Ec00-1207 Growing Onions, Shallots, And Chives, Susan Schoneweis, Laurie Hodges, Loren J. Giesler

University of Nebraska-Lincoln Extension: Historical Materials

The common onion (Allium cepa) is the most popular and widely grown Allium in Nebraska home gardens. Many members of the onion family (Alliaceae) are used in flower gardens because of their interesting flowers and foliage, but here we're going to focus on the onion as a vegetable crop.

This publication will discuss the different types of onions and how to grow onions from onion sets, plants, transplants, and seed. It will cover the different cultivars found here in Nebraska, the nutrition, fertilizing, harvesting, curing and storing of onions.

Season And Distance From Forest - Old Field Edge Affect Seed Predation By White-Footed Mice, Scott J. Meiners, James T. Mccormick Jan 2000

Season And Distance From Forest - Old Field Edge Affect Seed Predation By White-Footed Mice, Scott J. Meiners, James T. Mccormick

Faculty Research & Creative Activity

We studied the spatial pattern of seed predation across a forest-old field edge in both fall and winter to assess the potential for seed predators to influence plant spatial patterns. We used a 100 x 100 m grid that began 30 m inside the forest and extended 60 m into the old field. Inside this grid we placed seed stations at regular 10 m intervals and monitored seed removal. Seed predation varied significantly across the edge gradient in both fall and winter with the highest rate of seed removal at the edge in both trials. The spatial pattern of seed …

Metals And Metallothionein In The Liver Of Raccoons: Utility For Environmental Assessment And Monitoring, Joanna Burger, Christine G. Lord, Edward J. Yurkow, Lynn Mcgrath, Karen F. Gaines, I. Lehr Brisbin Jr., Michael Gochfeld Jan 2000

Metals And Metallothionein In The Liver Of Raccoons: Utility For Environmental Assessment And Monitoring, Joanna Burger, Christine G. Lord, Edward J. Yurkow, Lynn Mcgrath, Karen F. Gaines, I. Lehr Brisbin Jr., Michael Gochfeld

Faculty Research & Creative Activity

The relationship between metallothionein levels and concentrations of several metals and radionuclides was examined in liver tissues of raccoons (Procyon lotor, n = 47) from the Department of Energy’s Savannah River Site in South Carolina to determine the applicability of metallothioneins as an initial screening device for exposure assessment in free-living mammals and environmental monitoring. Using a fluorescent marker and a cell sorter to measure metallothionein, a significant positive correlation was found across animals between levels of metallothioneins and concentrations of selenium (Pearson’s r = .30) , mercury (Pearson’s r = .31) , and copper (Pearson’s r = .30) in …

Proximate Constraints On Intruder Detection In The Dragonfly Perithemis Tenera (Odonata: Libellulidae): Effects Of Angle Of Approach And Background, Paul V. Switzer, Perri K. Eason Jan 2000

Proximate Constraints On Intruder Detection In The Dragonfly Perithemis Tenera (Odonata: Libellulidae): Effects Of Angle Of Approach And Background, Paul V. Switzer, Perri K. Eason

Faculty Research & Creative Activity

The implications of insects’ vision for territorial defense have been relatively little studied in the field. In the dragonfly Perithemis tenera Say we investigated whether either the angle at which an intruder was viewed by a territorial resident or the background against which it was viewed affected the detection of that intruder. Residents detected intruders at a greater distance if the intruders were directly in front of them; they also detected more intruders in front of them than from other angles. Intruders viewed against distant vegetation were detected more readily than were intruders against near vegetation. Residents detected more intruders …

Genetic Diversity And Transcriptional Analysis Of The Bys1 Gene From Blastomyces Dermatitidis, James L. Bono, Basam Jaber, Mark A. Fisher, Raed O. Abuodeh, Erin O’Leary-Jepson, Gene M. Scalarone, Laurens H. Smith Jr.

