Isolation Of A Highly Active Photosystem Ii Preparation From Synechocystis 6803 Using A Histidine-Tagged Mutant Of Cp 47, Terry M. Bricker, Jason Morvant, Najy Masri, Holly M. Sutton, Laurie K. Frankel

Site-directed mutagenesis was used to produce a Synechocystis mutant containing a histidine tag at the C terminus of the CP 47 protein of Photosystem II. This mutant cell line, designated HT-3, exhibited slightly above normal rates of oxygen evolution and appeared to accumulate somewhat more Photosystem II reaction centers than a control strain. A rapidly isolatable (<7 >h) oxygen-evolving Photosystem II preparation was prepared from HT-3 using dodecyl-β-d-maltoside solubilization and Co2+ metal affinity chromatography. This histidine-tagged Photosystem II preparation stably evolved oxygen at a high rate (2440 μmol O2 (mg chl)-1 h-1), exhibited an α-band absorption maximum at 674 nm, …7>

Hsp70 And Larval Thermotolerance In Drosophila Melanogaster: How Much Is Enough And When Is More Too Much?, Robert A. Krebs, Martin E. Feder

Heat shock proteins (Hsps) and other molecular chaperones perform diverse cellular roles (e.g., inducible thermotolerance) whose functional consequences are concentration dependent. We manipulated Hsp70 concentration quantitatively in intact larvae of Drosophila melanogaster to examine its effect on survival, developmental time and tissue damage after heat shock. Larvae of an extra-copy strain, which has 22 hsp70 copies, produced Hsp70 more rapidly and to higher concentrations than larvae of a control strain, which has the wild-type 10 copies of the gene. Increasing the magnitude and duration of pretreatment increased Hsp70 concentrations, improved tolerance of more severe stress, and reduced delays in development. …

Understanding the factors that influence soil and plant nitrogen (N) spatial variability may improve our ability to develop management systems that maximize productivity and minimize environmental hazards. The objective of this study was to determine the field (65 ha) scale spatial variability of N and δ15N in soil and corn (Zea mays). Soil, grain, and stover samples were collected from grids that ranged in size from 30 by 30 m to 60 by 60 m. An Ab Initio Study Of Specific Solvent Effects On The Electronic Coupling Element In Electron Transfer Reactions, Thomas M. Henderson '98, Robert J. Cave

Specific solvent effects on the electronic coupling element for electron transfer are examined using two model donor–acceptor systems (Zn2+ and Li2+) and several model “solvent” species (He, Ne, H2O, and NH3). The effects are evaluated relative to the given donor–acceptor pair without solvent present. The electronic coupling element (Hab) is found to depend strongly on the identity of the intervening solvent, with He atoms decreasing Hab, whereas H2O and NH3 significantly increase Hab. The distance dependence (essentially exponential decay) is weakly affected by a single intervening solvent atom–molecule. Abstract—Toxic trace metals have been implicated as a potential cause of recent flamingo kills at Lake Nakuru, Kenya. Chromium (Cr), copper (Cu), lead (Pb), and zinc (Zn) have accumulated in the lake sediments as a result of unregulated discharges and because this alkaline lake has no natural outlet. Lesser flamingos (Phoeniconaias minor) at Lake Nakuru feed predominantly on the cyanobacterium Spirulina platensis, and because of their filter-feeding mechanism, they are susceptible to exposure to particle-bound metals. Trace metal adsorption isotherms to lake sediments and S. platensis were obtained under simulated lake conditions, and a mathematical model was …

For 36 years, the Council has been presenting these conferences for the purpose of advancing knowledge in the field of vertebrate pest management.
