In this study, we determined the contractile properties of single chemically skinned fibers prepared from the medial gastrocnemius (MG) and soleus (Sol) muscles of adult male rhesus monkeys and assessed the effects of the spaceflight living facility known as the experiment support primate facility (ESOP). Muscle biopsies were obtained 4 wk before and immediately after an 18-day ESOP sit, and fiber type was determined by immunohistochemical techniques. The MG slow type I fiber was significantly smaller than the MG type II, Sol type I, and Sol type II fibers. The ESOP sit caused a significant reduction in the diameter of …

Conventional light microscopy allows the observation of living as well as fixed cells and tissues to generate two-dimensional images. The out-of-focus information often obscures the ultrastructural details, especially in thick specimens with overlapping structures. The earliest available light microscopy visualized the objects in hydrated state in two-dimensions during their temporal development. The emergence of electron microscopy (EM) provided superb resolution of ultrastructural details, but it was applicable only for objects in the dehydrated state and thereby potentially introducing handling artifacts. Various forms of water erosion were present that cumulatively, would probably have a significant effect on the water quality of the Canning (and Swan) River. Much of the water erosion results from the installation and maintenance of infrastructure and developments, e.g. roads, subdivisions, stormwater drains etc. Reviewing standard practices, raising of awareness of consequences of erosion and training in urban erosion and sediment control techniques is recommended. A. Purpose. In 1990, the Base Realignment and Closure (BRAC) Commission designated Fort Ord, California for closure. At that time, studies were begun by the Army to determine the conditions of the facilities and properties comprising the base. The Fort Ord Reuse Authority (FORA) was established to develop plans for the reuse of the facilities. FORA was a coalition of representatives from the local city, county, and state governments. When the base closed in 1992, FORA took the lead in developing the Fort Ord Reuse Plan (FORP).

Toxin-producing Escherichia coli expressing F18 fimbriae colonizes the small intestines of weaned pigs and causes diarrhea, edema disease, or both. The F18 family is composed of two antigenic variants, F18ab and F18ac. Because many strains do not express F18 fimbriae in vitro, identification and differentiation of these two variants are difficult. Single-strand conformational polymorphism (SSCP) analysis is a rapid method for identifying genetic mutations and polymorphisms. The F18 major fimbrial subunit genes (fedA) of 138 strains were amplified by PCR, and genetic differences were detected by SSCP analysis. The SSCP analysis of the fedA gene differentiated F18ab+ …

Canada geese (Branta canadensis) recently have become management problems in some areas due to overpopulation. At Fish Springs National Wildlife Refuge (NWR), managers are concerned with the opposite situation. Despite attempts to boost production, only 18 to 34 goslings of the western Canada goose (B. c. moffitti) were produced per year, from 1989 to 1993. I studied the breeding population from March to July in 1996 and 1997. Results suggest that production is limited in 3 ways: low gosling survival, low nesting success for ground nests, and low number of breeding pairs.

1,25-dihydroxyvitamin D3 (1,25(OH)2D3) has been shown to mediate the rapid, non-nuclear stimulation of calcium and phosphate transport in chick intestine through binding to a receptor localized in the basal lateral membrane. By using an antibody to the N-terminus of the membrane receptor, studies were undertaken to determine whether a comparable protein exists in kidney and brain, and whether it is present in a particular subcellular fraction.

The first step was to establish fractionation protocols to separate subcellular organelles as judged by marker enzyme analyses. Differential centrifugation and Percoll gradient fractions were prepared from chick kidney …

The antioxidant activity of carnosine and phytic acid was investigated using several model systems. Carnosine and phytic acid alone inhibited metal ion-catalyzed deoxyribose degradation. Carnosine strongly inhibited metal ion-catalyzed lipid peroxidation in liposomes and in ground beef homogenates. Phytic acid facilitated oxidation of Fe (II) to Fe (III), and it inhibited hemeprotein + H2O2-catalyzed lipid peroxidation in linoleic acid micelles.

Antioxidant and color stabilizing effects of carnosine and phytate were investigated in a beef model system. Both compounds increased the rate of pH decline in pre-rigor beef muscle and stabilized fresh meat color by inhibiting metmyoglobin …

Changes In The Breeding Bird Community Of Subtropical Evergreen Forest In The Lower Rio Grande Valley Of Texas, 1970s-1990s, Timothy Brush, Alberto Cantu

