, this increase in GCs directly regulates the appearance of secondary fissure in the cerebellum. Protein engineering can be made far more powerful if a protein is not only expressed recombinantly but also altered covalently using synthetic chemistry. These two methods are brought together in the protein semi-synthesis technique Expressed Protein Ligation (EPL). In EPL, recombinant and synthetic polypeptides are joined together via a chemoselective ligation reaction. EPL was originally used to attach synthetic constructs to the C-terminus of recombinant proteins, but is now used to attach recombinant or synthetic polypeptides either at the N- or C-terminus of a protein or into the core of a protein. The toothless (tl) mutation in the rat is a naturally occurring, autosomal recessive mutation resulting in a profound deficiency of bone-resorbing osteoclasts and peritoneal macrophages. The failure to resorb bone produces severe, unrelenting osteopetrosis, with a highly sclerotic skeleton, lack of marrow spaces, failure of tooth eruption, and other pathologies. Injections of CSF-1 improve some, but not all, of these. In this report we have used polymorphism mapping, sequencing, and expression studies to identify the genetic lesion in the tl rat. We found a 10-base insertion near the beginning of the open reading of the Csf1 gene that yields a …

Digital color was introduced in the marketplace almost a decade ago. Both Indigo and Xeikon introduced key new products, and early projections were that these technologies would take off. Initially, as with a number of new technologies, there were technical issues. Presses were unreliable; ink and toner didn’t stick to the paper; and the cost of consumables was too high to generate any substantial application transfer from offset technology. D-fenfluramine (d-FEN) was once widely prescribed and was among the most effective weight loss drugs, but was withdrawn from clinical use because of reports of cardiac complications in a subset of patients. Discerning the neurobiology underlying the anorexic action of d-FEN may facilitate the development of new drugs to prevent and treat obesity. Through a combination of functional neuroanatomy, feeding, and electrophysiology studies in rodents, we show that d-FEN-induced anorexia requires activation of central nervous system melanocortin pathways. These results provide a mechanistic explanation of d-FEN's anorexic actions and indicate that drugs targeting these downstream melanocortin pathways may prove to …

Prey species show diverse antipredator responses to chemical cues signaling predation threat. Among terrestrial vertebrates, the red-backed salamander, Plethodon cinereus, is an important species in the study of these chemical defenses. During the day and early evening, this species avoids rinses from garter snakes, Thamnophis sirtalis, independent of snake diet, but late at night, avoids only those rinses from garter snakes that have recently eaten P. cinereus. We tested whether the selective, late-night response requires the ingestion or injury of salamanders. Injured prey often release alarm chemicals that induce antipredator behaviors in conspecifics. Injured or killed prey most likely release a wide array of chemicals in addition to alarm substances, such as sexual pheromones, which could enhance or compromise antipredator responses. Thus, damage-release cues provide an excellent opportunity to examine the influence of seasonally fluctuating sexual pheromones on antipredator behaviors. Responses to alarm chemicals from injured prey may influence predation risk and foraging success of receivers and senders, while learning can influence the strength of these responses. Thus, it is important to know when in ontogeny prey produce and detect alarm substances and how learning shapes their response, but surprisingly little is known about either of these topics. We assessed when in the life of red-spotted newts, Notophthalmus viridescens, alarm chemicals are produced and detected by comparing adult versus eft (terrestrial juveniles) and larval responses to rinses and tissue extracts from individuals in each life-history stage. IPC Synthase has been shown to be an essential membrane bound enzyme in yeast that catalyzes the transfer of PI to ceramide to yield the primary sphingolipid found in yeast, Inositolphosphorylceramide. A biochemical approach was taken to investigate the biological factors that regulate IC synthase activity in order to better understand the sphingolipid synthesis. A highly purified IPC synthase was purified over 1000-fold from the yeast Saccharomyces cerevisiae by a procedure that included Triton X-100 solubilization, as well as a series of ion exchange and hydrophobic column chromatography steps. Cytochrome P450 3A enzymes (CYP3A) are involved in the metabolism of two thirds of drugs and other xenobiotics, and highly induced by structurally diverse xenobiotics. Induction of CYP3A causes many drug-drug interactions, and an orphan nuclear receptor, the pregnane X receptor (PXR) is established to mediate the induction of CYP3A in mouse and human. Structurally, the PXR has a N-terminal uncharacterized domain, a conserved DNA binding domain (DBD) and a diverse C-terminal ligand binding domain (LBD). The pharmaceutical industry in Egypt is the largest in the Middle East and North Africa (“MENA”) region, and one that has attracted foreign investment despite the fact that Egyptian law has not always provided pharmaceuticals with patent protection. The situation changed with the signing of the Agreement on Trade-Related Aspects of Intellectual Property Rights (“TRIPS agreement” or “TRIPS”), strengthening the interest of the U.S. and other pharmaceutical companies in increasing their investment in Egypt. As more and more nations prosecute copyright piracy cases, it is far from clear whether these nations, in seeking to protect legitimate copyright interests, will also recognize the need to achieve three goals in the sentencing of such cases. The first is honesty in sentencing: that is, avoiding situations in which the nominal sentence that a court initially imposes at sentencing may later be substantially reduced through the parole process. The second is reasonable uniformity in sentencing, so that courts do not have wide disparities in the sentences they impose on similar offenders who commit similar criminal offenses. The third …
