Publications and Research

In peripheral nerve myelin, the intraperiod line results from compaction of the extracellular space due to homophilic adhesion between extracellular domains (ECD) of the protein zero (P0) glycoprotein. Point mutations in this region of P0 cause human hereditary demyelinating neuropathies such as Charcot-Marie-Tooth. We describe transgenic mice expressing a full-length P0 modified in the ECD with a myc epitope tag. The presence of the myc sequence caused a dysmyelinating peripheral neuropathy similar to two distinct subtypes of Charcot-Marie-Tooth, with hypomyelination, altered intraperiod lines, and tomacula (thickened myelin). The tagged protein was incorporated into myelin and was associated with the morphological …

Magnetic Clusters On Single-Walled Carbon Nanotubes: The Kondo Effect In A One-Dimensional Host, Teri W. Odom, Jin-Lin Huang, Chin Li Cheung, Charles M. Lieber Nov 2000

Magnetic Clusters On Single-Walled Carbon Nanotubes: The Kondo Effect In A One-Dimensional Host, Teri W. Odom, Jin-Lin Huang, Chin Li Cheung, Charles M. Lieber

Barry Chin Li Cheung Publications

Single-walled carbon nanotubes are ideal systems for investigating fundamental properties and applications of one-dimensional electronic systems. The interaction of magnetic impurities with electrons confined in one dimension has been studied by spatially resolving the local electronic density of states of small cobalt clusters on metallic single-walled nanotubes with a low-temperature scanning tunneling microscope. Spectroscopic measurements performed on and near these clusters exhibit a narrow peak near the Fermi level that has been identified as a Kondo resonance. Using the scanning tunneling microscope to fabricate ultra-small magnetic nanostructures consisting of small cobalt clusters on short nanotube pieces, spectroscopic studies of this …

Interactions Of Tillage And Soil Depth On Fluometuron Degradation In A Dundee Silt Loam Soil, R. M. Zablotowicz, M. A. Locke, L. A. Gaston, C. T. Bryson Nov 2000

Interactions Of Tillage And Soil Depth On Fluometuron Degradation In A Dundee Silt Loam Soil, R. M. Zablotowicz, M. A. Locke, L. A. Gaston, C. T. Bryson

Faculty Publications

Fluometuron (N,N-dimethyl-N'-[3-(trifluoromethyl)phenyl]urea) is the most widely used soil applied herbicide in cotton (Gossypium hirsutum) production in the Southeastern United States. Thus, there is a need to understand the persistence of this herbicide under various crop management systems in regards to both herbicide efficiency and environmental fate. The effects of long-term no-tillage practices on fluometuron degradation were studied in a Dundee silt loam under laboratory conditions. Soil was collected at four depths (0-2, 2-5, 5-10, and 10-25 cm) from field plots following 11 years of continuous no-tillage (NT) or conventional tillage (CT). Organic carbon content, microbial biomass, and fluorescein diacetate (FDA)-hydrolytic …

Molecular Dynamics Calculations Of The Electrochemical Properties Of Electrolyte Systems Between Charged Electrodes, Richard L. Rowley, Paul S. Crozier, Douglas Henderson Nov 2000

Molecular Dynamics Calculations Of The Electrochemical Properties Of Electrolyte Systems Between Charged Electrodes, Richard L. Rowley, Paul S. Crozier, Douglas Henderson

Faculty Publications

We investigate the interfacial electrochemical properties of an aqueous electrolyte solution with discrete water molecules in slab geometry between charged atomistic electrodes. Long-range intermolecular Coulombic interactions are calculated using the particle particle particle mesh method with a modification to account for the slab geometry. Density distribution profiles and potential drops across the double layer are given for 0, 0.25, and 1 M aqueous electrolyte solutions each electrode surface charges. Results are compared qualitatively with experimental x-ray scattering findings, other computer simulation results, and traditional electrochemistry theory. The interfacial fluid structure characteristics are generally in good qualitative agreement with the conclusions …

Dynamics And Folding Of Single Two-Stranded Coiled-Coil Peptides Studied By Fluorescent Energy Transfer Confocal Microscopy, David Talaga, Wai Leung Lau, Heinrich Roder, Jianyong Tang, Yiwei Jia, William F. Degrado, Robin M. Hochstrasser Nov 2000

Dynamics And Folding Of Single Two-Stranded Coiled-Coil Peptides Studied By Fluorescent Energy Transfer Confocal Microscopy, David Talaga, Wai Leung Lau, Heinrich Roder, Jianyong Tang, Yiwei Jia, William F. Degrado, Robin M. Hochstrasser

Department of Chemistry and Biochemistry Faculty Scholarship and Creative Works

We report single-molecule measurements on the folding and unfolding conformational equilibrium distributions and dynamics of a disulfide crosslinked version of the two-stranded coiled coil from GCN4. The peptide has a fluorescent donor and acceptor at the N termini of its two chains and a Cys disulfide near its C terminus. Thus, folding brings the two N termini of the two chains close together, resulting in an enhancement of fluorescent resonant energy transfer. End-to-end distance distributions have thus been characterized under conditions where the peptide is nearly fully folded (0 M urea), unfolded (7.4 M urea), and in dynamic exchange between …

Kinetic Characterization Of His-Tagged Cp47 Photosystem Ii In Synechocystis Sp. Pcc6803, Zhao Liang Li, Terry M. Bricker, Robert Burnap Nov 2000

Kinetic Characterization Of His-Tagged Cp47 Photosystem Ii In Synechocystis Sp. Pcc6803, Zhao Liang Li, Terry M. Bricker, Robert Burnap

Faculty Publications

Recently, construction of strains of Synechocystis sp. PCC6803 having a His6 extension (His-tag) of the carboxyl terminus of the CP47 protein has been reported (T.M. Bricker et al, Biochim. Biophys. Acta 1409 (1998) 50; M.J. Reifler et al., in: Garab, Pusztai (Eds.) Proc. XIth International Congress on Photosynthesis, 1998). While these initial reports suggest a minimal impact of the His-tag upon Photosystem (PS) II function, a more thorough analysis of the kinetic properties of the modified complex is essential. This communication reports on a more detailed kinetic analysis to assess possible perturbations of PS II due to the genetic addition …

Somite Development In Zebrafish, Heather L. Stickney, Michael J.F. Barresi, Stephen H. Devoto Nov 2000

Somite Development In Zebrafish, Heather L. Stickney, Michael J.F. Barresi, Stephen H. Devoto

Biological Sciences: Faculty Publications

A full understanding of somite development requires knowledge of the molecular genetic pathways for cell determination as well as the cellular behaviors that underlie segmentation, somite epithelialization, and somite patterning. The zebrafish has long been recognized as an ideal organism for cellular and histological studies of somite patterning. In recent years, genetics has proven to be a very powerful complementary approach to these embryological studies, as genetic screens for zebrafish mutants defective in somitogenesis have identified over 50 genes that are necessary for normal somite development. Zebrafish is thus an ideal system in which to analyze the role of specific …