Breeding-bird censuses conducted in 1994-1996 at the Santa Ana National Wildlife, southern Texas were compared to those done on the same site in 1973-1978. The study site was described as subtropical evergreen forest in the 1970s, but is now thorn-forest with remnant evergreen forest. Canopy cover was incomplete and lower and trees were smaller and more densely packed in 1994-1996 than in 1973-1978. Seven bird species typical of thorn-forest and thorn-scrub, which were not present in 1973-1978, made up 20% of the breeding bird community in 1994-1996. The problems of lead as an environmental toxicant are well established. These problems are complicated by the need for better analytical methods to determine trace quantities of lead in various biological samples, particularly biological fluids. The most widely used method which clinical laboratories use is the Graphite Furnace Atomic Absorption Spectroscopic (GFAAS) technique. The problems of determining lead in biological fluids such as blood are due to the low melting point of lead and the large amount of biological materials such as blood cell fragments. Camaric acid (3,25-epoxy-3a-hydroxy-22�(2-methyl-2z-butenoyloxy)-12-0Ieanen-28- oic acid), camarinic acid (22�-acetoxy-3,25-epoxy-3a-hydroxy-12-ursen-12-en-28-oic acid), lantic acid (3�,25-epoxy-3a-hydroxyrus-12-en-28-oic acid), and lantanilic acid (22�­ dimethylacryloyloxy-3�,25-epoxy-3a-hydroxyolean-12-en-28-oic acid) have been isolated by guided HPLC fractionation ofdichloromathane:methanol (1: 1) extract of Lantana camara obtained from the Environmental Chemistry and Toxicology Laboratory, Texas Southern University. The structures of the various compounds were confirmed through spectral methods: UV, IR, MS and IH_NMR and by comparison of their data with the published values. The purpose of this study is to report the chemical composition and structures of the local ornamental plants (Lantana camara).

Repetitive stimulation of motor nerves causes an increase in the number of packets of transmitter ("quanta") that can be released in the ensuing period. This represents a type of conditioning, in which synaptic transmission may be enhanced by prior activity. Despite many studies of this phenomenon, there have been no investigations of the quantal mechanisms underlying these events, due to the rapid changes in transmitter output and the short time periods involved. Tumor necrosis factor α (TNFα) is a pleiotropic cytokine that is predominantly produced by activated macrophages. The effects of TNFα are as diverse as the cells with which it interacts, e.g., stimulating fibroblast growth, exerting cytotoxic/cytostatic; activity against various human and murine cell lines, promoting inflammation through upregulation of endothelial adhesion molecules and IL-8 production. Yet TNFα is best known, and in fact was originally described, for its role in the bacterial-induced hemorrhagic necrosis of tumors and exacerbation of septic shock in which aberrant TNFα production leads to vascular collapse, cachexia, multiple organ failure, and ultimately death in as many …

Molecular analysis of biological specimens usually requires extraction of high-molecular-weight DNA free of foreign DNA contaminants. DNA was extracted from black flies at different life stages that had been preserved by 4 methods: larvae and adults in ethanol, larvae in Carnoy’s solution, adults on card-points, and adults hand-swatted and sun-dried. Using specific primers for the mitochondrial ND4 gene, a 257-bp amplicon was obtained from specimens preserved by ethanol, card-point mounting, and sun-drying. Successful amplification often required DNA dilutions ≥ 1:20 (<1–10 ng). DNA from specimens preserved in Carnoy’s solution (ethanol: acetic acid, 3:1) yielded degraded DNA, resulting in fewer successful amplifications. Parasitic nematodes and, to a lesser extent, gut contents resulted in extra products when amplified with randomly amplified polymorphic DNA (RAPD) primers. Sufficient DNA was extracted from the head of a larva for a successful polymerase chain reaction (PCR), eliminating the need to remove the contaminating gut and parasites.

The last systematic review of the yellow-shouldered bats of the Neotropical genus Sturnira in the Lesser Antilles was in 1976 (Jones and Phillips, 1976). At that point in time, two species--Sturnira lilium and Sturnira thomasi--were known from these islands. Sturnira lilium was represented by five subspecies, beginning with Trinidad and moving northward, these subspecies were lilium on Trinidad (Goodwin and Greenhall, 1961), paulsoni on St. Vincent (de Ia Torre and Schwartz, 19661, luciae on St. Lucia (Jones and Phillip, 19761, zygomaticus on Martinique (Jones and Phillips, 1976), and angeli on Dominica (de la Tom, 1966).

The role of chemical constituents in the foraging behavior of black bears (Ursus americanus) was investigated using two field studies. Vascular tissue samples were collected from Douglas-fir (Pseudotsuga menziesii) trees recently foraged by black bears. Samples were extracted and analyzed by liquid and gas chromatography to determine carbohydrates and terpenes, respectively. Chemical data were subjected to correlation analyses and multiple regression to examine if they adequately describe observed foraging preferences. Cladistic analysis based on comparative morphology was used to examine the subfamily-level relationships within the cestode order Proteocephalidea. A single most parsimonious tree (70 steps, CI = 0.571; RC = 0.295; HI = 0.471) is consistent with monophyly for the Proteocephalidea and showed a relatively high consistency at the family level with the diagnosis of two major subclades. Unambiguous support for a Proteocephalidae subclade, including the Corallobothriinae, Proteocephalinae, Gangesiinae and Sandonellinae, and a Monticelliidae subclade, including the Marsypocephalinae, Zygobothriinae, Monticelliinae, Rudolphiellinae, Ephedrocephalinae and Othinoscolecinae was evident.
