Phage Sensitivities Of Lactococci Isolated From Raw Milk And Whey, Pinar Şanlibaba, Mustafa Akçeli̇k

Turkish Journal of Biology

In this study, lactococcal strains isolated from raw milk and whey samples of different types of cheese produced without using industrial starter cultures in different regions of Turkey were tested for sensitivity to 22 lactic phages. In addition to the strain isolation materials, virulent lactic phages were also isolated from samples obtained from dairy plants using industrial starter cultures. 18 L lactis subsp. lactis strains out of 73 lactococcal strains were found to be resistant to all the phages. It was determined that the total phage resistance levels of the other 55 strains were between 18.18 and 90.90%.

Blood samples were taken from agricultural workers who had been exposed to pesticides for long periods in Içel and the surrounding agricultural areas (n=150) and levels of AST, ALT, ALP, LDH, total protein, albumin and globulin in serum were determined. Production Of Gibberellic Acid And Cytokinin By Phanerochaete Chrysosporiumme446 Immobilized On Polyurethane Foam, Serpi̇l Ünyayar

The fungus Phanerochaete chrysosporium ME446 was entrapped on polyurethane foam to synthesize gibberellic acid (GA 3 equivalent) and Cytokinin (zeatin equivalent). Maximum gibberellic acid was defined on the 6 th day (44.43 µg/mL and 61.22 µg/mL, respectively) in both free and immobilized cells. Maximum cytokinin was defined on the 12 th day (9.61µg/mL and 16.33 µg/mL, respectively) in both free and immobilized cells. An immobilized system of gibberellic acid and cytokinin production resulted in comparatively higher yield than free fungal mycelium.

Transcriptional regulation is of considerable importance in the expression of eukaryotic genes, and a considerable degree of regulation is controlled at the level of mRNA synthesis. As in prokaryotic organisms, transcriptional control in eukaryotes results from an interplay between regulatory DNA sequences and site-specific DNA-binding proteins. Transcription of eukaryotic genes is influenced by various regulatory elements, termed promoters, enhancers and silencers. These regulatory elements are composed of discrete DNA sequences, which constitute binding sites for sequence-specific DNA-binding proteins. These proteins bind DNA by some common structural motifs. The effects of different carbon sources (ethanol, methanol and acetic acid) and C/N ratios (0,5; 1; 1,5; 2; 2,5) on the microbial denitrification in batch culture were investigated. For this purpose Paracoccus denitrificans and Pseudomonas stutzeri were used as denitrifiers. For all experiments, the initial NO 3 concentrations were constant at 100 mg/l. During the incubation period of 66 hours, nitrate, nitrite and microorganism concentrations were determined spectrophotometrically at certain intervals. The effects of cadmium nitrate on the fecundity, larval survival and sex ratio of D. melanogaster were tested. According to our findings, the effects of cadmium nitrate can be summarized as follows: it appeared that when used as a feeding substance during the developmental period, cadmium nitrate did not have any effect on fecundity. On the other hand, if cadmium nitrate was given throughout adult life it caused a proportional decrease in the daily mean with increasing concentration. The effects of various concentrations (0.01, 1, 2mM) of spermine (Spm) on germination and some early growth parameters of sunflower ( Helianthus annuus L. cv. Santafe) seeds were investigated in media containing different concentrations of NaCl (50, 100, 200mM). Regardless of its concentrations, application of Spm under non-saline conditions remained ineffective on germination percentage as well as on the early growth parameters studied (length of radicle, fresh and dry weights). Samples of water, bottom clay, plankton and fish were collected over a period of one year. The boron contents of the samples taken were determined in the laboratory. The differences and relationships in the obtained data were identified using various statistical methods. Boron gradually accumulates in the plankton, bottom clay and water of Çatören and Kunduzar. Boron collects in the tissues of the fish Cyprinus carpio in both lakes, in the musce, brain and liver in Çatören, and in the musce, liver and brain in Kunduzlar. Apoptosis induced by hydrogen peroxide (H_2O_2 ) was investigated in Chinese hamster ovarian (CHO) cells, and compared to apoptosis induced by cisplatin (25 µM), a well-established apoptosis-inducing agent. In preliminary experiments, CHO cells were treated with low concentrations of H_2O_2 (250-500 µM) continuously or for 3 hours, followed by washing and further incubation in complete growth medium for 1-10 days. Fluorescence microscopy of acridine orange-stained cells indicated morphological changes of apoptosis. Similar changes were observed in cisplatin-treated cells. We report here the development of hybrid cells producing monoclonal antibodies specific for two different steroid hormones with mixed immunization using hybridoma technology. BALB/c mice were immunized with a mixture of three steroid antigens: progesterone, estradiole and testosterone linked to bovine serum albumine. These mice were used for fusion. In the two fusion experiments, ELISA tests showed that among 645 wells only 2 hybrids reacted with progesterone (MAM 3C2, MAM 3E3) and one of them reacted specifically with estradiol (MAM 7H7). This study was performed to determine the capability of embryonic stem (ES) cells at late passage number to form functional germ cells and to orientate the sexual development of chimeric mice produced from ES cell injection into embryos. When an R1 ES cell line was routinely cultured on mitotically inactivated feeder cells in culture medium supplemented with leukemia inhibitory factor (LIF), the colonies maintained an undifferentiated ideal ES cell morphology. Thermomonospora fuscaBD25 produces relatively high levels of activity of endoxylanases. Only oat spelt xylan, RBB-xylan and birchwood xylan reacted with endoxylanases with the maximum specific activity of 47.65 U mg -1 protein on birchwood xylan. The endoxylanase remained stable up to 70 °C. At 60 °C endoxylanase activity showed stability (92 %) for 16 h, however it showed a half-life of 8 h at 70 °C and 30 min at 80 °C in the absence of substrate at pH 8.0. Thermomonospora fuscaBD25 produces relatively high levels of activity of \beta-xylosidase and \alpha-L-arabinofuranosidase. The aim of the work described in this study was to characterize some properties of \beta-xylosidase and \alpha-L-arabinofuranosidase produced by T. fusca BD25 when growing on oat spelt xylan as the main carbon and energy sources. The substrates p-NPX and p-NPA reacted with \beta-xylosidase and \alpha-L-arabinofuranosidase with specific activities of 4.01 U mg -1 protein and 0.35 U mg -1 protein, respectively. Production Of Auxin And Abscisic Acid Byphanerochaete Chrysosporiumme446 Immobilizedon Polyurethane Foam, Serpi̇l Ünyayar, Eli̇f Ünal, Ali̇ Ünyayar

