The Use Of Polypeptide Probes Selected Fromartificial Peptide Libraries For The Recognitionand Differentiation Of Dna Sequences, Erol Ömer Atalay, Berri̇n Erdağ, Beyazit Çirakoğlu

Turkish Journal of Medical Sciences

In the novel approach suggested here, we examined the use of polypeptide probes selected from artificial peptide libraries for the identification of the differences in dsDNA sequences which are potential targets for the genetic diseases. As a model system two target DNAs which are the polymerase chain reaction (PCR) products of the two hage-display vector multiple cloning sites (MCS) differing at 18 base pair long has been used. In the production of the target sequences biotinylated primers were used in PCR. The polypeptide probes obtained from artificial peptide library by the selection of phage-display techniques. Identified clones bearing the candidate …

Technical Approach To Generate Polyclonalantibodies Against Bacterially Expressed Gst-Pyk-C: Evaluation Of Cross-Reaction Andrecognition In Some Mammalian And Yeast Celllysates, Ayşegül Ayyildiz

Turkish Journal of Medical Sciences

Pyk-2 is a protein tyrosine kinase, highly expressed in human hippocampus, dentate gyrus and olfactory bulb. In this study, polyclonal antibodies against the C-terminal of pyk-2, starting from Pro-759, were generated by injecting rabbits with the bacterially expressed GST fusion construct, generated by cloning the C-terminal domain of pyk-2 into pGEX-3X vector and affinity purification to increase the specificity of recognition. Anti-pyk-C was shown to recognize a 123 kD band in Western Blots of 293 cells transfected with pyk-2, as well as PC-12, COS and 3T3 cells and not to cross-react with FAK transfected Sf-21 lysates, in spite of high …

Opioid Modulation Of The Pre-Ovulatory Lh Surge In The Conscious Rat: Involvement Of Indolamines, Bayram Yilmaz, Desmond P. Gilmore

Turkish Journal of Medical Sciences

Endogeonus opioid peptides play an important role in the generation of the pre-ovulatory LH surge. We investigated the interrelationship between the oploid and indolaminergic systems in the regutalion of LH secretion, and the involvement of \mu-and \kappa-opioid subtypes in this process. Concsious female rats bearing cannula in the femoral artery were injected intraperitoneally with \mu-(diamorphine) and \kappa -(U-50488H) opioid agonists either alone or with their respective antagonists, naloxone and MR2266, on pro-oestrus. Blood samples were collected hourly between 15.00 and 19.00 and plasma LH levels were measured by RIA. 5-HT and 5-HIAA concentrations were determined by HPLC-ECD in the MPOA, …

The Effects Of Hyaluronic Acid On Myringoplasty Inrats, Ahmet Kutluhan, Serdar Uğraş, Emi̇n İnalkaç, Nusret Akpolat

Turkish Journal of Medical Sciences

Tympanic membrane perforations arise from a variety of causes. Some of them require myringoplasty. In this study, we used hyaluronic acid, a polysaccharide with high-molecular- weight which may be obtained commercially and affects modulations of cell function. An experimental myringoplasty was performed on rats in which a chronic dry tympanic membrane perforation was formed. While physiological saline was embedded in Gelfoam ® which was supported under and over the graft, in group 1, hyaluronic acid embedded gelfoam was used in group 2. Better epithelization, less calcification and scar tissue were established in group 2 myringoplasties. This study shows that hyaluronic …

The effect of primaquine on the enzymatic antioxidant defence mechanisms of erythrocytes were determined by measuring the activities of the below enzymes, before and after 30 minutes of incubation of hemolyzates with 1 mM primaquine at 37°C. Among the enzymes studied glucose-6- phosphate dehydrogenase seemed the most sensitive enzyme; 57 % and 78 % inhibition at zero (due to the binding of primaquine to NADP binding site) and 30 minutes of incubation (due to the reactive oxygen species), respectively. Catalase, superoxide dismutase, and glutathione-S-transferase were less sensitive to primaquine-derived reactive oxygen species: 33, 11, and 0 % inhibition at the …

Estimation Of The Prevalence Of A Disease From Screening Tests, Ergun Karaağaoğlu Jan 1999

Estimation Of The Prevalence Of A Disease From Screening Tests, Ergun Karaağaoğlu

Turkish Journal of Medical Sciences

Since most secreening tests are not 100% accurate, the proportion of subjects screened positive in such a test cannot be used as an estimate of the population prevalence. Methods which take sensitivity and the specificity into consideration should be employed in such circumstances. Estimation of the population prevalence as defined by Gartand Buck may produce results which are outside the range of 0 to 1. A Bayesian approach avoids results of this kind, but requires complicated computations. Lew and Levy proposed an approximation to the Bayesian estimate of the population prevalence. To simplify the computations, I propose a method which …

Turkish Journal of Medical Sciences

It has been estimated that between 2% to 28% of the general adult population has radiographic evidence of elongated styloid process. In adults, the mean radiographic lenght of the styloid process is 20 to 30 mm and its tip is located between the external and internal carotid arteries, just lateral to the tonsillar fossa. Eagle's syndrome occurs when an elongated styloid process or ossified stylohyoid ligament causes recurrent throat pain or foreign body sensation, dysphagia, or facial pain. It may develop inflammatory changes or impinge on the adjacent arteries or sensory nerve endings, leading to the symptoms described. To study …

Diastolic Dysfunction: Pathogenesis, Therapy Andthe Importance Of Doppler Echocardiography, Fati̇h Yalçin, James Thomas Jan 1999

Diastolic Dysfunction: Pathogenesis, Therapy Andthe Importance Of Doppler Echocardiography, Fati̇h Yalçin, James Thomas

Turkish Journal of Medical Sciences

Diastolic dysfunction is characterized by increased resistance to ventricular filling, inappropriate upward shift of the diastolic pressure-volume relationship and exercise intolerance. Inappropriate tachycardia, decreased diastolic compliance and impaired systolic relaxation lead to diastolic dysfunction. Optimal therapy will depend on the type and pathophysiologic phase of disease. Doppler echocardiography is a valuable tool to diagnose diastolic dysfunction and therapeutic effects of drugs on diastolic function in different groups of patients. Recently a lot of studies have been going on this subject.

In-Situ Measurement Of Denitrification In The Elk Valley Aquifer, Allen Schlag Jan 1999

In-Situ Measurement Of Denitrification In The Elk Valley Aquifer, Allen Schlag

Theses and Dissertations

Groundwater serves more than half of North Dakota's population as a drinking water source. Nitrate (NO₃¯) is a common groundwater contaminant associated with fertilizers, septic systems and livestock waste in shallow unconfined aquifers. As a suspected carcinogen and cause of potentially lethal methemoglobinemia in infants, the natural attenuation of NO₃¯ by denitrification can be an important process in keeping groundwater as a reliable source of clean drinking water for North Dakota. Organic carbon is often considered the most important electron donor for the chemical reduction of NO₃¯ into harmless nitrogen gas. However, several studies have shown that inorganic electron donors …
