Because the Sand Hills are, for the most part, a treeless landscape of grass-covered sand dunes, most of the insects are adapted to sandy habitats and a dry, continental climate, as well as to the kinds of plants and other animals that live there. Unlike most forest-adapted species, insects in the Sand Hills tolerate drier and windier conditions and greater solar radiation. They have also been successful in surviving the periodic fires that are so necessary for maintaining native grasslands. Identification Of A G(Iα) Binding Site On Type V Adenylyl Cyclase, Carmen W. Dessauer, John J.G. Tesmer, Stephen R. Sprang, Alfred G. Gilman

The stimulatory G protein α subunit G(sα) binds within a cleft in adenylyl cyclase formed by the α1-α2 and α3-β4 loops of the C₂ domain. The pseudosymmetry of the C₁ and C₂ domains of adenylyl cyclase suggests that the homologous inhibitory α subunit G(iα) could bind to the analogous cleft within C₁. We demonstrate that myristoylated guanosine 5'-3-O- (thio)triphosphate-G(iα1) forms a stable complex with the C₁ (but not the C₂) domain of type V adenylyl cyclase. Thiazolidinediones Inhibit Lipoprotein Lipase Activity In Adipocytes, Subramanian Ranganathan, Philip A. Kern

The thiazolidinediones troglitazone and BRL 49653 improve insulin sensitivity in humans and animals with insulin resistance. Adipose tissue lipoprotein lipase is an insulin-sensitive enzyme. We examined the effects of thiazolidinediones on lipoprotein lipase expression in adipocytes. When added to 3T3-F442A, 3T3-L1, and rat adipocytes in culture, troglitazone and BRL 49653 inhibited lipoprotein lipase activity. This inhibition was observed at concentrations as low as 0.1 μM and within 2 h after addition of the drug. Lipoprotein lipase activity was inhibited in differentiated adipocytes as well as the differentiating cells. Despite this decrease in enzyme activity, these drugs increased mRNA levels in …

Cryptosporidiosis (crypto), a disease affecting both animals and humans, is caused by the organism Cryptosporidium. This is not a bacteria or virus but a group of parasitic protozoans (single-celled organisms that live off other organisms). There are several species of Cryptosporidium, most of which infect only specific species of animals.

Assessing The Accuracy Of A New Diagnostic Test When A Gold Standard Does Not Exist, Todd A. Alonzo, Margaret S. Pepe

UW Biostatistics Working Paper Series

Often the accuracy of a new diagnostic test must be assessed when a perfect gold standard does not exist. Use of an imperfect test biases the accuracy estimates of the new test. This paper reviews existing approaches to this problem including discrepant resolution and latent class analysis. Deficiencies with these approaches are identified. A new approach is proposed that combines the results of several imperfect reference tests to define a better reference standard. We call this the composite reference standard (CRS). Using the CRS, accuracy can be assessed using multistage sampling designs. Maximum likelihood estimates of accuracy and expressions for …
