Genetic And Transcriptional Analyses Of The Vibrio Cholerae Mannose-Sensitive Hemagglutinin Type 4 Pilus Gene Locus, Jane W. Marsh, Ronald K. Taylor

The mannose-sensitive hemagglutinin (MSHA) of the Vibrio cholerae O1 El Tor biotype is a member of the family of type 4 pili. Type 4 pili are found on the surface of a variety of gram-negative bacteria and have demonstrated importance as host colonization factors, bacteriophage receptors, and mediators of DNA transfer. The gene locus required for the assembly and secretion of the MSHA pilus has been localized to a 16.7-kb region of the V. cholerae chromosome. Sixteen genes required for hemagglutination, including five that encode prepilin or prepilin-like proteins, have been identified. Examination of MSHA-specific cDNAs has localized two promoters …

Internal Temperature Of Douglas-Fir Buds Is Altered At Elevated Temperature, Martha E. Apple, Melissa S. Lucash, David M. Olszyk, David T. Tingey, Donald L. Phillips

Pseudotsuga menziesii (Douglas-fir) saplings were grown in sun-lit controlled environment chambers at ambient or elevated (4°C above ambient) temperature. We measured internal temperatures of vegetative buds with thermocouple probes and compared temperatures of normal buds and abnormal buds with loosened, rosetted outer scales in elevated temperature chambers. The abnormal buds had higher and earlier peak daily temperatures than normal buds. Elevated temperature may influence the internal temperature of buds and contribute to the development of abnormal, rosetted buds with loosened outer scales. Abnormal bud development may alter branching patterns and allometry of Douglas-fir trees subjected to climatic change.

Baseline carbaryl susceptibility and potential resistance mechanisms were identified in U.S. populations of western corn rootworm, Diabrotica virgifera virgifera LeConte, using lethal concentration, diagnostic concentration, and synergism bioassays. Twenty-two adult rootworm populations were examined from the states of Indiana, Illinois, Iowa, Kansas, Nebraska, Ohio, Pennsylvania, South Dakota, Texas, and Virginia. Lethal concentration bioassays, conducted within carbaryl-coated glass vials on 12 populations, identified resistance in only 2 Nebraska populations. A diagnostic concentration of carbaryl was subsequently identified which caused 100 and 40% mortality in standard susceptible and known resistant populations, respectively. Influence Of Rhopus Nigroclavatus (Hymenoptera: Encyrtidae) On The Mealybugs Tridiscus Sporoboli And Trionymus Sp. (Homoptera: Pseudococcidae), Tiffany Heng-Moss, Frederick P. Baxendale, Terrance P. Riordan, Linda J. Young

This research investigated parasitism of the buffalograss mealybugs Tridiscus sporoboli (Cockerell) and Trionymus sp. by Rhopus nigroclavatus (Ashmead) through rearing and dissection studies, paired comparison tests, and a field survey. Rates of parasitism reported in this article reflect combined parasitism of the 2 mealybug species. In the rearing study, parasitism of adult female mealybugs by R. nigroclavatus was 48.6%, with an average emergence of 1.77 parasitoids per parasitized mealybug. A maximum of 7 R. nigroclavatus adults emerged from a single mealybug female. Mealybug dissections documented parasitism rates by R. nigroclavatus of 78.5, 67.5, and 4.3%, respectively, for adult females, 3rd …

It has been observed previously that plasma selenium and glutathione levels are subnormal in HIV-infected individuals, and plasma glutathione peroxidase activity is decreased. Under these conditions the survival rate of AIDS patients is reduced significantly. In the present study, using 75Se-labeled human Jurkat T cells, we show that the levels of four 75Se-containing proteins are lower in HIV-infected cell populations than in uninfected cells. These major selenoproteins migrated as 57-, 26-, 21-, and 15-kDa species on SDS/PAGE gels. In our earlier studies, the 57-kDa protein was purified from T cells and identified as a subunit of thioredoxin reductase. …

