Rhodococcus Equi And Arcanobacterium Haemolyticum: Two "Coryneform" Bacteria Increasingly Recognized As Agents Of Human Infection, R. Linder

Rhodococcus equi and Arcanobacterium haemolyticum, formerly classified in the genus Corynebacterium, are members of the loosely defined taxon "coryneform" bacteria. Although they are the etiologic agents of distinct human infections, both organisms are frequently overlooked, which results in missed or delayed diagnoses. R. equi, long known as an important pathogen of immature horses, has become in the past three decades an opportunistic pathogen of severely immunosuppressed humans. Most cases are secondary to HIV infection. When specifically sought in throat swab cultures, A. haemolyticum is found responsible for 0.5% to 2.5% of bacterial pharyngitis, especially among adolescents. These two microorganisms represent …

Dendroctonus jeffreyi and D. ponderosae are sibling species of bark beetles (Coleoptera: Scolyti- dae) with few morphological and molecular genetic differences. The two species are believed to have di- verged relatively recently. Dendroctonus jeffreyi colo- nizes only Pinus jeffreyi, while D. ponderosae colonizes up to thirteen Pinus spp., but not P. jeffreyi. Adult beetles of both D. jeffreyi and D. ponderosae carry sym- biotic fungi in mycangia located on the maxillary car- dines. Dendroctonus ponderosae was known to carry two fungi, Ophiostoma clavigerum and 0. montium, in its mycangia. However, it was not known which fungi might be carried by …

Management of ecosystems at large regional or continental scales and determination of the vulnerability of ecosystems to large-scale changes in climate or atmospheric chemistry require understanding how ecosystem processes are governed at large spatial scales. A collaborative project, the Vegetation and Ecosystem Modeling and Analysis Project (VEMAP), addressed modeling of multiple resource limitation at the scale of the conterminous United States, and the responses of ecosystems to environmental change. In this paper, we evaluate the model-generated patterns of spatial variability within and between ecosystems using Century, TEM, and Biome-BGC, and the relationships between modeled water balance, nutrients, and carbon dynamics. …

The increase in phenotypic or morphological complexity in organisms may stem from a corresponding increase in the complexity of the underlying genetic architecture, driven by the process of gene duplication. Gene duplication is a mutational mechanism that can impact the genome through the gradual birth and death of individual genes or clusters of genes, and through infrequent episodic events of whole genome duplication. Functional and pleiotropic differences among genes may impact the probability of fixation of duplicated genes and the facility of gene families to record duplication events across deep history. The mechanisms governing neuronal differentiation, including the signals underlying the induction of voltage-dependent sodium (Na+) channel expression by neurotrophic factors, which occurs independent of Ras activity, are not well understood. Therefore, Na+ channel induction was analyzed in sublines of PC12 cells stably expressing platelet-derived growth factor (PDGF) beta receptors with mutations that eliminate activation of specific signalling molecules. Mutations eliminating activation of phosphatidylinositol 3-kinase (PI3K), phospholipase C gamma (PLC gamma), the GTPase-activating protein (GAP), and Syp phosphatase failed to diminish the induction of type II Na+ channel alpha-subunit mRNA and functional Na+ channel expression by PDGF, as determined by RNase …

Salmonellosis continues to be a major infectious disease in both the United States and elsewhere. The outer membrane porin proteins (Omp) from Salmonella typhimurium play a key role in the initial adherence of this microorganism to murine macrophages and this mechanism is a critical event in the pathogenesis of infections. In this study macrophages were found to make contact with an OmpC-like protein of S. typhimurium and inhibition assays using the purified protein significantly reduced bacterial binding to macrophage. The ompC gene in a strain of S. typhimurium that is resistant to killing by macrophage was inactivated by transposon mutagenesis. …

