Apoptosis Following Peripheral Sensory Deafferentation In The Olfactory Blub Of Adult Zebrafish, Danio Rerio, Akiko M. Vankirk

Removal of the olfactory organ in the adult zebrafish results in a significant decrease in volume of the ipsilateral olfactory bulb. Our lab has been investigating the potential role of apoptosis in this phenomenon. My hypothesis is that cells in the adult olfactory bulb normally undergo minimal apoptosis and that apoptosis will increase when sensory stimulation is removed. The TUNEL method allowed detection of cells undergoing DNA-fragmentation, which indicates an apoptotic response. Double-label immunohistochemistry was used to mark which apoptotic cells are neurons by using antibody to Hu, a neuron specific protein. Triple-labeling with bis-benzirnide confirmed the apoptotic nature of …

To determine the number of cavity-containing aspens in old-growth (>80 years), we counted the number of stems containing cavities in 132 0.02-ha plots in Wyoming. There were 8.7 cavities/ha of aspen type. At least 84% of the cavity stems were alive when the initial cavity was constructed; 60% were alive when examined. Fruiting bodies and Phellinus tremulae (a heartrot fungus) were present on 71% of all cavity-bearing stems but on only 9.6% of all stems >15 cm d.b.h. Cavities were present in 7.7% and 0.2% of living stems with and without fruiting bodies, respectively. Average d.b.h. of cavity stems …

Understanding the molecular and biochemical basis of cellular growth and division involves the investigation of regulatory events that most often occur in a cell-cycle phase-dependent fashion. Studies examining cell-cycle regulatory mechanisms and progression invariably require cell-cycle synchronization of cell populations. Thus, many methods have been established to synchronize cells at specific phases of the cell cycle. Several of the common methods involve pharmacological agents, which act at various points throughout the cell cycle. Because of adverse cellular perturbations resulting from many of the synchronizing drugs used, other synchrony methods that involve less perturbation of biological systems, such as serum deprivation, …

Chemokines induce chemotaxis, cell migration, and inflammatory responses. We report the identification of an interleukin-8 (IL-8) homolog, termed vIL-8, encoded within the genome of Marek's disease virus (MDV). The 134-amino-acid vIL-8 shares closest homology to mammalian and avian IL-8, molecules representing the prototype CXC chemokine. The gene for vIL-8 consists of three exons which map to the BamHI-L fragment within the repeats flanking the unique long region of the MDV genome. A 0.7-kb transcript encoding vIL-8 was detected in an n-butyrate-treated, MDV-transformed T-lymphoblastoid cell line, MSB-1. This induction is essentially abolished by cycloheximide and herpesvirus DNA polymerase inhibitor phosphonoacetate, indicating …

Since the implementation of legislation that bans yard waste from landfills, there has been a need to characterize the impact that large scale composting has on groundwater quality. Allen (1993) and Weaver (1995) have attempted to characterize a leaf composting leachate plume located at the WMU-Asylum Lake Well Field #2 in Kalamazoo, Michigan. Allen was not able to base any conclusions on geochemical results and Weaver delineated the plume using fewer monitoring wells than are now available.
The data from twenty-one monitoring wells indicates that local groundwater is being impacted by the large-scale composting operation. The plume is characterized by …

