Production Of Monoclonal Antibody Specific For Human Hemoglobin, Selma Öztürk, Beyazit Çirakoğlu Jan 2001

Production Of Monoclonal Antibody Specific For Human Hemoglobin, Selma Öztürk, Beyazit Çirakoğlu

Turkish Journal of Medical Sciences

Hemoglobin is not normally present in sputum, urine or feces except in minute amounts. Demonstration of the presence of hemoglobin in these materials is an important diagnostic clue that usually implies bleeding within the respiratory, urinary or gastroentestinal tract, respectively. In this work, a hybridoma line secreting monoclonal antibody (Mab) highly specific for human hemoglobin A (hHbA) was generated by fusion of immunized BALB/c mouse splenic lymphocytes with non-secreting mouse myeloma cells followed by selection in hypoxanthine-aminopterin-thymidine (HAT) medium and the screening of culture fluids for hHb A affinity in ELISA. The Mab produced by MAM 2/3B6 is highly specific …

Flow Cytometric Analysis Of The Effects Of Methotrexate And Vepesid On The Hep-2 Cell Cycle, Ni̇zami̇ Duran, Adi̇l M. Allahverdi̇yev, Sali̇h Çeti̇ner Jan 2001

Flow Cytometric Analysis Of The Effects Of Methotrexate And Vepesid On The Hep-2 Cell Cycle, Ni̇zami̇ Duran, Adi̇l M. Allahverdi̇yev, Sali̇h Çeti̇ner

Turkish Journal of Medical Sciences

Objective: To determine the mechanism of action of Methotrexate and Vepesid on the HEp-2 cells isolated from human laryngeal cancer cells morphologically and flow cytometrically (G1, G2, S and PI). Materials and Methods: The HEp-2 continuous cell line was used. Cultivation of the cells was realized in EMEM medium with 10% fetal bovine serum at an atmosphere of 37ºC with 5% CO_2. Six different concentrations of Vepesid and Methotrexate were prepared by diluting with deionized water (5 µg/ml, 50 mg/ml, 500 µg/ml). The morphological and cell cycle parameters of HEp-2 cells were determined by inverted microscope and flow cytometer respectively. …

In Vitro Mic Determination Of Haemophilus Influenzae Strains Using E-Test, Ni̇lay Çöplü, Orhan Cem Aktepe, Selda Uluutku, Engi̇n Güvener Jan 2001

In Vitro Mic Determination Of Haemophilus Influenzae Strains Using E-Test, Ni̇lay Çöplü, Orhan Cem Aktepe, Selda Uluutku, Engi̇n Güvener

Turkish Journal of Medical Sciences

H. influenzae is a fastidious bacteria, so the selection of a reliable method for in-vitro susceptibility tests is particularly important. The E-test is a practical method for detecting in-vitro antimicrobial susceptibility of antibiotics by MIC values. This study was performed using the E-test for direct patient management and in order to obtain epidemiological data on H. influenzae. The detection of antimicrobial agent resistance among fifty-seven H. influenzae isolates from lower respiratory tract infections in childhood was studied by the E-test. The E-test was performed on HTM agar according to the instruction manual recommendations. The susceptibility ratios and the MIC90 values …

Effects Of Experimental Diabetes And Insulin Treatment On Rabbit Renal Morphology: A Quantitative And Qualitative Study, Hüseyi̇n Eray Bulut, Bi̇lge Onarlioğlu, Celal Kaloğlu, Öztürk Özdemi̇r, Semi̇h Ayan Jan 2001

Effects Of Experimental Diabetes And Insulin Treatment On Rabbit Renal Morphology: A Quantitative And Qualitative Study, Hüseyi̇n Eray Bulut, Bi̇lge Onarlioğlu, Celal Kaloğlu, Öztürk Özdemi̇r, Semi̇h Ayan

Turkish Journal of Medical Sciences

Diabetes causes metabolic defects and related vascular alterations such as atherosclerosis. These vascular alterations may also reflect the renal structural degeneration under the influence of diabetes. Therefore, the aim of the present study was to determine qualitatively and quantitatively the effect of experimental diabetes on rabbit renal cortical structure, and also to evaluate the reversal rate of the renal structural alterations by daily insulin treatment. Eighteen rabbits, bred and fed in standard laboratory conditions, were divided into 3 groups each containing 6 animals. While control group animals received no treatment, the rest were injected with a single dose of intravenous …

Analysis Of Ras Gene Mutation In Human Oral Tumours By Polymerase Chain Reaction And Direct Sequencing, Si̇nan Süzen, James M. Parry Jan 2001

Analysis Of Ras Gene Mutation In Human Oral Tumours By Polymerase Chain Reaction And Direct Sequencing, Si̇nan Süzen, James M. Parry

Turkish Journal of Medical Sciences

Genetic alterations in proto-oncogenes or tumour suppressor genes are believed to be one of the key events in the multistage process of carcinogenesis. Activating point mutations occurring in either one of the three ras proto-oncogene families are common genetic alterations in human and animal neoplasms. However, the mechanisms leading to oral cancer are not completely understood. Activation of the ras oncogene in oral carcinogenesis, although absent or rare in the western world, accounts for up to 35% of all malignancies in India and South Asia. Recognised aetiological agents of oral cancer include tobacco and alcohol. Tobacco-associated compounds such as nitrosamines …

Cytomorphologic Analysis Of Papanicolaou Stained Smears Collected From Floor Of The Mouth Mucosa In Patients With Or Without Oral Malignancy, Nur Mollaoğlu, Jonathan George Cowpe, Rita Walker Jan 2001

Cytomorphologic Analysis Of Papanicolaou Stained Smears Collected From Floor Of The Mouth Mucosa In Patients With Or Without Oral Malignancy, Nur Mollaoğlu, Jonathan George Cowpe, Rita Walker

Turkish Journal of Medical Sciences

Cytology has been found to be a reliable and accurate technique when used to diagnose early malignant transformation in areas of the body where visual examination is difficult or impossible. The factors initiating neoplastic growth are still not clearly understood. Thus, automated instruments, capable of objective and quantitative cell analysis, have been used in descriptive morphology, for the assessment of tumour cell heterogeneity. Smears were collected from a group of 33 patients presenting with squamous cell carcinoma. Nine females and 24 males presenting with squamous cell carcinoma of the floor of the mouth underwent quantitative cytological assessment. These slides were …