Loosening of total joint replacements continues to be a major mode of failure in cemented total joint arthroplasty. Clinical investigations report failure of the stem-cement interface after total hip replacement may be an important factor in loosening of hip prostheses. There are basically two design philosophies to reduce the risk of cemented stem failure: promote bonding (i.e. pre-coat, rough surface) and promote debonding (i.e. polished surface). The latter approach is examined in this study. The influence of stem-cement debonding and the viscoelasticity of the bone cement was investigated using a smooth tapered collarless stem. Treatment of acid mine drainage (AMD) through the use of anoxic limestone drains (ALDs) is a technology that has shown mixed success. Precipitation of solid phase materials, primarily Fe{dollar}\\sp{lcub}3+{rcub}{dollar} and Al{dollar}\\sp{lcub}3+{rcub}{dollar} hydroxides, inhibits limestone dissolution by presumably creating a barrier between the limestone surface and the AMD, which impedes neutralization at an optimum rate. A laboratory project was initiated to: monitor geochemical transformations of reactive species ({dollar}\\rm Al\\sp{lcub}3+{rcub},\\ Fe\\sp{lcub}3+{rcub}{dollar}, and SO{dollar}\\sb4\\sp{lcub}2-{rcub}{dollar}), determine solid phases controlling precipitation reactions, and determine a kinetic model for limestone dissolution in ALDs. Water column samples from three vastly contrasting coastal regimes (Framvaren Fjord, Norway, the Amazon and Mississippi River outflow regions) were analyzed to study the estuarine transport and scavenging processes of $\sp{210}$Pb, $\sp{210}$Po, $\sp{234}$U and $\sp{238}$U. The Amazon and Mississippi River outflow regions are two of the world's largest river-ocean mixing systems that contrast each other sharply in many physiographical and geochemical features. The Amazon is a tropical, high energy, shelf environment where estuarine physico-chemical processes are strongly influenced by sediment-water interactions. A spreadsheet program was successfully developed which determines the engineering requirements for pipeline transport of sediments from dredge sites to wetland restorations sites. The program called SLURRY.XLS involves several new correlations. A new correlation for determining the critical velocity of particle transport is presented. New correlations were statistically developed for determining the drag coefficient of particles. The spreadsheet also successfully determines settling velocities of the individual particle sizes in any settling regime. A second spreadsheet program entitled "CONSOL.XLS" uses geotechnical principles of consolidation for determining marsh surface elevation with and without hydraulic fill versus time. Subjects who had sustained a mild head injury (MHI) and uninjured control subjects were examined before and after 36 hours of sleep deprivation. MHI subjects and uninjured controls were selected from among individuals who scored above the 50$\rm\sp{th}$ percentile on the Postconcussion Syndrome Checklist (PCSC), a measure designed to assess the frequency, intensity and duration of postconcussion symptoms. Sixty subjects were divided into four groups: Head injured/sleep deprived, head injured/non-sleep deprived, uninjured/sleep deprived, and uninjured/non-sleep deprived. The purpose of the present research was to examine the contribution of age and individual differences to everyday problem solving, focusing on goal preference. Three pilot studies were conducted to generate interpersonal problem solving goals that were high in emotional arousal and low in emotional arousal. In the Experiment Proper, 40 young and 40 older adults were given 6 vignettes depicting everyday problems. Subjects rated their preference of both high and low experimenter-provided goals. Subjects then completed 4 individual difference questionnaires that measured stimulation intensity, affect intensity, and emotional control. Thiarubrine A (TA), a plant secondary metabolite synthesized by members of the family Asteraceae possesses antifungal and antiviral properties and is a potential pharmaceutical. The objective of the work was the optimization and scale-up of a bioprocess for the production of TA using hairy root cultures of Ambrosia artemisiifolia. At the inception of the work an established hairy root culture of the model system was used as inoculum in reactor experiments for the development of a protocol for the quantification of TA and in subsequent shake flask experiments to determine optimum culture conditions. In 1982, Iwata, Dorsey, Slifer, Bauman, and Richman developed a systematic assessment method to identify the operant functions of self-injurious behavior. In this study, the Iwata et al., method was used to assess the operant functions of aggression displayed by children and adolescents with developmental disabilities. Although researchers have demonstrated that aggression serves operant functions, there has been no comprehensive analysis of aggression using the Iwata et al. method. Twelve children and adolescents participated. Mechanisms modulating avian seasonal reproduction involve a complex interaction between exogenous environmental information and internal physiologic systems, such as functional changes in the hypothalamic-gonadal axis and changes in the influence of monoaminergic neurotransmitters on hypothalamic neuroendocrine cells. This project was undertaken to describe seasonal changes of the testes and immunoreactive-(ir)-gonadotropin-releasing hormone (GnRH) neurons in House Sparrows in southeastern Louisiana, and to examine the role of monoaminergic neurotransmitters in modulating seasonal reproduction through the use of dietary monoamine precursors. The current study investigated possible etiological risk factors in the development of social anxiety. The risk factors examined in this study were family environment, negative peer interactions, neuroticism, self-perceived attractiveness, public self-consciousness, social behavior goal/ability discrepancy, and fear of negative evaluation. These variables were hypothesized to have both direct and indirect influences on the development of social anxiety. The research presented in this dissertation generally involves aspects of molecular spectroscopy and contains two areas of focus. In the first section, the influences of organized media on the excited-state phenomena of intramolecular proton transfer (ESIPT) and photoisomerization have been investigated. Specifically, the photochemical properties of the novel ESIPT molecule 10-hydroxybenzo(h) quinoline (HBQ), as well as 2-(2$\sp\prime$-hydroxyphenyl)benzazoles (HBAs), have been examined in the presence of cyclodextrins (CDs) and micelles using absorbance, steady-state, and time-resolved emission spectroscopies. Diethyltoluenediamine (CAS 68479-98-1) is a ring-ethylated analog of toluenediamine (TDA) and like TDA, consists of the 2,4- and 2,6-diamine isomers. DETDA was developed by Albermarle Corporation as a substitute for TDA in some applications. To determine the uptake and distribution of 2,4- and 2,6-DETDA, 5 groups of 4 adult male Sprague Dawley rats were gavaged at 179 $\mu$mol/kg body weight with ($\sp3$H) -2,4- or ($\sp3$H) -2,6-DETDA. Multiple tissues were collected at 1, 4, 8, 24 and 48 hours. Very high levels of label occurred in the gastrointestinal system and bladder during the first 8 hours while liver and kidney exhibited …

