Pcr As A Diagnostic And Quantitative Technique In Veterinary Parasitology, Dante S. Zarlenga, James Higgins Jan 2001

Pcr As A Diagnostic And Quantitative Technique In Veterinary Parasitology, Dante S. Zarlenga, James Higgins

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Over the past 15 years, there has been a dramatic evolution in molecular approaches to study parasites and parasitic diseases. Many of these advancements have been brought about through the development of new applications of the polymerase chain reaction (PCR). Enhancements in sensitivity that can be achieved using PCR now permit scientists to investigate changes at the level of a single cell, far below what is often needed for parasite-derived applications. PCR has had a substantial impact on advances made in the areas of parasite systematics and epidemiology, immunology and host–parasite interactions, recombinant DNA vaccine development and most re- cently, …

Methyl-Β-Cyclodextrin: An Alternative Carrier For Intravenous Infusion Of Palmitate During Tracer Studies In Swine (Sus Scrofa Domestica), Diane Wray-Cahen, Thomas J. Caperna, Norman C. Steele Jan 2001

Methyl-Β-Cyclodextrin: An Alternative Carrier For Intravenous Infusion Of Palmitate During Tracer Studies In Swine (Sus Scrofa Domestica), Diane Wray-Cahen, Thomas J. Caperna, Norman C. Steele

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Fatty acid-free albumin has been the standard carrier for intravenous infusion of fatty acids to study in vivo lipid metabolism. However, subjects can have adverse reactions to infusion of albumin. We sought an alternative to albumin as a carrier for intravenous infusion of fatty acids, using the pig as a model. Cyclodextrins are naturally occurring water-soluble molecules that can serve as carriers for lipid-soluble compounds. 13C-palmitate was complexed to either 20% methyl-β-cyclodextrin, 20% 2-hydroxypropyl-β-cyclodextrin, or 5% porcine albumin (isotopic purity of infusates: 99.22±0.06%). 13C-palmitate-albumin was infused under fed conditions and 13C-palmitate-methyl-β-cyclodextrin was infused under fasted and fed …

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Bemisia tabaci was described over 100 years ago and has since become one of the most important pests worldwide in subtropical and tropical agriculture as well as in greenhouse production systems. It adapts easily to new host plants and geographical regions and has now been reported from all global continents except Antarctica. In the last decade, international transport of plant material and people have contributed to geographical spread. B. tabaci has been recorded from more than 600 plant species and there may be many additional hosts not yet formally documented. Biotypes have been identified in different areas of the world …

Food Webs, Risks Of Alien Enemies And Reform Of Biological Control, D. R. Strong, R.W. Pemberton Jan 2001

Food Webs, Risks Of Alien Enemies And Reform Of Biological Control, D. R. Strong, R.W. Pemberton

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

As ilwasions of alien species mount, biological control will become an increasingly important tool of conservation and agriculture. In an effort to understand indirect interactions in biological control, we review food web ecology in terms of resource competition, trophic cascades, intraguild predation, apparent competition, omnivory and a diverse set of tritrophic interactions. The most inclusive study suggests that food webs in biological control are simpler than in natural communities. Risks to non-target species created by biological control have been studied seriously for only about 20 years, and knowledge of these risks is incomplete. The greatest risks are known to be …

Innovative 15N Microplot Research Techniques To Study Nitrogen Use Efficiency Under Different Ecosystems, Ronald F. Follett

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Use of labeled 15N techniques can help improve and refine knowledge required to understand the dynamics of the N cycle under agricultural systems. The examples reported in this paper illustrate that, with creativity and careful planning, use of 15N-enriched or depleted fertilizers can be used in many types of N experiments with various crops, cropping systems, and in many types of agroecosystems. The objective is to report examples of a wide range of field studies that researchers can hopefully draw upon to design and utilize N isotope techniques that accomplish the objectives for their own research. Examples of …

Uniform Random Number Generation Using Pseudorandom Binary Sequences, Susan Rea Jan 2001

Uniform Random Number Generation Using Pseudorandom Binary Sequences, Susan Rea

Theses

Random numbers have conventionally been used in a range of applications such as numerical analysis, lotteries and computer simulation and as cryptographic keys. The algorithms used for the production of random numbers are termed pseudorandom, as the numbers produced are not truly random as they emanate from a deterministic periodic source that is they are predictable if a subset of the sequence is known. However, for most applications these pseudorandom number sources are entirely suitable.

For cryptographic key sources, however, the random number sequences must be statistically good and difficult to predict. Linear feedback shift registers are presented in this …

Backdoor Liability From Internet Telecommuters, Mark J. Maier Jan 2001

Backdoor Liability From Internet Telecommuters, Mark J. Maier

Computer Law Review and Technology Journal

Internet telecommuters work from home and access their employer's internal network over the Internet. This relatively new style of work has increased in popularity through the years, with an estimated 4.5 million workers Internet telecommuting in 1999. While employers enjoy compelling benefits such as reducing overhead costs, Internet telecommuting raises new security concerns for employers by exposing their internal networks to "backdoor attacks" that exploit the telecommuter's connection. The recent backdoor attack on Microsoft's source code illustrates the extent to which such attacks threaten confidential information. If such confidential information belongs to a client or third party, then the telecommuter's …

Performing Arts Manuscript Collections: Balancing Access And Privacy, Linda B. Fairtile Jan 2001

Performing Arts Manuscript Collections: Balancing Access And Privacy, Linda B. Fairtile

University Libraries Faculty and Staff Publications

This essay examines some of the issues involved in administering performing arts manuscript collections. After briefly discussing the evolving notion of privacy in both its legal and moral senses, it will turn to the relationships and interlocking responsibilities of the four groups concerned with access to manuscript collections: donors, custodians, users, and "third-party" contributors. Finally, the results of a survey of performing arts repositories will reveal the variety of ways in which these responsibilities are addressed. It will be demonstrated that despite the attention paid to issues of access by professional organizations, agreements in theory, much less in practice, have …

The Investigation Of Rna Viruses Using The Atomic Force Microscope, Michelle Moloney Jan 2001

The Investigation Of Rna Viruses Using The Atomic Force Microscope, Michelle Moloney

Theses

The subject of viral entry into, and subsequent progeny virus egress, from cultured cells, has been extensively studied using numerous scientific techniques in fields ranging from cell culture to biochemistry to microscopy. The atomic force microscope (AFM) is a novel, developing instrument with unprecedented capabilities and this study was undertaken to observe enveloped Semliki Forest virus (SFV) and non-enveloped Theiler's Murine Encephalomyelitis virus (TMEV) entry into and egress from cultured cells using the AFM. Virus particle structure of both enveloped and non-enveloped viruses was also directly examined.

Cytopathic effect studies revealed that TMEV's are unable to produce progeny viruses in …