Aspects of the life cycle of the nematomorph Gordius robustus were investigated. Gordius robustus larvae fed to Tenebrio molitor (Coleoptera: Tenebrionidae) readily penetrated and subsequently encysted in the posterior portion of the midgut wall. Parasite development was then arrested in each infected beetle. Upon feeding these cysts to laboratory-reared Gryllus firmus. worm larvae developed to adults in the cricket hemocoel. In an additional experiment, G. robustus larvae fed to G. firmus did not develop to adults. These findings are in contrast to other studies that found direct infection to be an alternative mode of infection. This experiment is the …

The safe drinking water amendments of 1996 placed a strong emphasis on the goal to establish a nationwide effort to protect drinking water sources. As part of that goal, the legislation provided for a preliminary assessment of drinking water sources and an inventory of surrounding adjacent land use (ALUs). This nationwide effort will result in the first comprehensive look at the nation's drinking water sources from an assessment perspective. Pilin antigenic variation in Neisseria gonorrhoeae may result following intrachromosomal recombination between homologous pi/ genes. Despite extensive study, recA is the only previously characterized gene known to be involved in this process. In this study, the gonococcal recD gene, encoding one subunit of the putative RecBCD holoenzyme, was characterized and its role in pilin variation assessed. The complete recD gene of N. gonorrhoeae M S l l was cloned and its nucleotide sequence determined. The gonococcal recD gene complemented a defined Escherichia coli recD mutant, based on plaque formation of bacteriophage 1 and the restoration of ATP-dependent nuclease activity. Inactivation …

Ocular toxoplasmosis is a potentially blinding intraocular inflammation. The intent of this study was to investigate the role of Fas-FasL interaction in a murine model of acquired ocular toxoplasmosis induced by intracameral inoculation of Toxoplasma gondii. Intraocular inflammation, Fas and FasL expression on lymphocytes and on ocular tissues, the occurrence of apoptosis, and the frequency of CD8+ and CD4+ T cells in the infected eyes were analyzed in C57BL/6 (B6) mice. Susceptibility to parasite-induced intraocular inflammation was observed in Fas-deficient (B6-lpr) and FasL-deficient (B6-gld) mice. Inoculation of 5,000 T. gondii tachyzoites induced significant …

Interaction Between The Mec1-Dependent Dna Synthesis Checkpoint And G1 Cyclin Function In Saccharomyces Cerevisiae, Elizabeth Ann Vallen, F. R. Cross

The completion of DNA synthesis in yeast is monitored by a checkpoint that requires MEC1 and RAD53. Here we show that deletion of the Saccharomyces cerevisiae G1 cyclins CLN1 and CLN2 suppressed the essential requirement for MEC1 function. Wild-type levels of CLN1 and CLN2, or overexpression of CLN1, CLN2, or CLB5, but not CLN3, killed mec1 strains. We identified RNR1, which encodes a subunit of ribonucleotide reductase, as a high-copy suppressor of the lethality of mec1 GAL1-CLN1. Northern analysis demonstrated that RNR1 expression is reduced by CLN1 or CLN2 overexpression. Because limiting RNR1 expression would be expected to decrease dNTP …

Physiological And Morphological Correlates Of Presynaptic Inhibition In Primary Afferents Of The Lamprey Spinal Cord, I. V. Batueva, E. A. Tsvetkov, A. K. Sagatelyan, James T. Buchanan, N. Vesselkin, V. Adanina, E. I. Suderevskaya, J. -P Rio, J. Repérant

Patch-clamp recordings in a whole-cell mode were performed on dorsal sensory cells enzymatically isolated from the spinal cord of two lamprey species, Ichthyomyzon unicuspis and Lampetra fluviatilis. The voltage-activated currents through calcium channels were analysed. GABA and the specific GABAB receptor agonist baclofen reduced the peak amplitude of inward Ba2+ current, as a robust alternate charge carrier through voltage-dependent Ca2+ channels. These effects were dose-dependent and reversible. GABAB receptor antagonists, 2-hydroxysaclofen and δ-amino-n-valeric acid, blocked the reduction of Ba2+ currents by GABA and baclofen, while bicuculline, a GABAA receptor antagonist, had no blocking …