A significant issue surrounding the increasing dependence of U.S. society on technological products surrounds questions of equity. Research suggests that our increased dependence on technology has acted to further stratify our society in regards to gender. A growing body of literature which takes a critical stance of technology acknowledges that there are gaps between technology 'haves' and 'have-nots' (Aronowitz, 1988; Brosnan, 1998; Harvey, 2000; Loader, 1998; Millar, 1998), suggesting that utopian perspectives of technology need to be re-examined. Fibrinogen and fibrin play important, overlapping roles in blood clotting, fibrinolysis, cellular and matrix interactions, inflammation, wound healing, and neoplasia. These events are regulated to a large extent by fibrin formation itself and by complementary interactions between specific binding sites on fibrin(ogen) and extrinsic molecules including proenzymes, clotting factors, enzyme inhibitors, and cell receptors. Fibrinogen is comprised of two sets of three polypeptide chains termed Aα, Bβ, and γ, that are joined by disulfide bridging within the N-terminal E domain. Forest fires are as old as human habitation on Earth. Traditionally, legal systems considered them to be isolated events and the responsibility of the national state within which they took place. Events have proven this legal construct to be inadequate. As Earth's population has grown, human settlements increasingly encroach on forest lands, and natural resource exploitation of forests has intensified. In 1997 and 1998, large-scale forest fires raged on five continents. Satellite sensors measured the fires, as well as the EI Niño induced dry climate conditions that set the stage for the fires. Methods for detecting microorganisms on surfaces are needed to locate biocontamination sources and to relate surface and airborne concentrations. Research was conducted in an experimental room to evaluate surface sampling methods and quantitative PCR (QPCR) for enhanced detection of a target biocontaminant present on flooring materials. QPCR and culture analyses were used to quantitateBacillus subtilis (Bacillus globigii) endospores on vinyl tile, commercial carpet, and new and soiled residential carpet with samples obtained by four surface sampling methods: a swab kit, a sponge swipe, a cotton swab, and a bulk method. The purpose of this investigation was to study the effects of a 17-day spaceflight on the contractile properties of individual fast- and slowtwitch fibers isolated from biopsies of the fast-twitch gastrocnemius muscle of four male astronauts. Single chemically skinned fibers were studied during maximal Ca2+-activated contractions with fiber myosin heavy chain (MHC) isoform expression subsequently determined by SDS gel electrophoresis. Spaceflight had no significant effect on the mean diameter or specific force of single fibers expressing type I, IIa, or IIa/IIx MHC, although a small reduction in average absolute force (Po) was observed for the type …

The purpose of this study was to examine the impact of peer pressure upon eighth, tenth, and twelfth grade use of alcohol and other drugs. Data related to the use of cigarettes, alcohol, and marijuana were analyzed to determine the effect that peer pressure has on the use of these substances. Other relationships considered were sex, grade level, and race, based on the literature in this area.
The data that were chosen for this study included 141 Michigan public school districts and were based on the responses of 16,760 students in 1992-93, and 15,283 students in 1994-95.

This thesis explores the social implications involved with the technological decision to use grog (crushed potsherds) as a ceramic tempering agent by potters affiliated with the Plum Bayou culture of central Arkansas. The analytical technique of point-counting ceramic thin sections is used to search for patterns of grog-temper use at a single Plum Bayou culture site, the Ink Bayou site (3PU252). While the thermal properties of grog-temper may help to explain the variability of use observed at the Ink Bayou site, the social implications of producing grog-tempered pots are best illuminated by the sequence of productive operations employed by the …

Lepidopteran insects present a complex organization of appendages which develop by various mechanisms. In the mulberry silkworm, Bombyx mori, a pair of meso- and meta-thoracic discs located on either side in the larvae gives rise to the corresponding fore- and hind-wings of the adult. These discs do not experience massive cell rearrangements during metamorphosis and display the adult wing vein pattern. We have analysed wing development in B. mori by two approaches, viz., expression of patterning genes in larval wing discs, and regulatory capacities of larval discs following explantation or perturbation. The youth-based, patriarchal culture in which we live bestows valuable status, power and prestige upon young, beautiful women. Within the standards of this society, the natural aging process progressively deteriorates a woman's beauty, and, therefore, her social worth. A vast network of media and societal messages prompt middle-aged women to react to this devaluation. Several pathways or behavioral alternatives are available as reactions, adaptive reconstructions, and/or the medicalization of the aging process in women. The goal of this sociological/gerontological exploratory research is to identify, examine, label, and categorize the multiple options available to midlife women.
