The efficacy of triple drug therapy for HIV-1 infection encourages its early use to prevent damage to the immune system. We monitored the effects of such therapy on 12 patients with 14-75-mo histories of minimal disease, i.e., CD4+ counts constantly >500/μl and little or no lymph node enlargement. In this way, we could first determine the extent of viral replication and immunoarchitectural changes in unenlarged nodes early in disease, and second follow the response to triple therapy in plasma and lymphoid tissue in tandem. In eukaryotic cells the segregation of the genome in a closed organelle requires an efficient mechanism to ensure the constant exchange of material between the nucleus and the cytoplasm. Nuclear pore complexes (NPCs) provide the only known sites for exchange of material across the nuclear envelope (NE). A wealth of evidence has accumulated throughout the years that strongly suggest that the NPCs are structurally linked to the nuclear interior. It has long been proposed that this structural continuity is essential for the efficient exchange of material between the nuclear interior and the cytoplasm. This paper will provide the reader with an overview of popular Cuban music and dance forms as well as an understanding of how the island’s history of cultural diversity has empowered its music to become an influential component to all of Latin music. In particular, African cultural elements that have effected Cuba’s popular culture as well as its national identity will be discussed. Beginning with African tribal influences as well as other cultural elements, known prior to the abolition of slavery, the paper will move into the post-slavery evolution of Cuba’s popular culture. Regulation And Role Of Calcium In Pancreatic Exocytosis, Stacey Jeanne Hanc

The pancreas is an organ designed to synthesize, package, and release enzymes required by the digestive system. These enzymes are packaged in secretory vesicles and are released by exocytosis following stimulation of the pancreatic acinar cells by gastrointestinal hormones. A recognized aspect of this signal is an increase in the concentration of intracellular Ca2+. Previous work has demonstrated that the increase occurs preferentially in the apical pole of the cell in the region of the secretory vesicles prior to the onset of exocytosis. Secretory vesicles were determined to contain significant amounts of Ca2+ by FURA-2 fluorescence. Purified secretory vesicle membranes …

Until recently, the measurement of mRNA levels was very time consuming and required specific parameters. With the advent of RT-PCR, mRNA fragments could be amplified and easily detected and measured. The amount of mRNA loaded into the reaction is proportional to the amount of product DNA. Here, we propose a new method for mRNA detection and quantification. The product of the amplification reaction is resolved electrophoretically on a polyacrylamide gel and then transferred to a positively charged nylon membrane. This membrane is then stained for DNA with a non-mutagenic dye. The membrane is converted to a visual image via a …

The Justice Who Wouldn't Be Lutheran: Toward Borrowing The Wisdom Of Faith Traditions, Marie Failinger

Although many authors have debated the propriety of the use of religious arguments in public policy discussions and lawmaking, few have critically reviewed the jurisprudence of particular judges through the lens of their own faith-traditions. Preliminarily, this article attempts a modest contribution to the discussion about the use of religious argument in public discussions by suggesting that judicial opinions should be viewed rhetorically and that religious assumptions and claims may legitimately be "borrowed" analogically into such opinions, at least their forensic and epideictic elements. More concretely, it analyzes themes in some of Justice William Rehnquist's opinions to determine how consistent …

Phorbol Ester Activation Of An Nhe-Like Electroneutral Na+ /H+ Antiporter In Isolated E-Cells Of Lobster (Homarus Americanus) Hepatopancreas, Jeff Duerr, Gregory A. Ahearn

The basolateral membrane of Atlantic lobster (Homarus americanus) epithelium possesses an electroneutral Na+ /H+ antiporter that functionally resembles members of the vertebrate NHE family. Regulatory mechanisms of this antiporter in purified hepatopancreatic E-cell suspensions, produced with a centrifugal elutriation technique, were investigated. Suspensions routinely consisted of greater than 95% E-cells displaying greater than 90% viability. Intracellular pH (pHi) was monitored by loading cells with the fluorescent dye 2´,7´-bis(carboxyethyl)-5,6- carboxyfluorescein (BCECF), and placing suspensions in a spectrofluorometer. Recovery from induced acid-loading was mediated by a Na+ -dependent, dimethylamiloride-sensitive proton efflux. Antiport activation was a sigmoidal function of pHi at values below …