Roman L. Hruska U.S. Meat Animal Research Center: Reports

Blastomyces dermatitidis, a pathogenic fungal organism, is able to exist in two differentmorphologies, a multicellular mycelium or a unicellular yeast, according to temperature, 25˚C and 37˚C respectively. The switching between morphologies must be accompanied by a cascade of signaling events in which expression of genes responsible for the change of morphology is increased or decreased. bys1, a gene from B. dermatitidis isolate #58, is expressed at high levels in the unicellular yeast, but gradually diminishes as the temperature is lowered and the organism converts to the mycelial phase where there is no transcription of bys1. We explored …

Effect Of Antimicrobial Agents On Livestock Waste Emissions, Vincent H. Varel, Daniel N. Miller Jan 2000

Effect Of Antimicrobial Agents On Livestock Waste Emissions, Vincent H. Varel, Daniel N. Miller

Roman L. Hruska U.S. Meat Animal Research Center: Reports

Various antimicrobial agents were evaluated with the purpose of reducing the microbial fermentation in stored cattle waste and the resulting odor emissions. Duplicate sealed 2-L flasks with 500 ml waste slurry, with and without antimicrobial inhibitors, were used to measure the production of short-chain volatile fatty acids, lactate, and total fermentation gas over 27–30 days. A combination of chlorhexidine diacetate (2 mM), iodoacetate (2 mM), and α-pinene (3.8 mM) reduced gas production 80% (1000 ml to 200 ml) and total volatile fatty acid production 50% (145 mM to 72 mM). Pinene had little antimicrobial effect; rather, it served as an …

Mapping Quantitative Trait Loci For Bovine Ovulation Rate, Brian W. Kirkpatrick, Becky M. Byla, Keith E. Gregory Jan 2000

Mapping Quantitative Trait Loci For Bovine Ovulation Rate, Brian W. Kirkpatrick, Becky M. Byla, Keith E. Gregory

Roman L. Hruska U.S. Meat Animal Research Center: Reports

An elite, three-generation family from the USDA Meat Animal Research Center twinning population was examined for evidence of ovulation rate quantitative trait loci (QTL). This work was both a continuation of previously reported results suggesting evidence for ovulation rate QTL on bovine Chromosome (Chr) 7 and an extension of a genome-wide search for QTL. Additional markers were typed on Chr 7 to facilitate interval mapping and testing of the hypothesis of one versus two QTL on that chromosome. In addition, 14 other informative markers were added to a selective genotyping genome screening of this family, and markers exhibiting nominal significance …

Comparative Map Alignment Of Bta27 And Hsa4 And 8 To Identify Conserved Segments Of Genome Containing Fat Deposition Qtl, Tad S. Sonstegard, Wes M. Garrett, Melissa S. Ashwell, Gary L. Bennett, Steven M. Kappes, Curtis P. Van Tassell

Roman L. Hruska U.S. Meat Animal Research Center: Reports

Quantitative trait loci (QTL) associated with fat deposition have been identified on bovine Chromosome 27 (BTA27) in two different cattle populations. To generate more informative markers for verification and refinement of these QTL-containing intervals, we initiated construction of a BTA27 comparative map. Fourteen genes were selected for mapping based on previously identified regions of conservation between the cattle and human genomes. Markers were developed from the bovine orthologs of genes found on human Chromosomes 1 (HSA1), 4, 8, and 14. Twelve genes were mapped on the bovine linkage map by using markers associated with single nucleotide polymorphisms or microsatellites. Seven …

Development And Demonstration Of Rna Isolation And Rt-Pcr Procedures To Detect Escherichia Coli O157:H7 Gene Expression On Beef Carcass Surfaces, E. D. Berry Jan 2000

Development And Demonstration Of Rna Isolation And Rt-Pcr Procedures To Detect Escherichia Coli O157:H7 Gene Expression On Beef Carcass Surfaces, E. D. Berry

Roman L. Hruska U.S. Meat Animal Research Center: Reports

Preventing the development of pathogen resistance to processing and preservation techniques will require an understanding of the genetic mechanisms that pathogens use in situ to adapt and develop tolerance to stresses they encounter in the food environment. RNA isolation and reverse-transcription (RT)-PCR protocols were developed as tools to detect gene expression in bacteria on beef carcass surfaces. The utility of these procedures was demonstrated by detecting the expression of a selectively-inducible green fluorescent protein (GFP) gene in a plasmid-transformed strain of Escherichia coli O157:H7 inoculated onto beef carcass surface tissue. These procedures should serve as useful tools for studying the …

