Characterization Of Catalytic Phenotype Of \Beta-Galactosidasefrom Laci Mutant, E. Coli Csh-36,As A Tool For The Management Of Lactoseintolerance, Ayşegül Ayyildiz

Turkish Journal of Medical Sciences

Objectives: Lactose intolerance is a common public health problem with 5-100% prevalance among different populations of the world. Use of \beta-galactosidase to reduce the lactose content in food or consumption of special products of milk or exogeneous lactase enzyme are among the approaches taken to manage lactose intolerance. Recombinant \beta -galactosidase as well as \beta -galactosidase derived from different sources have variances in their bioefficiency. Thermodynamic properties of \beta -galactosidase determines the industrial efficiency. In this \beta -galactosidase from E. coli CSH-36, a constitutive expressor of enzyme was investigated for enzyme biofunctionality. Methods: The enzyme was purified by cell lysis, …

The Intracellular Concentration Of Components Ofthe Ubiquitin System In A549 Culture Cellsduring Uv Treatment, İbrahi̇m Pi̇ri̇m

Turkish Journal of Medical Sciences

This study attempts to determine a link between the intracellular components of the ubiquitin (Ub) system and the process of DNA repair in eukaryotes. It is possible to investigate this link in two ways: i. studying the concentration of mRNA for specific Ub components in DNA damaged and controlled cells. ii. studying the concentration of proteins of the Ub system in both DNA damaged and control cells. The specific aim of this study was to try to establish such a link between repair and Ub, by studying and making a quantitative analysis of the intracellular concentrations of both the Ub …

S-Aroylmethyl N, N-Disubstituteddithiocarbamates With Antiparkinson Activity, Rahi̇me Şi̇mşek, Ci̇hat Şafak, Kevser Erol, Kami̇l Vural Jan 1999

S-Aroylmethyl N, N-Disubstituteddithiocarbamates With Antiparkinson Activity, Rahi̇me Şi̇mşek, Ci̇hat Şafak, Kevser Erol, Kami̇l Vural

Turkish Journal of Medical Sciences

Seven S-aroylmethyl N,N-disubstituted dithiocarbamate derivatives have been synthesized and their effects on oxotremorine induced tremor have been investigated on mice pretreated with the proparkinsonian drug haloperidol. While the compound 1f inhibited oxotremorine-induced tremors at doses of 50 mg/kg and 100 mg/kg levels, compounds 1a, 1c and 1e had this effect at the dose of 100 mg/kg. The compounds 1b, 1d and 1g had no effect on oxotremorine-induced tremors in mice. These results suggest that some of these derivatives have central antimuscarinic effects and antiparkinson activities.

Turkish Journal of Medical Sciences

The aim of this study is to examine expression of Proliferating cell nuclear antigen (PCNA) and p53 oncoprotein in gastric neuroendocrine tumors. 35 gastric neuroendocrine tumors were analyzed for first histopathologic features and then, immunohistochemical study was done to detect labeling index of PCNA and accumulation of p53 protein by using monoclonal antibodies and the Labeled strepavidin-biotin method. PCNA labeling index and p53 protein expression were compared with histopatholoic features in statistical methods of Fisher's exact, chi square and logistic regression tests. No significant correlation was found between them except PCNA score and depth of invasion. In conclusion, our findings …

Turkish Journal of Medical Sciences

There are both histomorphometric and nonhistomorphometric studies confirming that etidronate reduces bone resorption. In this study, we have examined urinary Calcium/Creatinine ratio (uCA/Cr) as a biochemical marker of bone turnover to show the effectiveness of etidronate and whether it could be used as a follow-up parameter of treatment. Eighty-one postmenopausal women aged 40 to 65 included into the study to investigate the effects of etidronate on uCa/Cr in a prospective, randomised, placebo controlled clinical trial. All necessary criteria matched 81 women were divided into 3 groups at random, each group consisted of 27 patients. Prior to treatment, uCa/Cr was calculated …

Turkish Journal of Veterinary & Animal Sciences

Pulmonary aspergillosis were examinated in a total of 22 geese (2-13 week-old), brought from A Geese Production Station in Kars and geese breeders. The lesions were seen as a hard nodules in all cases in the lungs, and also in 3 cases in the air sacs. They were multifocalty localized in each lobules, and 1-4 mm. in diameter and yellow in color. The lesions were characterized by granulomatous pneumonie and airsacculitis. The characteristic hyphae of Aspergillus spp. were demonstrated in granuloma in histopathological sections stained by periodic acid Schiff stain.

Turkish Journal of Veterinary & Animal Sciences

The ultrastructure of endogenous stages of Cryptosporidium muris in the stomach of laboratory mice was studied by transmission electron microscopy. Electron microscopy showed the various developmental stages as free in the gastric gland lumina and attached to the mucus cells. The changes in organelle composition and structure of various stages from the sporozoite to young trophozoites and matured oocysts were demonstrated in detail. Two types of the meront were discriminated. The finding of a microgamete on the parasitophorous vacuolar membrane surrounded a macrogamont was interesting for the stage of fertilization in C. muris. Types 1 and 2 wall-forming bodies were …

Usually basal-parabasal type of cells appeared in vaginal cytologic examination, invasion of neutrophyl leucocytes and in some cases degenerative cell changes proved inflammatory changes. In bitches with CEH-pyometra complex there was a very significant increase in AST (Aspartate aminotransferase) and decrease in the …

Turkish Journal of Veterinary & Animal Sciences

Ovine immunoglobulin M was isolated from sheep serum using gel filtration and affinity chromatography. Polyclonal and monoclonal antibodies against this isotype were prepared. The specificity of the monoclonal antibody was investigated using SDS-Page, immunoblotting, an enzyme-linked immunosorbent assay (ELISA) and affinity chromatography. On immunoblotting, monoclonal anti-IgM reacted with a 70 kD protein which corresponded to ovine IgM.

Turkish Journal of Veterinary & Animal Sciences

The location of extrinsic afferent and efferent neurons projecting to the rat gastrointestinal tract were mapped using the retrograde tracer, True Blue (TB). Injection of retrograde tracer, TB into stomach, proximal colon and distal colon resulted in labelled cells in the dorsal root ganglia. Cells were also labelled in the nodose ganglia following injection of tracer into stomach and proximal colon. TB-labelled cells were also seen in the prevertebral ganglia (coeliacomesenteric complex and inferior mesenteric ganglion), after injection of TB into the stomach, proximal and distal colon. Many TB-labelled cells were detected in the dorsal motor nucleus of the vagus …
