Expression Characteristics Of The Transfer-Related Kilb Gene Product Of Streptomyces Plasmid Pij101: Implications For The Plasmid Spread Function, G. S. Pettis, N. Ward, K. L. Schully

Intermycelial transfer of Streptomyces plasmid pIJ101 occurs prior to cellular differentiation and is mediated by plasmid functions that are also required for production of zones of growth-inhibited recipient cells (i.e., pocks) that develop around individual donors during mating on agar medium. Several other pIJ101 functions, including that of the kilB gene, whose unregulated expression on pIJ101 is lethal, are required for normal pock size and so have been postulated to mediate intramycelial spread of the plasmid throughout recipient cells. Using antibodies raised against a KilB fusion protein expressed in Escherichia coli, native KilB protein was detected throughout development of pIJ101-containing …

Fish Hosts And Culture Of Mussel Species Of Special Concern: Annual Report For 2000, M. Christopher Barnhart

This report describes results of the third year of a 3-year investigation of reproductive biology of freshwater mussels (unionoids). The first two years of this project focused on identification of hosts and on development of methods related to propagation of threatened species. In the third year of the grant we continued propagation efforts with Neosho muckets (Lampsilis rafinesqueana), and began propagation efforts with scaleshell (Leptodea leptodon). We also investigated the effects of chronic exposure to low oxygen on survival of juvenile Lampsilis. We plan to continue propagation of threatened species and investigations of the biology of juvenile mussels over the …

At present a network of over 100 field sites are measuring carbon dioxide, water vapor and sensible heat fluxes between the biosphere and atmosphere, on a nearly continuous basis. Gaps in the long term measurements of evaporation and sensible heat flux must be filled before these data can be used for hydrological and meteorological applications. We adapted methods of gap filling for NEE (net ecosystem exchange of carbon) to energy fluxes and applied them to data sets available from the EUROFLUX and AmeriFlux eddy covariance databases. Heightened awareness of global change issues within both science and political communities has increased interest in using the global network of eddy covariance flux towers to more fully understand the impacts of natural and anthropogenic phenomena on the global carbon balance. Comparisons of net ecosystem exchange (FNEE) responses are being made among biome types, phenology patterns, and stress conditions. The comparisons are usually performed on annual sums of FNEE; however, the average data coverage during a year is only 65%. Therefore, robust and consistent gap filling methods are required.

Tropical South America is one of the three main centers of the global, zonal overturning circulation of the equatorial atmosphere (generally termed the “Walker” circulation). Although this area plays a key role in global climate cycles, little is known about South American climate history. Here we describe sediment cores and down-hole logging results of deep drilling in the Salar de Uyuni, on the Bolivian Altiplano, located in the tropical Andes. We demonstrate that during the past 50,000 years the Altiplano underwent important changes in effective moisture at both orbital (20,000-year) and millennial timescales.