This article pits Ronald Dworkin against Fyodor Dostoyevsky. The article critiques Ronald Dworkin's answer to the question of fit: how judges reconcile general legal rules with particular situations. Dworkin's heavy focus on legal principles under-emphasizes the importance of facts in judicial decisionmaking. This work was conducted to determine the frequency and distribution of stable cytogenetic damage (translocations and insertions) in human lymphocytes using chromosome painting. Included were cells exposed to gamma radiation in vitro and selected individuals from a healthy unexposed California population (n = 27), and a population of Russians exposed during cleanup work the Chernobyl nuclear accident (n = 120) and Russian controls (n = 69). The in vitro results showed that the frequencies of radiation-induced stable aberrations in chromosomes 3, 5 and 6 were proportional to chromosome size. This study examines the interaction between two genera of bacteria and the biochemical reduction of uranium. The bacteria studied are the obligate anaerobe, Desulfovibrio desulfuricans, and the facultative aerobe, Escherichia coli. Preliminary experiments established the pH, cell concentration, substrate concentration, and type of substrate that favored reduction of hexavalent uranium in aqueous solutions. Uranium reduction is most complete at pH 7 and 9, with cell concentrations of at least 33.5 mg/ml (dry cell weight). Whole cells of D. desulfuricans were found to reduce uranium in the presence of lactate and hydrogen whereas in E. coli, reduction was achieved only in …

Flight muscle histolysis was investigated in the dorsal longitudinal muscles (DLMs) of the cricket. Histolysis in the DLMs was characterized by analysis of total protein content, tissue wet weight, and relative percent muscle shortening. The purpose of this study was to examine the provisional prevalence rates and characteristics of self-mutilative behavior in a community sample of adolescents. A total of 368 adolescents between the ages of 12 and 19 participated in this investigation. Thirty-nine percent of high school students sampled (n = 143) engaged in self-mutilation within the past year. Commonly endorsed behaviors were biting self, hitting self on purpose, and cutting/carving skin. Self-mutilators were likely to engage in these behaviors to reduce internal tension, as well as to gain attention.
