Primers were used to amplify a 561-bp region of the 16S rRNA gene of Ehrlichia phagocytophila from Ixodes scapularis ticks and small mammals collected in Rhode Island and Connecticut. DNA sequences for all 50 E. phagocytophila-positive samples collected from 1996 through 1998 in southwestern Connecticut were identical to the sequence reported for E. phagocytophila DNA from confirmed human cases. In contrast, the sequences from 92 of 123 E. phagocytophila-positive Rhode Island samples collected from 1996 through 1999 included several variants differing by 1-2 nucleotides from that in the agent infecting humans. The Tabanidae of Rhode Island were surveyed using Rhode Island canopy traps placed at 20 locations in the state during the summers of 1999 and 2000. In total, 5,120 flies were collected, which included 55 species in the genera Chrysops, Hybomitra, Tabanus, Merycomyia, and Stonemyia. Distributional and ecological information is provided for each species in Rhode Island.

We provide new information from archaeological samples on the historical freshwater mussel fauna (Mollusca: Bivalvia: Unionoidea) of the Cuyahoga River (South Park site: occupied between ca. A.D. 950 and 1650) and Black River (White Fort site, occupation centered at ca. A.D. 1350), northeast Ohio. Data from these prehi storic sites are compared with information on extant mussel populations of the Cuyahoga River published between 1890 and 2000. The high representation at both archaeological sites of the species Actinonaias ligamentina, Elliptio dilatata, and Ptychobranchus fasciolaris suggests that these were· among the important clean water species in northeast Ohio prior to European …

Many difficulties still exist in translating innovation theories into action in the tourism industry. A substantial literature base is now in place addressing innovation typologies and methodologies and assessing organisational structures, cultures and sizes. Despite this, transferring leading-edge research on organisational innovation and creativity into pragmatic strategies for tourism enterprises has not been fully successful. The most cited and obvious form of innovation is product innovation. It is on this basis that the success of organisations is often judged. Deletion of the nef gene from simian immunodeficiency virus (SIV) strain SIVmac239 yields a virus that undergoes attenuated growth in rhesus macaques and offers substantial protection against a subsequent challenge with some SIV wild-type viruses. We used a recently described model to identify sites in which the SIVΔnef vaccine strain replicates and elicits immunity in vivo. A high dose of SIVΔnef was applied to the palatine and lingual tonsils, where it replicated vigorously in this portal of entry at 7 days. The immune system generally avoids horror autotoxicus or autoimmunity, an attack against the body's own constituents. This avoidance requires that self-reactive T cells be actively silenced or tolerized. We propose that dendritic cells (DCs) play a critical role in establishing tolerance, especially in the periphery, after functioning T cells have been produced in the thymus. In the steady state, meaning in the absence of acute infection and inflammation, DCs are in an immature state and not fully differentiated to carry out their known roles as inducers of immunity. Peripheral immune tolerance is believed to be induced by the processing and presentation of self-tissues that die during physiologic tissue turnover. To examine the mechanism that mediates tolerance, we injected mice with dying syngeneic TAP-/- splenocytes loaded with small amounts of the protein antigen, ovalbumin (OVA). After ingestion and presentation of cell-associated OVA by the CD8+ subset of dendritic cells in situ, large numbers of antigen-reactive, CD8+ T cell receptor (TCR) transgenic T lymphocytes were driven into cell cycle, but then the T cells were deleted. Dendritic cells (DCs) are able in tissue culture to phagocytose and present antigens derived from infected, malignant, and allogeneic cells. Here we show directly that DCs in situ take up these types of cells after fluorescent labeling with carboxyfluorescein succinimidyl ester (CFSE) and injection into mice. The injected cells include syngeneic splenocytes and tumor cell lines, induced to undergo apoptosis ex vivo by exposure to osmotic shock, and allogeneic B cells killed by NK cells in situ. Immature dendritic cells (DCs), unlike mature DCs, require the viral determinant nef to drive immunodeficiency virus (SIV and HIV) replication in coculture with CD4+ T cells. Since immature DCs may capture and get infected by virus during mucosal transmission, we hypothesized that Nef associated with the virus or produced during early replication might modulate DCs to augment virus dissemination. Adenovirus vectors expressing nef were used to introduce nef into DCs in the absence of other immunodeficiency virus determinants to examine Nef-induced changes that might activate immature DCs to acquire properties of mature DCs and drive virus replication. Information on the establishment of immunodeficiency virus infection through transmission of infected cells is sparse. Dendritic cells (DCs) and T cells may be central to the onset and subsequent spread of infection following mucosal exposure.