The fungus Phanerochaete chrysosporiumME446 was entrapped on polyurethane foam to synthesize auxin and abscisic acid (ABA). The maximum amounts of auxin produced on the 18 th day in both free and immobilized cells were 55.91mg/mL and 76.07mg/mL, respectively. Maximum ABA concentrations on the 12 th day in both free and immobilized cells were 6.87 mg/mL and 10.69 mg/mL, respectively. An immobilized system of auxin and ABA production resulted in a comparatively higher yield than free fungal mycelium.

Temporal And Spatial Determination Of Germinbiosynthesis In Wheat Tissues, Mahmut Çalişkan, Andrew C. Cuming

Turkish Journal of Biology

In the current study, the biosynthesis of germin was determined in the organs of wheat embryos during germination. The temporal and spatial determination of germin protein was carried out by employing an anti-germin sera in gel-blot analysis. Germin was detected in the coleoptile, roots and vascular transition region of and 6-day-old germinated wheat seedlings. It was shown that the leaf tissue acquires germin accumulation only at the late stage of germination. The pellet fraction of protein homogenate obtained from embryos at different ages revealed higher germin accumulation than the supernatant fraction from the same sources.

Native-polyacrylamide gel electrophoresis (N-PAGE) was used to characterize Candida albicansstrains isolated from vaginal specimens of patients suffering from vaginitis in Ankara hospitals, Turkey. Three different protein band profiles were observed among these 38 C. albicansisolates when whole cell protein extracts of these strains were used in N-PAGE. 34 of the 38 isolates were shown to belong to a single group or clone with a dissemination percentage of 89, while three isolates belonged to another group with a percentage of 8. Furthermore only one C. albicans isolate was detected with a dissemination percentage of 3 and assigned to a third group. …

The present study was undertaken to determine the relationship between Methicillin-Resistant Staphylococcus aureus (MRSA) strains isolated from various clinical specimens in Gülhane Military Medical Academy, in Ankara, Turkey. In 1997, at this hospital, the percentage of S. aureus among all nosocomial isolates was 20% and the percentage of MRSA among these S. aureus isolates was 51%. Both sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and Native-Page (N-PAGE) were used for typing MRSA isolates which have common antibiotic susceptibility test results. In this study, 18 live specimens collected from six localities in the distribution areas of Mesocricetus brandti and Mesocricetus auratus were examined using SDS-PAGE (sodium dodecyl sulphate-polyacrylamide gel electrophoresis). The findings showed that there is no difference in globulin and albumin proteins of M. brandti and M. auratus, and supported the claim that M. auratus is very closely related to M. brandti.

This study was undertaken to obtain uricase enzyme from Aspergillus niger and also to determine the production conditions and the effects of some factors on enzyme activity. The results of the experiments, in consideration of the production of the uricase enzyme and the factors affecting the production, demonstrated that enzyme activity was maximum when the production time was 3 days at a temperature of 30ºC with the initial pH at 6.0 and inductor concentration at 0.1%. Sulphur dioxide as a criterion of air pollution supresses macrophage functions. Some immunological changes in diabetes mellitus have been detected. In the present study, we examined the effect of sulphur dioxide on macrophage phagocytic activity in diabetic rats. The animals were separated into four groups as: 1) Control, 2) Sulphur dioxide, 3) Diabetes mellitus, 4) Diabetes mellitus+sulphur dioxide. Qualitative seasonal distributions of sulfate-reducing bacteria, colorless sulfur bacteria, nitrifying and denitrifying bacteria in the water and sediment samples in the inner part of İzmir Bay were investigated in relation to certain physico-chemical parameters (pH, oxygen, hydrogen sulfide, nitrate, nitrite, ammonium). Denitrifying bacteria and sulfate-reducing bacteria were present in the sediment and even in the aerobic water samples. Hydrogen sulfide was found almost in all water samples measured but it was at lower concentrations. In Vitro Propagation Of Thymus Sipyleus Boiss. (Lamiaceae), Bengi̇ Baba Erdağ, A. Kami̇l Yürekli̇

In vitro propagation of Thymus sipyleus Boiss. (Lamiaceae), an endemic from western Anatolia was investigated. Sterilized seeds of the plant were germinated on modified Murashige-Skoog medium (mMS) and Heller's nutrient media. Then, seedlings were transferred to Murashige-Skoog basal medium containing 0.4 mg/l Naphtaleneacetic acid, (NAA) and 3 mg/l Benzyladenin (BA) and then they formed callus where they were in contact with the medium. Plantlets which regenerated on the same medium formed multiple shoots within 2 weeks and they were rooted after the 4th subculture.