Porcine Iga Allotypes Are Not Equally Transcribed Or Expressed In Heterozygous Swine, P. Navarro, R.K. Christensen, P. Weber, M. Rothschild, G. Ekhardt, J.E. Butler

Roman L. Hruska U.S. Meat Animal Research Center: Reports

The prediction of 1:1 expression of constant region allotypes in heterozygous animals assumes that productive VDJ rearrangements occur at random between chromosomes, switch recombination is random, there is no allele-related defect in switching and there is no selection for a B-cell receptor bearing a certain constant region allotype. In data reported here, this prediction was often not fulfilled for the transcripts encoding the IgAa and IgAb alleles of porcine IgA including those from late term fetal piglets that are not in contact with environmental antigens or maternal regulatory factors. In the spleen, thymus, mesenteric lymph node (MLN), ileal …

Identification And Characterization Of A New Allele For The Beta Subunit Of Follicle-Stimulating Hormone In Chinese Pig Breeds, M. D. Li, G. A. Rohrer, T. H. Wise, J. J. Ford

Roman L. Hruska U.S. Meat Animal Research Center: Reports

During evaluation of follicle-stimulating hormone- beta (FSHB) expression in anterior pituitary glands by an RNase protection assay (RPA), the expected fragment of 205 nucleotides at positions 759-963 was not detected in one boar that had moderate plasma and pituitary FSH concentrations. After subcloning and sequencing, mRNA from this boar lacked an 11-bp fragment (59-CATTTGGAAAC-39) at nucleotide positions 807-817 of the 39-untranslated region (39-UTR, D allele). Wild-type FSHB (WT allele) was present in pituitary RNA and genomic DNA in both Meishan (MS) and White Composite (WC) pigs; whereas the D allele was present only in MS pigs (P < 0·01; 5/6 MS vs. 0/6 WC). Also, we found the D allele in …

Porcine Erythropoietin Receptor: Molecular Cloning And Expression In Embryonic And Fetal Liver, P. L. Pearson, T. P. L. Smith, T. S. Sonstegard, H. G. Klemcke, R. K. Christenson, J. L. Vallet Jan 2000

Porcine Erythropoietin Receptor: Molecular Cloning And Expression In Embryonic And Fetal Liver, P. L. Pearson, T. P. L. Smith, T. S. Sonstegard, H. G. Klemcke, R. K. Christenson, J. L. Vallet

Roman L. Hruska U.S. Meat Animal Research Center: Reports

The full coding sequence for porcine erythropoietin receptor (EPOR) was elucidated using reverse transcription polymerase chain reaction (PCR) (rtPCR) and 39 and 59 rapid amplification of cDNA ends (RACE) procedures. Total RNA collected from Day 30 fetal liver was used as starting material. A 1843 bp sequence was obtained from which could be inferred a 509 amino acid protein which was 79–85% identical to the amino acid sequence of erythropoietin receptor from other species. Total RNA samples collected from white crossbred intact, white crossbred UHO and Meishan gilts on Days 24, 30 and 40 of gestation were subjected to Northern …

Roman L. Hruska U.S. Meat Animal Research Center: Reports

Although live-attenuated vaccines have been used for some time to control clinical symptoms of the porcine reproductive and respiratory syndrome (PRRS), the molecular bases for the attenuated phenotype remain unclear. We had previously determined the genomic sequence of the pathogenic PRRSV 16244B. Limited comparisons of the structural protein coding sequence of an attenuated vaccine strain have shown 98% homology to the pathogenic 16244B. Here we have confirmed the attenuated phenotype and determined the genomic sequence of that attenuated PRRSV vaccine and compared it to its parental VR- 2332 and the 16244B strains. The attenuated vaccine sequence was colinear with that …
